All samples continue to show a downward trend in pH. The Intralipid emulsions showed the most variation when measurements were taken for a year or more. The results are given of a 90 day pH experiment on I.V. fat emulsions that were purposely frozen. Ten emulsions were selected at random for the experiment. Three 20cc samples were removed from each bottle with vacutainers and stored in a freezer at -5C. The pH of the frozen and unfrozen samples are compared: Unfrozen emulsions showed a decrease of -0.539 in pH units; frozen samples showed an increase of 0.072. Results are also reported of the thin layer chromatography tests on intravenous fat emulsions done at monthly intervals since June 1965. (Author).

Book excerpt: A systematic procedure and some results are given for examination of Lipofundin and Intralipid samples by separation of the emulsions into aqueous and oil phases, followed by a combination of thin layer chromatography, gas liquid chromatography and other separation and test procedures to separate and partially identify the various components present. Te procedure also serves to quantitatively determine the water-soluble components and the triglyceride and lecithin (posphatidyl choline) components, and also the total of ''other components'' or impurities. the relative proportions of the latter are unexpectedly large. Small proportions of various sterols are present, these being identified and measured by gas chromatography. The possibility is discussed of a tendency toward inversion of an oilin-water emulsion to water-in-oil, as a consequence of the antagonism in lecithin-cholesterol systems and the possible inversion of the lecithin-cholesterol ratio upon infusion of an intravenous emulsion. Book excerpt: The report gives further data on chemical composition, particle size and pH variation in intravenous fat emulsions in current and recent use. Some retests were made on old emulsions of the previous reports. All of the new emulsions that have been received were tested. New colorimetric procedures with ferrous thiocyanate reagent show the presence of peroxy and hydroperoxy derivatives in varying amounts in most of the emulsions, apparently associated with triglycerides and phospholipids. Other colorimetric tests have been introduced, to detect carbohydrates and amine-type compounds. Improved thin layer chromatography development procedures indicate the presence of bound lipids in the water phase. A twodimensional TLC technique is illustrated. Practically all emulsions are continuing to show a decrease in pH as time passes. Some of the most recently submitted emulsions are showing a more rapid pH drop than has been previously noted. It can thus serve as a guide in animal and clinical testing of emulsions. The pH of stored I.V. emulsions continues to show a downward trend. Thin Layer Chromatography (TLC) analysis done after each pH analysis shows no drastic changes in lipid composition. Chemical studies with chromatography will continue to follow pH changes until emulsions break. The introduction of a densitometric scanning device for standard TLC plates will further the development of methods combining well-established chromatographic, chemical and spectrophotometric procedures for the characterization and quantitation of closely related chemical classes of compounds. The main arguments in favor of scanning procedures combined with TLC are productivity and economy of time and effort. With the exception of GLC, the scanning method appears to be the only procedure in which the speed of quantitative determination will match the speed of the chromatographic method of separation. The method furthermore, has the advantage that trace components of samples can be measured directly without using isolation procedures. This new approach may well prove to be important in biochemical research, particularly with I.V. fat emulsions where a large number of samples require characterization and estimations. (Author).
