The optimal dosing regimen of ibuprofen remains debatable since information on target exposure and pharmacology in this population is limited. This study aims to develop a population pharmacokinetic model for ibuprofen to improve the treatment of PDA in preterm infants. MethodSeventy-three neonates born after 24 to 32 weeks gestation that received ibuprofen treatment for PDA closure were included in a prospective study. Median gestational age (GA) was 26.1 weeks (24.0-30.1) and median postnatal age (PNA) at treatment initiation was 3 days (1-12). 218 plasma samples were collected in which either R-ibuprofen, S-ibuprofen or both could be quantified. Using NONMEM software, a model was developed to describe the pharmacokinetics of ibuprofen.
