Most forensic laboratories routinely employ Gas Chromatography/ Mass Spectrometry (GC/MS) as the preferred method for this examination. The technique provides a rapid, semi-automated analysis of the sample and typically yields sufficient information to identify the compounds in question. However, the application of GC/MS for drug analysis does have its limitations. Certain drugs yield minimal mass spectral fragmentation patterns using electron impact MS, while other compounds, such as some diastereomers and positional isomers, are not readily differentiated technique to mass spectroscopy. Infrared spectroscopy (IR, meaning FTIR) provides an alternate technique to mass spectroscopy for the identification of organic compounds. Recent improvements in the hyphenated technique, Gas Chromatography/Infrared Spectroscopy (GC/IR) may provide a simple alternative or supplemental approach to GC/MS for identification of certain compounds. A newly introduced instrument collects GC effluent on a liquid nitrogen cooled, IR transparent window that allows the direct analysis of the deposited solid material. This technique is superior to the IR light pipe in sensitivity, IR spectral quality, and allows direct comparison of the collected spectra to existing IR databases. Our research developed procedures and protocols for the analysis of drugs and determined the benefits and limitations of this technology. The research focused on the routine identification of commonly encountered drugs, designer drugs, closely related drug isomers, as well as the fundamentals of the gas chromatography and infrared systems. Statement of purpose: The research was undertaken to develop this technology into a viable technique for the forensic community. The instrument was studied for repeatability, sensitivity, and selectivity while optimizing for analysis of a wide range of drug samples. Based upon this work the instrument proved to be a powerful forensic tool providing complimentary data to GC/MS. Acceptable levels of sensitivity, linearity, and reproducibility were achieved using the GC split-less injection mode. Concern about cross contamination of samples on the collection disk were dispelled as the deposited GC vapor produced solid "tracks" that were unique to each sample and appropriately documented by the instrument. Analytical methods were developed for the routine analyses of drugs and synthetic cannabinoids. Through these studies the instrument was verified for casework analysis and is presently in operational use in our laboratory. Software limitations hindered research progress, although software and hardware upgrades were made by the vendor (Spectra Analysis) some of which were driven by feedback provided by staff at the Vermont Forensic Laboratory (VFL).

Book excerpt: The metabolism of absorbed xenobiotic compounds in humans results in a mixture of target compounds applicable for analysis, trapped in complex biological matrices. Gas chromatography-mass spectrometry (GC-MS) is a powerful analytical technique that has been successfully applied in the analysis of volatile and semi-volatile compounds from complex biological samples. This is due to the ability of GC-MS to separate different sample constituents at trace levels while providing accurate molecular structural information for the resolved compounds. The complexity of biological specimens and their largely aqueous nature, combined with the physicochemical properties of target analytes resulting from metabolism, greatly precludes direct analysis of biosamples by GC-MS. Traditionally, highly laborious and time consuming sample preparation procedures are performed to isolate and chemically alter target analytes to attain suitable amenity for the detection system. Furthermore, routine analytical procedures in clinical toxicology laboratories are signified by short specimen turn-around times. The commonplace use of GC-MS in modern-day laboratories still suffer from prolonged turn-around times that result from both sample preparation steps and lengthy instrumental analysis. Simplified and cost-effective analytical procedures capable of extracting multiple analytes, with divergent functional groups, from biological matrices in a timely manner are therefore required. To address this issue, this work describes the development of validated extractive-derivatization methods combined with fast GC-MS analysis for expedient and accurate quantitation of different analytes in occupational monitoring and workplace drug testing. Extractive alkylation of acidic analytes phenol, o-cresol, mandelic acid, hippuric acid, and (o-, m-, p- ) methylhippuric acid for simultaneous urinary bio-monitoring of occupational exposure to benzene, toluene, ethylbenzene, and xylene, respectively, is performed. Extractive acylation for simultaneous urinary confirmation of basic analytes amphetamine, methamphetamine, norephedrine, methcathinone, ephedrine, methylenedioxyamphetamine (MDA), methylenedioxymethamphetamine (MDMA), methylenedioxyethylamphetamine (MDEA) and N-methyl-1-(3,4 methylenedioxyphenyl)-2-butanamine (MBDB) in workplace drug testing is performed. The successful combination of abovementioned techniques alongside fast GC-MS allows increased sample throughput and decreased turn-around time for routine analysis while maintaining bioanalytical quantitative criteria, as required in a clinical toxicology laboratory setting.

Retention times and UV-spectra were used for the identification of various adulterants and synthetic by-products in street samples. This method is compared with gas-chromatography/mass spectrometry (GC/MS) Forty street samples were analyzed by both methods. It is concluded that the combined use of HPLC/DAD and GC/NPD as a routine analytical procedure for the analysis of illicit samples could be a good alternative to GC/MS for forensic purposes. It is due to their high accessibility and the possibility to perform simultaneously qualitative and quantitative analysis.

In gas chromatography, the components of a sample are dissolved in a solvent and vaporized in order to separate the analyses by distributing the sample between two phases: a stationary phase and a mobile phase. The mobile phase is a chemically inert gas that serves to carry the molecules of the analyze through the heated column. Gas chromatography is one of the sole forms of chromatography that does not utilize the mobile phase for interacting with the analyze. The stationary phase is either a solid adsorbent, termed gas-solid chromatography (GSC), or a liquid on an inert support, termed gas-liquid chromatography (GLC). Helium remains the most commonly used carrier gas in about 90% of instruments although hydrogen is preferred for improved separations. This inert gas goes through a glass column packed with silica that is coated with a liquid. Materials that are less soluble in the liquid will increase the result faster than the material with greater solubility. The purpose of this book entitled Applications of Gas Chromatography is to provide a better understanding on its separation and measurement techniques and its application. Since chromatography techniques are separating and analyzing methods, this book will help other researchers and young scientists to choose a suitable chromatography technique.
