Available in PDF, EPUB and Kindle. Book excerpt: Phospholipases generate lipid signaling molecules through their hydrolytic action on phospholipids and are known to regulate function of a variety of cells under normal and diseased conditions. While several physiological, biochemical and molecular techniques have identified key players involved in different disease processes, phospholipases have also emerged as critical players in the pathogenesis of a number of different diseases including cancer and heart disease. In addition, phospholipases are also implicated in such conditions as brain disorder/injury, kidney and immune cell dysfunction. Phospholipases in Health and Disease is a compilation of review articles dedicated to the study of the field with respect to biochemical and molecular mechanisms of normal and abnormal cell function. The wide range of area covered here is of interest to basic research scientists, clinicians and graduate students, who are engaged in studying pathophysiological basis of a variety of diseases. Furthermore, this book highlights the potential of the different phospholipases as therapeutic targets as well as part of prevention strategies. Twenty three articles in this book are organized in four sections that are designed to emphasize the most characterized forms of the phospholipases in mammalian cells. The first section discusses general aspect of phospholipases. Section two covers the role and function of phospholipase A in different pathophysiological conditions. The third section is focussed on phospholipase C which is believed to play a central role in transmembrane signaling. The final section covers phospholipase D which is present in a variety of different cells. The book illustrates that the activation of phospholipases is of fundamental importance in signal transduction affecting cell function. Overall, this book discusses the diverse mechanisms of phospholipase mediated signal transduction in different pathophysiological conditions and raises the possibility of specific forms of phospholipases serving as novel targets for drug development.

Available in PDF, EPUB and Kindle. Book excerpt: The NATO-Advanced Research Workshop "Esterases, Lipases and Phospholipases: From Structure to Clinical Significance" was held at the University of Bordeaux II, France from 22nd- 24th September 1993 under the Directorship of Professor Michel Clerc of the University of Bordeaux II. The meeting was organised by Hugues Chap (INSERM U 326, Toulouse, France), Georges Ferard (University of Strasbourg, France), Wolfgang Junge (University of Kiel, Germany) and Michael Mackness (University of Manchester, UK). In recent years it has become increasingly apparent that hydrolytic enzymes of the esterase, lipase and phospholipase type play central roles in the pathophysiology of many human diseases. The purpose of this NATO-ARW was to bring together experts (both clinical and scientific) in all three interrelated fields to review the current basic and clinical position and discuss future developments particularly with respect to future research aimed at determining the basic biochemical lesion involving hydrolytic enzymes involved in human disease and the use of these enzymes in diagnosis. As well as formal lectures from established researchers, the meeting also involved a number of lively round-table discussions on future developments and presentations from younger research workers, all of which are recorded in this Proceedings and which contribute to the success of the meeting.

Book excerpt: This volume provides a state-of-the-art overview of the role of phospholipase A2 (PLA2) in the pathophysiology of inflammatory diseases. Our knowledge about PLA2 has increased tremendously in the last two decades and investigations of the enzyme have revealed a whole family of different gene products. A rough distinction can be made between intracellular and secretory PLA2. The involvement of PLA2 in various life-threatening inflammatory diseases as well as in carcinomas is documented by many contributions to this volume. Recently developed transgenic and knockout mouse models are a powerful new tool for PLA2 research. A number of specific inhibitors of the various PLA2s offer new therapeutic approaches to control unwanted acute and chronic inflammatory responses and to intervene in malignant diseases. Due to the enzyme's important role in these pathophysiological processes, this update of PLA2 research will interest basic scientists, physicians, oncologists, ICU specialists and surgeons.

Available in PDF, EPUB and Kindle. Book excerpt: This volume in the Advances in Experimental Medicine and Biology Series is dedicated to developing an overall view of the "state-of the-art" of knowledge in the field of phospholipase A2 and to exploring the role and function of this enzyme in various inflammatory diseases. This book grew out of two major symposia on phospholipase A2 held in 1989: I). "Phospholipase A2: Pathophysiological Role of Soluble and Membrane-Bound Enzymes" organized by Dr. Doug Morgan and Dr. Ann Welton of Hoffmann-La Roche and sponsored by the New York Academy of Sciences (January 24, 1989). II). "Release and Function of Phospholipase A2 from Inflammatory Cells" organized by Dr. Patrick Wong and Dr. Edward Dennis at the FASEB meeting in New Orleans (March 22, 1989). Readers will find exciting advances in our understanding of the structure, function and molecular biology of phospholipase A2 research which is presented in this volume. The elucidation of gene structures of phospholipase A2 should lead to new insights and new approches in the control and regulation of the enzyme activity. This is primary to better understanding the function of phospholipase A2 and the determination of its activity in cellular and animal systems. Of great importance is the development of animal models for inflammatory disease in order to evaluate the role of phospholipase A2 and its inhibitors. We are convinced that this volume will be an excellent reference that surveys this field in which numerous investigators are currently actively at work.

This book was released on 2013-11-11 with total page 268 pages. Available in PDF, EPUB and Kindle. Book excerpt: During the past decade there has been a dramatic expansion of our knowledge on phospholipases in general, and phospholipase A2 (PLA2) in particular. Progress in this field has been evident on many fronts, with novel information rapidly accumulating in the literature regarding the chemistry and molecular biology of this enzyme and its role in many important physiological processes. These include cellular signal transduction via the G-protein cycle, and in the generation of many cellular mediators, such as the platelet activating factor (PAF) and the eicosanoids that participate in the initiation and propagation of inflammation, to mention a few. This symposium was organized to obtain an overview of current investigations on this enzyme from the standpoint of its chemistry, molecular biology and physiology. Another important focus of this symposium concerns the regulation of PLA2, including endogenous and synthetic inhibitors and activators of this enzyme. To review these important areas in PLA2 research we invited scientists who made significant contributions in this field. The papers in this volume are organized to emphasize the recent advances in several areas of investigation, including: (I) the structure and mechanism of action of PLA2, (2) mechanism of activation of PLA2, (3) molecular biology, physiology and endogenous inhibitors of this enzyme and finally, (4) clinical investigations emphasizing the pathophysiological role of this enzyme in human diseases. The first article in this volume is by Dr.
