Book excerpt: The aim of this book is to provide methods and algorithms for the optimization of input signals so as to estimate parameters in systems described by PDE’s as accurate as possible under given constraints. The optimality conditions have their background in the optimal experiment design theory for regression functions and in simple but useful results on the dependence of eigenvalues of partial differential operators on their parameters. Examples are provided that reveal sometimes intriguing geometry of spatiotemporal input signals and responses to them. An introduction to optimal experimental design for parameter estimation of regression functions is provided. The emphasis is on functions having a tensor product (Kronecker) structure that is compatible with eigenfunctions of many partial differential operators. New optimality conditions in the time domain and computational algorithms are derived for D-optimal input signals when parameters of ordinary differential equations are estimated. They are used as building blocks for constructing D-optimal spatio-temporal inputs for systems described by linear partial differential equations of the parabolic and hyperbolic types with constant parameters. Optimality conditions for spatially distributed signals are also obtained for equations of elliptic type in those cases where their eigenfunctions do not depend on unknown constant parameters. These conditions and the resulting algorithms are interesting in their own right and, moreover, they are second building blocks for optimality of spatio-temporal signals. A discussion of the generalizability and possible applications of the results obtained is presented.

Book excerpt: Reaction networks are systems consisting of several species interacting with each other through a set of predefined reaction channels.Models of real world reaction systems often contain several parameters which play a significant role in determining the system's dynamics. Therefore, parametric sensitivity analysis is an essential tool for the modeling and parameter estimation process. Due to the complex and random nature of the reaction systems, among all approaches for sensitivity analysis, Monte Carlo simulation is the most suitable for the parametric sensitivity analysis because its complexity does not grow dramatically as the problem dimension grows. Most Monte Carlo methods for sensitivity analysis can be classified into three categories, the pathwise derivative (PD), the finite difference (FD) and the Girsanov transformation (GT). Comparisons of these methods for specific examples have been done by many researchers, which showed that when applicable, the PD method and FD method tend to outperform the GT method. However, to the best of our knowledge, no existing literature studies these observations from a theoretical point of view. In this thesis, we provide a theoretical justification for these observations in terms of system size asymptotic analysis. We also examine our result by testing several numerical examples. Other than the analysis for the efficiency of these Monte Carlo estimators, we also provide some sufficient conditions which guarantee the validity of the GT method. Finally, for an ergodic system, there exists a steady state distribution and hence it is reasonable for us to consider the steady state sensitivity estimation problem. We establish an asymptotic correlation result and use this result to justify the ensemble-averaged correlation function method introduced in the literature.

To a certain extent the study of the subject had been piecemeal, different criteria of optimality having been studied separately. Also to a certain extent the topic was regarded as being largely of theoretical interest and as having little value for the practising statistician. However during this decade two significant developments occurred. It was observed that the various different optimality criteria had several mathematical properties in common; and general algorithms for constructing optimal design measures were developed. From the first of these there emerged a general theory of remarkable simplicity and the second at least raised the possibility that the theory would have more practical value. With respect to the second point there does remain a limiting factor as far as designs that are optimal for parameter estimation are concerned, and this is that the theory assumes that the model be collected is known a priori. This of course underlying data to is seldom the case in practice and it often happens that designs which are optimal for parameter estimation allow no possibility of model validation. For this reason the theory of design for parameter estimation may well have to be combined with a theory of model validation before its practical potential is fully realized. Nevertheless discussion in this monograph is limited to the theory of design optimal for parameter estimation.
