Book excerpt: The work embodies development and systematic optimization of oral controlled release (CR) mucoadhesive hydrophilic matrices of atenolol using "Design of Experiments". A central composite design was used to optimize drug release profile and bioadhesive strength. Two polymers, namely, Carbopol 934P and sodium CMC, were taken as synergistic factors. The 3-D response surfaces and 2-D contour plots indicated significant effect of both the polymers on ex vivo bioadhesive strength of tablet against porcine gastric mucosa, diffusional release exponent and drug release extension. The optimized drug delivery formulation was selected by feasibility and grid searches based on maximization of the response variables. Validation of drug delivery optimization study using experimental runs indicated high prognostic ability of the chosen experimental design. Besides unraveling the effect of the two factors on various response variables, the study helped in finding the "optimized" formulation with excellent bioadhesive strength and extended release for 24 h, holding immense promise for once-a-day gastroretentive CR formulation and minimizing fluctuations in plasma drug levels.

Available in PDF, EPUB and Kindle. Book excerpt: Polymer systems can be developed into a variety of functional forms to meet industrial and scientific applications. In general, they are presented in four common physical forms: (1) linear free chains in solution, (2) covalently or physically cross-linked reversible gels, (3) micro and nanoparticles, and (4) chains adsorbed or in surface-grafted form. Hydrogels are polymeric particles consisting of water-soluble polymer chains, chemically or physically connected using, in general, a cross-linking agent. These materials do not dissolve in water but may swell considerably in aqueous medium, demonstrating an extraordinary ability (>20%) to absorb water into the reticulated structure. Such features make these materials promising tools in the biomedical field, especially as controlled drug release systems. This chapter describes recent progress in the development and applications of polymeric nanostructured hydrogels, mainly in the context of biomedical devices. Additionally, it reports the significant advances in synthesis and characterization strategies of these materials. Special attention is devoted to smart or stimuli-responsive bionanogels, which mimic the property of living systems responding to environmental changes such as pH, temperature, light, pressure, electric field, chemicals, or ionic strength, or a combination of different stimuli. Consequently, these bionanogels offer an efficient solution to various biomedical limitations in the field of drug administration.

Available in PDF, EPUB and Kindle. Book excerpt: Physical hydrogels present direct interactions between polymeric chains and forms non-permanent networks, providing an attractive alternative to chemically cross-linked systems. These hydrogels allow incorporation of macromolecular drug substances under mild process conditions, comprising properties such as water permeability, biocompatibility and tensile strength. The aim of the present work was to provide an overview of physical hydrogels for drug delivery applications. The main focus of this review was based on chemical and structural composition, and on the physicochemical properties necessary for drug release. The structural basis of these drug delivery systems was discussed on the many different macromolecular structures presented: physical entangled networks of linear polymers, polymer blends and composites block or graft copolymers and hydrogels formed by aggregation or complexation of polymers, intended for drug, peptide and protein delivery applications.
