Model substrates were synthesized which allowed investigation into Diels-Alder, radical cascade, and palladium-catalyzed bond-forming reactions as possible routes to the morphine alkaloid skeleton. As a secondary objective, three separate series of aromatic substrates were subjected to whole-cell oxidation with Escherichia coli JM 109 (pDTG601), a recombinant organism over-expressing the enzyme toluene dioxygenase. Included in this study were bromothioanisoles, dibromobenzenes, and cyclopropylbenzene derivatives. The products of oxidation were characterized by chemical conversion to known intermediates. The synthetic utility of one of these bacterial metabolites, derived from oxidation of o-dibromobenezene, was demonstrated by chemical conversion to (-)conduritol E.

Book excerpt: This thesis describes the approach towards chemoenzymatic total synthesis of ent-dihydroisocodeine and chemoenzymatic total synthesis of ent-oxycodone as well as the development of a new method for the preparation of rearranged allylic isocyanates. The synthesis of ent-dihydroisocodeine started from phenethyl acetate and included a microbial oxidation of phenethyl acetate by E. coli JM109 (pDTG601A), a Mitsunobu reaction to the couple A- and C-rings, Heck cyclization to construct the E-ring and Henry reaction to introduce the nitrogen functionality as key steps. The construction of the B-ring proved to be challenging and neither radical cyclization nor attempts to perform photochemistry or nucleophilic opening of an epoxide gave any positive results. The chemoenzymatic total synthesis of ent-oxycodone was accomplished starting from phenethyl acetate in 23 steps. The tricyclic intermediate was furnished in the same manner as described above. The olefin to ketone conversion and a double Henry reaction allowed the construction of the B-ring. Unfortunately, it was established that the resulting hydroxyl group at C14 and amino group at C9 were of the undesired trans stereochemistry. To complete the morphine skeleton the transformation of the side chain at C13 to an N-methyl-p-toluenesufonamide via Mitsunobu reaction as well as the elimination of the amino group at C9 via formation of an N-oxide were performed. Subsequent radical cyclization of the side chain at the C9 position formed the last D-ring. The silyl ether deprotection followed by oxidation provided ent-oxycodone. The other approach to construct the D-ring was based on the formation of a lactone and the elimination of the amino group via an N-oxide. The nitrogen functionality was reinstalled using sodium azide and was accompanied by the introduction of the C10 hydroxyl group. Reduction of the azide and subsequent formation of the amide allowed access to the core skeleton of the target compound. The removal of the C10 hydroxyl group accomplished the synthesis. The last project involved the development of a new method for the preparation of rearranged allylic isocyanates from allylic alcohols using 1-cyano-4-dimethylaminopyridinium as the source of electrophilic cyanide. Experimental and spectral data are provided for all the compounds.

Book excerpt: This book provides an interdisciplinary, integrative overview of environmental problem-solving using mild reaction conditions, green reagents, waste free and energy efficient synthesis in both industry and academic world. Discussions include a broad, integrated perspective on sustainability, integrated risk, multi-scale changes and impacts taking place within ecosystems worldwide. Features: This book serves as a reference book for scientific investigators who need to do greener synthesis of organic compounds, drugs and natural products under mild reaction condition using green reagents, eco-friendly catalysts and benign reaction mediums over traditional synthetic processes which is a key driving force of scientists. Greener synthesis of multiple value-added heterocycles opens up a new horizon towards the organic catalysis and for this purpose, development of natural resources acts as an effective catalyst. Using environmentally friendly reaction medium e.g. ACC, WETSA, WEBSA have been used for the synthesis of some crucial heterocyclic scaffolds such as bisenols and 2-amino-4H-pyrans, tetraketones, pyrans, and biaryls. This book can also be used as a textbook for graduate and post graduate level courses for students. Furthermore, the problems with answers in book will add better understanding for students.

Unique in its approach, it deals with almost every aspect of modern synthesis. The first part of the book describes methods and reagents, with particular emphasis on rapidly developing organometallic and biooriented procedures. In the second part, these tools are applied to the syntheses of interesting target compounds and natural compounds with remarkable physiological properties. Mechanistic discussions and retrosynthetic analyses are included. More than 1000 up-to-date references help the reader to pursue the topics highlighted here. This book gives both the active researcher and the advanced student insight into the competitive atmosphere, creativity, and resourcefulness so characteristic of organic synthesis today.

Book excerpt: Isoquinolines form one of the largest groups of plant alkaloids and they in clude a number of valuable clinical agents such as codeine, morphine, eme tine and tubocurarine. Research into different aspects of isoquinolines con tinues in profusion, attracting the talents of botanists, chemists, bioche mists, analysts, pharmacists and pharmacologists. Many of these aspects are of an interdisciplinary nature, and in April 1984, The Phytochemical Society of Europe arranged a 3-day symposium on The Chemistry and Bi ology of Isoquinoline Alkaloids in order to provide a forum for scientists of differing disciplines who are united by a common interest in this one class of natural product. Each chapter in this volume is based on a lecture given at this symposium. Attempts have been made to make the aims and objectives, experimental findings and conclusions reached, intelligible to scientists of differing backgrounds. The introductory chapter, which is mainly based on a historical discus sion, stresses that plants containing isoquinolines have proved to be both a boon and a curse to mankind. The Opium Poppy, Papaver somniferum, produces the medicinally used alkaloids morphine, codeine, noscapine and papaverine whilst it also continues to provide drugs of abuse, particularly morphine and its readily prepared O,O-diacetyl derivative, heroin. Numer ous other alkaloids have been isolated from other members of the Papaver acea, and a knowledge of their presence and distribution within the various species has proved a useful adjunct to systematic botanical studies.

Available in PDF, EPUB and Kindle. Book excerpt: This third edition is a comprehensive and extended study about the best known approaches for preparing the main types of glycosides, covering the classic and more recent glycosylation reactions used for preparing simple and challenging glycosides currently used as potent antiviral and antineoplastic drugs, or fluorogenic substrates used for enzymatic detection in cell biology. Besides, this new edition provides more examples of the glycosidic methodologies followed for preparing complex glycoconjugates such as glycoproteins and glycosphingolipids and gangliosides used as adjuvants or as synthetic vaccines candidates. Also, additional mechanistic evidence is presented for better understanding of the glycosylation reaction, trying to identify the variables mainly depending on protecting and leaving groups, as well as catalyst and reaction condition which altogether directs the anomeric stereo control. A chapter on the glycoside hydrolysis is included in view of the increasing interest in the use of biomass as a natural and renewable source for obtaining important intermediates or products used in food or valuable materials. The author includes information in the characterization of glycosides section with the aim of giving additional tools for the structural assignment through NMR, X-Ray and mass spectra techniques.
