Recently, this class of signalling molecules and receptors has been found to mediate communication between neurons and non-neuronal cells (glia) in the central and peripheral nervous systems. Glia are critical for normal brain function, development and response to injury. Neural impulse activity is detected by glia and purinergic signalling is emerging as a major means of integrating functional activity between neurons, glia and vascular cells in the nervous system. These interactions mediate effects of neural activity on the development of the nervous system and in association with injury, neurodegeneration, myelination and cancer. Book excerpt: Over the last two decades, the recognition that astrocytes - the predominant type of cortical glial cells - could sense neighboring neuronal activity and release neuroactive agents, has been instrumental in the uncovering of many roles that these cells could play in brain processing and the storage of information. These findings initiated a conceptual revolution that leads to rethinking how brain communication works since they imply that information travels and is processed not just in the neuronal circuitry but in an expanded neuron-glial network. On the other hand the physiological need for astrocyte signaling in brain information processing and the modes of action of these cells in computational tasks remain largely undefined. This is due, to a large extent, both to the lack of conclusive experimental evidence, and to a substantial lack of a theoretical framework to address modeling and characterization of the many possible astrocyte functions. This book that we propose aims at filling this gap, providing the first systematic computational approach to the complex, wide subject of neuron-glia interactions. The organization of the book is unique insofar as it considers a selection of “hot topics” in glia research that ideally brings together both the novelty of the recent experimental findings in the field and the modelling challenge that they bear. A chapter written by experimentalists, possibly in collaboration with theoreticians, will introduce each topic. The aim of this chapter, that we foresee less technical in its style than in conventional reviews, will be to provide a review as clear as possible, of what is “established” and what remains speculative (i.e. the open questions). Each topic will then be presented in its possible different aspects, by 2-3 chapters by theoreticians. These chapters will be edited in order to provide a “priming” reference for modeling neuron-glia interactions, suitable both for the graduate student and the professional researcher.

The nervous system of the freshwater crayfish, Procambarus clarkii was the preparation of choice for most of the studies the axon has been well characterized electrically and metabolically by a number of investigators including the P.I. The glial cell investment of the crayfish medial giant axon has been partially characterized by this investigator. At the time this research program was started the crayfish was only one of two such axon-Schwann cell preparations characterized for this work. The other being a tropical squid axon Schwann cell preparation not available in this country. In latter phases of this research program the emphasis, as originally planned, was shifted to the development of a mammalian cell culture model for axon-Schwann cell interaction studies for the purpose of determining the generality of our findings in the invertebrate systems. Book excerpt: "A fundamental aspect of biological organisation is intercellular communication, which is necessary for the coordination and function of multicellular assemblies, and ultimately, for the survival of organisms in an everchanging environment. Within the developing nervous system, the different cell types - neuronal and glial - must work together for the formation, maintenance, and refinement of brain circuits. To further our understanding of activity-dependent circuit plasticity, two distinct phenomena of neuron-glia interactions in the developing visual system were investigated. First, the developmental progression of early myelin ensheathment in the Xenopus laevis was characterised. Based on this work, an appropriate developmental window was identified to investigate myelin plasticity, specifically, by examining the effects of early patterned sensory visual experience on myelin ensheathment in the Xenopus retinotectal circuit. We observed that patterned sensory experience evoking robust neuronal activity also increased early myelin ensheathment, whereas dampened neuronal activity reduced it. These findings thus lend support to the possibility of shared mechanisms for the control of timing-dependent plasticity and myelination during circuit development. In parallel, we explored the role of D-serine, which is a co-agonist of the N-methyl-D-aspartate glutamate receptor (NMDAR) and its availability is regulated by neuron-glia interactions. D-serine administration appears to prematurely stabilise the dendritic arbours and synapses of recently differentiating tectal neurons, though chronic exposure may also lead to functional compensation in overall synaptic density and visually evoked calcium responses in the optic tectum. Thus, the neuron-glia-dependent availability of D-serine at glutamatergic synapses may contribute to activity-dependent NMDAR-mediated refinement of the developing tectal circuit. By examining some of the neuron-glia interactions underlying activity-dependent plasticity, this dissertation aims to contribute to our understanding of the complex and dynamic processes necessary for biological systems to function optimally in their environment"--
