Book excerpt: A veritable mountain of literature has been published showing the causal relationship of reactive oxygen/nitrogen species in human disease conditions, and there has been an explosion in the understanding of oxidative stress, the protective role of antioxidants and molecular events involved in the regulation of transcription, editing, and translation of key events leading to disease processes. Strategies need to be developed for prevention of diseases by allowing scientists and clinicians to obtain information on new and emerging advances. The molecular mechanisms involved in several diseases including Alzheimer's disease, atherosclerosis, diabetes, arthritis, and Parkinson's disease, as well as disorders of the eye, skin, cardiac, and pulmonary systems are discussed in this volume, along with scientific evidence supporting the value of dietary supplementation with antioxidants in the prevention of cellular damage leading to chronic disease. Special in vivo techniques are also discussed at length, along with the role of molecular studies in human risk assessment.

Available in PDF, EPUB and Kindle. Book excerpt: This first volume of the comprehensive, two-volume work on oxidative stress in lung disease introduces the molecular mechanisms, and the role of oxidants in the progression of different lung diseases. The lungs of humans and animals are under constant threat from oxidants from either endogenous (e.g. in situ metabolic reactions) or exogenous sources (e.g. air pollutants). Further, oxidative stress causes the oxidation of proteins, DNA and lipids, which in turn generates secondary metabolic products. The book consists of sections, each focusing on different aspects of oxidant-mediated lung diseases. As such it is a unique reference resource for postgraduate students, biomedical researchers and also for the clinicians who are interested in studying and understanding oxidant-mediated lung diseases. The second volume will incorporate other aspects of oxidant-mediated lung diseases, including prevention and therapeutics.

Available in PDF, EPUB and Kindle. Book excerpt: Oxygen free radicals and other reactive oxygen species are being postulated as causal agents in an increasing number of pathological conditions. Indeed, some investigators are suggesting that highly destructive reactive oxygen species are the final common path lead ing to tissue damage following a wide variety of insults including trauma, hypoxia, ischemia, hyperoxia, radiation, some toxins, and even strenuous athletic pursuits. But, as Robert Floyd points out, "Proof of the importance of oxygen free radicals and the oxidative damage they initiate depend on unequivocal evidence for the pres ence of free radicals and a clear association of their formation with the induction of the dysfunction of pathological conditions. " Since such proof does not come easily, there have been and will continue to be many controversies regarding the role played by reactive oxygen species in tissue damage. There have been many recent reviews of the chemistry and pos sible role of reactive oxygen species in many types of organ dys functions, tissue damage, degenerative diseases, and aging. This book is not such a review. Rather it presents for a diverse audience of physical-organic chemists, biochemists, medical researchers, and other investigators of pathophysiology, discussions of a variety of is sues important for understanding reactive oxygen species and their role in tissue damage.

Available in PDF, EPUB and Kindle. Book excerpt: Many physiological conditions such as host defense or aging and pathological conditions such as neurodegenerative diseases, and diabetes are associated with the accumulation of high levels of reactive oxygen species and reactive nitrogen species. This generates a condition called oxidative stress. Low levels of reactive oxygen species, however, which are continuously produced during aerobic metabolism, function as important signaling molecules, setting the metabolic pace of cells and regulating processes ranging from gene expression to apoptosis. For this book we would like to recruit the experts in the field of redox chemistry, bioinformatics and proteomics, redox signaling and oxidative stress biology to discuss how organisms achieve the appropriate redox balance, the mechanisms that lead to oxidative stress conditions and the physiological consequences that contribute to aging and disease.

Available in PDF, EPUB and Kindle. Book excerpt: The rapid expansion of the area of free radical biology in the last 25 years has occurred within a framework of assumptions and preconceived notions that has at times directed the course of this movement. The most dominant of these notions has been the view that free radical production is without exception a bad thing, and that the more efficient our elimination of these toxic substances, the better off we will be. The very important observation by Bernard Babior and colleagues in 1973 that activated phagocytes produce superoxide in order to kill micro organisms, served to illustrate that constructive roles are possible for free radicals. For many in the field, however, this merely underscored the deadly nature of oxygen-derived radicals, both from the microbe's point of view and from the host's as well. (Phagocyte-produced superoxide is responsible in part for the tissue injury manifested as inflammation. See Harris and Granger, Chapter 5, and Leff, Hybertson and Repine, Chapter 6.) Mother Nature, however, has a penchant for being able to make a silk purse from a sow's ear. If one is dealt a bad hand, one must simply make the best of it. After two decades of focusing on the destructive side of free radicals, the last few years have begun to reveal a new and finer perspective on free radical metabolism - a role in regulation of cellular function (see Schulze-Osthoff and Baeuerle, Chapter 2). Evidence from a number of sources suggests that an increase in the oxidative status of cell encourages that cell to grow and divide. Increasing the expression of mangnese superoxide dismutase can suppress the malignant phenotype of melanon cells (see Oberley and Oberley, Chapter 3). Oxidative stress beyond a certain poitosis (from the Greek, literally "to fall apart"). Is this suicide response an evolutionary fail-safe device to curtail tumorogenesis? Does oxidative stress-induced apoptosis account for the loss of immune cells in AIDS (see Flores and McCor Chapter 4)? This volume attempts to present the spectrum of roles, both good and bad played by active oxygen species as understood at this point in the evolution of this field of free radical biology.

Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.
