Book excerpt: To improve current chemotherapy treatments, there is a need for non-harmful imaging modalities that can serve to treat and longitudinally monitor patient tumors. Fluorine MRI provides a safe tracer technology that allows recognition of the size and anatomical location of the tumor. Polymeric nanoparticles were designed with an azide termini, high fluorine content for MRI, self-assembly, and minimal toxicity. Copolymers were polymerized using fluorinated and PEG monomers and shown to self-assemble to colloidal nanoparticles for passive tumor targeting. To improve the imaging properties of the nanoparticle, a library of copolymers, with various molecular weights and compositions, were synthesized to compare imaging efficiency. The signal properties remained consistent between the different polymer compositions indicating a need for external modalities that can be conjugated to the azide handle. These studies indicate that development of the polymeric system can be used to improve the theranostic properties for dual 19F MRI tracer and treatment.

That is the reason why, although mainly dealing with synthetic drug carriers, the first chapter is related to preparation and physicochemical properties of nanoparticles constructed from natural polymers. This is followed by two chapters involving the application of polymeric drug carriers in cancer therapy. The second chapter extensively describes in vitro and in vivo behaviour of purely synthetic nanoparticles. As ub the case of liposomes, a problem can be that some particles are preferentially taken up by the reticuloendothelial system. Magnetic drug carriers provide a partial answer to this problem since their tropism can be managed by an external magnet. The third chapter is entirely devoted to this aspect. A brief but original application of nanoparticles is given in the fourth chapter by reviewing their use in ocular therapy. Finally, two general applications of polymeric microspheres are discussed in chapters 5 and 6.
