Available in PDF, EPUB and Kindle. Book excerpt: Plasma Protein Metabolism: Regulation of Synthesis, Distribution, and Degradation covers the concepts concerning the physiological and pathophysiological factors regulating the distribution, degradation, and synthesis of plasma proteins. This book is organized into nine parts encompassing 32 chapters. The first parts present the assumptions and methodology involved in the various in vivo and in vitro techniques that provide insights to protein metabolism. The next parts describe the techniques of protein isolation, characterization, labeling, and mathematical analysis of the data, as well as the methods for directly quantitating protein synthetic rates in nonsteady state conditions. Other parts are devoted to the factors involved in regulating the serum levels of albumin, acute phase reactants, immunoglobulins, clotting factors, complement and hormone-binding proteins. The controlling factors include such general and specific physiological regulators of protein synthesis and catabolism as levels of specific serum proteins, hormonal regulators, variations of temperature and oncotic pressure, antigenic stimulation, and nutritional factors. The remaining parts analyze the pathophysiological factors including disorders of protein synthesis, distribution, exogenous catabolism, and external loss and stress that produce abnormal levels of serum proteins. This book is intended primarily intended to protein chemists and researchers.

Book excerpt: This book represents a factual account of the proceedings of an international symposium on the pathophysiology of plasma protein metabolism, which was organised in October 1982 by the Plasmaprotein and Immunology Division of the C.N.R. Institute of Clinical Physiology at the University of Pisa (Italy). Several of the contributors are former members of the International Study Group on Plasma Protein Metabolism, the last meeting of which was held in Turin (Italy) in 1974, under the auspices of the scientific organisation of the same institute. The symposium took the form of a series of lectures, with the main objective of providing a positive contribution to the state of the art of several topics related to the kinetic and pathophysiological factors regulating the synthesis, distribution and degradation of plasma proteins. The first four chapters form a group, each one considering a special aspect of the kinetics of turnover and distribution of plasma proteins in general; particular attention is paid to the recent advances in the field of kinetic modelling, the choice of the best models and the optimisation of the experimental designs. The next seven chapters consider the regulation of synthesis, distribution and catab olism of various classes of plasma proteins including albumin, immunoglobulins, complement fractions and acute-phase proteins. The remaining chapters deal with metabolic studies of various plasma proteins (including tumour markers, coagUlation proteins and lipoproteins) in different disease states, such as malig nancies, coagulative disorders, malnutrition and the extensive group of athero sclerotic cardiovascular diseases.

Book excerpt: It was the year of 1969 when this monograph was originally published in Japanese by Professor TADASHI KAWAI, titled as "The Plasma Proteins, Their Fundamental and Clinical Aspects." After I read through the Japanese edition, I was impressed by its rather complete coverage of the subjects and their detailed descriptions. I have felt that this excellent monograph should be distributed not only among our Japanese scien tists but also among many other colleagues throughout the world. I am happy, the refore, to know that the English edition of his monograph, partly revised, is ready to be published at this time. Professor KAWAI received his postgraduate medical training in U.S.A. for seven years, and was certified by the American Board of Pathology in both Anatomical and Clinical Pathology in Fall, 1962. Thus, I believe, he is the most suitable fellow for publishing the English edition of this kind.

Book excerpt: Less than 50 years ago it was discovered that steady-state protein concentrations in plasma are the net result of continuous elimination and synthesis of protein molecules. The first quanti tative studies on the turnover and distribution of plasma pro teins were made around 1950, after the introduction of radio labeled protein preparations. Around 1970, another development in quantitative interpre tation of circulating proteins was initiated in clinical enzy mology. Estimation of cumulative release into plasma of cellular enzymes can be helpful in a variety of diseases to assess the extent of tissue damage and to evaluate therapy. Enzymes can be considered as biological tracers, i.e. minute quantities of protein can be accurately determined by their spe cific catalytic activities. However, radioactive tracers permit direct estimates of turnover and distr ibution by measurement of excreted radioactivity, possibilities that are not available for enzymes. Consequently, only a few techniques used in tracer studies with radiolabeled proteins can be applied to circulating tissue enzymes and this may explain the lack of communication between the fields of plasma protein metabolism and quantitative clinical enzymology. In the present study a summary is given of the basic methods used in both fields, with emphasis on the equivalence of various models and formalisms used by different authors. It is shown that major limitations in the study of circulating tissue enzymes can be overcome if two different, but simultaneously released, en zymes can be measured. The resulting method will also be applied to plasma protein metabolism.

Available in PDF, EPUB and Kindle. Book excerpt: Mammalian Protein Metabolism, Volume I focuses on the processes, methodologies, biosynthesis, protein formation, and reactions involved in mammalian protein metabolism. The selection first elaborates on the origin and growth of concepts of protein metabolism, including the development of nitrogen balance as a technique for the study of protein metabolism; identification of proteins as a chemical class; discovery of nitrogen and its biological consequences; and recognition of the importance of nitrogenous compounds. The text then examines protein digestion and absorption in nonruminants and digestion and absorption of nitrogenous compounds in ruminants. Topics include passage of nitrogenous compounds from the rumen; utilization of nitrogenous compounds in the rumen; and endogenous nitrogen entering the reticulum and rumen. The book examines free amino acids and peptides in tissues, metabolic fate of amino acids, protein biosynthesis in mammalian tissues, and metabolism of plasma proteins. Discussions focus on the distribution of proteins between plasma and lymph, interpretation of plasma radioactivities, amino acid activating enzymes, ribosomes and protein synthesis, pathways of amino acid degradation, and synthesis of urea. The selection is a dependable source of data for researchers interested in mammalian protein metabolism.

The book summarizes the biochemistry of protein synthesis and breakdown, and explains the methods that are used to examine protein metabolism in humans, together with their limitations. Chapters review the effects of nutrition, hormones, metabolic substrates, and physical activity, while various topics of clinical interest include cancer, diabetes, tissue injury, pregnancy, renal disease, muscular dystrophies, and other conditions. Normal values are presented for turnover of proteins in the whole body and individual organs, and for turnover of many individual proteins. This is thus a valuable resource for physiologists, nutritionists, and clinicians interested in the regulation of body protein stores in health and disease. For scientists primarily interested in the basic aspects of protein metabolism, it shows how the basic knowledge is being applied to the study of humans.
