The 29-amino acid peptide (named after its N-terminal glycine and C-terminal alanine) was identified in 1983 by its C-terminal amidation. This 'reverse' approach, that is to discover a substance through a distinct chemical feature, and only subsequently to characterize its biological activity, was novel and has been successful in the identification of several other peptides. After the structure of galanin was determined in 1983, functional studies were performed with material purified from natural sources until the synthetic form of the peptide became available. Galanin can act as transmitter, modulator and trophic factor, and is involved in a number of physiological processes such as hormone secretion, cardiovascular mechanisms, feeding and cognition. This peptide may also be of significance for a number of pathological processes/disorders including pain, depression, Alzheimer's disease, epilepsy, addiction and cancer. This wide diversity of actions is mediated by three galanin receptor subtypes. The studies reviewed in this volume give a fairly complete overview of the spectrum of the biological actions and functions of galanin and its receptors and on possible therapeutic applications in a number of pathological conditions.

Since anxiety disorder are highly comorbid with mood disorders, understanding the common symptoms of these illnesses could indicate functional changes that elicit these symptoms. Also, alterations in the hypothalamo-pituitary-adrenal (HPA) axis, the system involved in maintaining homeostasis, are observed in many mood and anxiety disorders. The amygdala has been shown to modulate anxiety behavior and HPA axis function. The amygdala receives inputs from many sensory cortices and has projections reaching the hypothalamus and mid- and hindbrain regions. These connections allow integration of sensory signal and can produce the necessary signaling to induce responses to maintain homeostasis following stress. This signaling occurs through various neurotransmitters. The longest lasting and farthest reaching of these are neuropeptides. Two neuropeptides, corticotropin-releasing hormone (CRH) and neuropeptide Y (NPY), have been shown to alter anxiety behavior and HPA axis function. These changes are mediated through multiple receptors. This dissertation endeavors to understand how certain CRH and NPY receptors are regulated by glucocorticoid and chronic stress. The receptors to be investigated in this thesis include the CRH type 2 receptor (CRH-R2), NPY type 1 receptor (NPY-Y1) and the glucocorticoid induced receptor (GIR). In vitro analysis was used to investigate changes in the expression of CRH-R2, NPY-Y1 and GIR following glucocorticoid exposure. CRH-R2 and GIR could be down regulated by glucocorticoids and NPY-Y1 was up regulated. Possible mechanisms for these dexamethasone-mediated changes are discussed. In vivo studies determined the regulation of CRH-R2, NPY-Y1 and GIR expression by chronic stress in the central and medial amygdala, both known to mediate anxiety behavior and HPA axis function. The Chronic Variable Stress (CVS) paradigm used is known to elicit changes similar to those observed in depression, such as altered HPA axis function, increased anxiety and anhedonia. Alteration in neuropeptide receptor expression could contribute to these changes. In the central amygdala, CRH-R2 is down regulated following 14 days of CVS, while NPY-Y1 and GIR had no effect. In the medial amygdala, GIR is up regulated while CRH-R2 and NPY-Y1 are unchanged.

We have known since study of the lymphocyte and plasma cells really began in earnest in the early 1940's that the pituitary adrenal axis under intimate control of the hypothalamus could influence immunological functions profoundly. We have also known for at least 20 years in my recollection that female sex hor mones can maximize certain immunity functions while male sex hormones tend to suppress many immunological reactions. The thyroid hormones accelerate antibody production while at the same time sp~eding up de gradation of antibodies and immunoglobulins and thyroidectomy decreases the rate of antibody production. Further, much evidence has accumulated indicating that the brain, yes even the mind, can influence in significant ways susceptibility to infections, cancers and to development of a variety of autoimmune diseases. More than 20 years ago, my colleagues and I convinced ourselves, if no one else, that hypnosis can exert major in fluences on the effector limb of the classical atopic allergic reactions. We showed with Aaron Papermaster that the Prausnitz-Kustner reaction may be greatly inhibited, indeed largely controlled, by post-hypnotic suggestion. And it was not even necessary for us to publish our discovery because scientists in John Humphrey's laboratory at Mill Hill Research Center in London had beaten us to the punch. They described hypnotic control of both the PK reaction and delayed allergic reactions to tuberculin by hypnosis.
