Book excerpt: The NATO Advanced Study Institute held in the Hotel Metropole Llandrindod Wells, Powys, Wales from July 26th to August 4th 1981 provided an excellent platform for presentation and appraisal of our current knowledge of neurotransmitter systems. The proceed ings comprise the present volume and these were arranged to allow study of the pathways and the interactions of specific transmitter systems, one with another, in the various sub-regions of the central nervous system. In addition several of the key factors involved in neurotransmitter release were highlighted. These include the structural and molecular organisation of the nerve terminal, its organelles (such as vesicles, neurotubules and synaptic densities) and the mechanisms involved in neurotransmitter release including the participation of calcium. The organisation of transmitter synthesis, storage and transport were discussed together with their linkage to other streams of metabolism, and the cellular compartmentation of these processes. In addition to the principal lectures given at the Institute, many part1c1pants presented short reports for discussion and these are all included in this volume. The success of the Study Institute was due to the careful planning of the scientific programme to allow a smooth flow and integration of the topics included and this is reflected in the organisation of this book. Schneider xi Introduction The term metabolic compartmentation, as related to the chemistry of the brain, appeared in print for the first time about 15 years ago. The concept was developed in the laboratory of Dr. Heinrich Waelsch as a result of studies related to the metabolism of glutamic acid and glutamine. It was welcomed by a number of neurochemists who felt that the concept had validity and would help explain metabolic phenomena that were otherwise quite puzzling. The concept gradually achieved general acceptance, and by 1971 the amount of information that could profit by being examined from the point of view of metabolic compartmentation had increased sufficiently to warrant a symposium on the subject ("Metabolic Compartmentation in the Brain," Balazs and Cremer, eds. , MacMillan, 1973). Almost all the participants at that initial symposium were present at the second, on which this volume is based.

MCGAUGH Understanding of the nature and functions of neurotransmitter systems in the brain has increased enormously in recent decades. Lack of knowledge required us, not too long ago, to use the adjective "putative" when discussing transmitters. Such caution is no longer essential (at least for a number of transmitters). Impressive progress has been achieved in understanding the pharmacology, biochemistry and anatomy of transmitter systems. There has, however, been relatively less progress in understanding the functioning of brain transmitters in regulating and mediating behavior. A simple and certainly correct explanation for this is, of course, that understanding of neurotransmitter functions requires prior detailed knowledge of basic pharmacology, biochemistry and anatomy. Beyond that, it now seems likely that progress in understanding the functions of brain neurotransmitters will proceed only as we examine the interactions of neurotransmitter systems in regulating behavioral functions. This premise is, of course, suggested by the findings of studies of the chemical neuroanatomy of the brain: Neurotransmitter systems are influenced by other neurotransmitter systems and, in tum, influence the same as well as other systems. No system works alone. The chapters in this book explicitly examine the interactions of neurotransmitter systems involved in the regulation of cognitive processes. The facts and interpretations offered provide compelling support for the premise that cognitive processes are orchestrated by interactions among neurotransmitter systems. And, they offer promise that understanding of such interactions will be of critical importance in the develop ment of treatments for brain diseases affecting cognitive functioning.

Nowhere have the interactions of these levels of analysis been more apparent and borne more fruit than in the last several years of study of the develop ment of the vertebrate retina. Fields of investigation have a natural evolution, rdoving through periods of initial excitement, of framing of questions and controversy, to periods of synthesis and restatement of questions. The study of the development of the vertebrate retina appeared to us to have reached such a point of synthesis. Descriptive questions of how neurons are generated and deployed, and ques tions of mechanism about the factors that control the retinal neuron's type and distribution and the conformation of its processes have been posed, and in good part answered. Moreover, the integration of cellular accounts of development with genetic, molecular, and whole-eye and behavioral accounts has begun.

The existence of re ceptor substances on the surface of cells that recognize and bind to extracellular molecules was proposed at the beginning of the century by the pharmacologist and immunologist Paul Ehrlich and the physiologist J. N. Langley. Since then, receptors have been found to play an important role in numerous physiological and pathological processes. Over the years many attempts have been made to physically isolate and chemically characterize receptors, but because of the receptors' extremely low concentration and membrane localization, these ef forts have met with limited success. Yet, despite the failure to characterize receptor substances, the concept of the presence of such molecules has had considerable heuristic value. Using pharmacological, physiological, and im munologic approaches, researchers have identified several specific receptors, e. g. , a-and ~-adrenergic, nicotinic and muscarinic cholinergic, and histami nergic. With the characterization of various types of receptors on cell mem branes, many drugs were developed that proved to be experimentally and ther apeutically useful. It was only in the early 1970s that methods for the specific measurement, chemical characterization, and physical isolation of cell membrane receptors were developed. These advances were made possible by the availability of ligands with high specific radioactivity that retained their biological activity and of experimental procedures that differentiated between specific and non specific binding of ligands.

Actually, this recently formed hybrid, neuroscience, is in the process of merging with another well-established discipline, molecular genetics (frequently called molecular biology, and itself a hybrid), which appears to have sufficient hybrid vigor to form yet a new community of scientists, which, for want of a more imaginative term, has been called molecular genetic neuroscience. Clearly, advantages resulting from such mergers or hybridizations accrue not only from the merging discipline (neurochemistry in this case) to the new community (molecular genetic neuroscience), but also in the reverse direction. Clearly an exhaustive analysis of all aspects of the cerebellar system, as they relate to the legacy of Ramon y Cajal, would be impossible to contain in just one volume, given its far-reaching impact. Instead, we deliberately steered away from the traditional handbook approach that some of us have taken in the past and selected those aspects of cerebellar research currently under vigorous study that would also represent the widest scope of interest for neuroscientists in general and for cerebellar specialists in particular. In particular, we felt that as the discrete anatomy of the cerebellum is quite well known, only certain aspects of the structure should be discussed here.
