Book excerpt: One of the primary functions of the human auditory system is to separate the complex mixture of sound arriving at the ears into neural representations of individual sound sources. This function is thought to be crucial for survival and communication in noisy settings, and allows listeners to selectively and dynamically attend to a sound source of interest while suppressing irrelevant information. How the brain works to perceptually organize the acoustic environment remains unclear despite the multitude of recent studies utilizing microelectrode recordings in experimental animals or non-invasive human neuroimaging. In particular, the role that brain areas outside the auditory cortex might play is, comparatively, vastly understudied. The experiments described in this thesis combined classic behavioral paradigms with electrical recordings made directly from the cortical surface of neurosurgical patients undergoing clinically-indicated invasive monitoring for localization of epileptogenic foci. By sampling from widespread brain areas with high temporal resolution while participants simultaneously engaged in streaming and jittered multi-tone masking paradigms, the present experiments sought to overcome limitations inherent in previous work, namely sampling extent, resolution in time and space, and direct knowledge of the perceptual experience of the listener. In experiment 1, participants listened to sequences of tones alternating in frequency (i.e., ABA-) and indicated whether they perceived the tones as grouped ("1 stream") or segregated ("2 streams"). As has been reported in neurologically-normal listeners since the 1950s, patients heard the sequences as grouped when the frequency separation between the A and B tones was small and segregated when it was large. Evoked potentials from widespread brain areas showed amplitude correlations with frequency separation but surprisingly did not differ based solely on perceptual organization in the absence of changes in the stimuli. In experiment 2, participants listened to sequences of jittered multi-tone masking stimuli on which a regularly-repeating target stream of tones was sometimes superimposed and indicated when they heard the target stream. Target detectability, as indexed behaviorally, increased throughout the course of each sequence. Evoked potentials and high-gamma activity differed strongly based on the listener's subjective perception of the target tones. These results extend and constrain theories of how the brain subserves auditory perceptual organization and suggests several new avenues of research for understanding the neural mechanisms underlying this critical function.

At the same time, much of the functional role of the various sub-areas in human auditory cortex is still relatively unknown, and a more sophisticated understanding is only now emerging through the use of contemporary electrophysiological and neuroimaging techniques. The integration of results across the various techniques signify a new era in our knowledge of how human auditory cortex forms basis for auditory experience. This volume on human auditory cortex will have two major parts. In Part A, the principal methodologies currently used to investigate human auditory cortex will be discussed. Each chapter will first outline how the methodology is used in auditory neuroscience, highlighting the challenges of obtaining data from human auditory cortex; second, each methods chapter will provide two or (at most) three brief examples of how it has been used to generate a major result about auditory processing. In Part B, the central questions for auditory processing in human auditory cortex are covered. These perceptual representations provide information on the putative sound sources in the environment, which are combined with knowledge of auditory categories to both imbue sounds with meaning and inform appropriate behavioral actions. Critical aspects regarding the cortical mechanisms responsible for regularity representation and perception as well as how prior knowledge of auditory categories influences perceptual judgments remain unanswered. This dissertation had two main goals: (1) to test how neural activity encodes regularity representation and perception; and (2) to test how a listener uses prior category knowledge to inform categorical judgments when a stimulus’ category membership is ambiguous. To achieve these goals, I employed a combination of neurophysiological, behavioral, and computational analyses in humans. I found that the phase of population-level neural activity is a more reliable indicator of regularity than power and that a variety of brain regions exhibited reliable modulations that distinguished stimulus and behavioral differences related to regularity violation. Additionally, I found that human listeners learn approximations of auditory categories and are varied in their ability to use prior category information to inform categorical judgments. Finally, I found that categorization behavior was consistent with an ideal decision strategy that includes trial-by-trial variability in a listener’s estimates of the prior probability of each category. These findings build upon previous work on the mechanisms underlying regularity processing in auditory perception and that future research should focus on a variety of brain regions beyond the classical auditory pathways in cortex. Additionally, the categorization findings are the first to extend previous work in visual categorization into the auditory domain and reformulates the issue of categorization in a manner that can help to interpret the results of previous research within a generative framework.

Book excerpt: Since the 1950s, there has been continued advancement in technology for obtaining long-term neural recordings in mammals, including present-day techniques yielding data sets with increasing spatial and temporal resolution. Discovering dynamical patterns encoded in high fidelity neural measurements is a challenging, yet rewarding, task equipped with techniques applicable to data obtained with various methods. Emerging approaches employ data-driven methods to identify low-dimensional structures from large-scale recordings, which encapsulate key characteristics of the measured neural populations. In this thesis, we consider electrocorticography recordings in primary auditory cortex from human patients performing an auditory streaming task designed to elicit bistable perception. Listeners are presented with 5-minute sequences of alternating pure tones and self-report changes in their perception of the stimulus. Our goal is to analyze the neural recordings obtained concurrently during stimulus presentation for correlates with subject-reported perception. To this end, we employ the data-driven methods of (1) time-delay coordinates, (2) diffusion maps, and (3) the extended dynamic mode decomposition to yield a low-dimensional collection of Fourier-like features, which are adapted to intrinsic dynamics encoded in the neural data. We find two sets of features, one which correlates with the stimulus, and one with the reported percept. In particular, we characterize a slow rhythm correlated with switches in perception that manifests and modulates the phase of a subset of the derived features. By leveraging the dynamical techniques, we identify neural characteristics associated with perception, which may remain undetected by traditional static methods from data analysis. The data-driven methods implemented in this thesis can be applied in contexts outside of neuroscience and are part of the growing movement towards equation-free modeling and analysis. This emerging paradigm is highly applicable to physical processes or systems that exhibit rich dynamics but are accompanied by incomplete, or entirely unknown, governing equations.

The first three steps in the anal ysis of these complex waveforms are proper placement of electrodes for recording, the proper choice of electrical or sensory stimulus parameters, and the establish ment of behavioral control. The fourth is development of techniques for reliable measurement. Measurement consists of comparison of an unknown entity with a set of standard scales or dimensions having numerical attributes in preassigned degree. A physical object can be described by the dimensions of size, mass, density, etc. In addition there are dimensions such as location, velocity, weight, hardness, etc. Some of these dimensions can be complex (e. g. size depends on three or more subsidiary coordi nates), and some can be interdependent or nonorthogonal (e. g. specification of size and mass may determine density). In each dimension the unit is defined with refer ence to a standard physical entity, e. g. a unit of mass or length, and the result of measurement is expressed as an equivalence between the unknown and the sum of a specified number of units of that entity. The dimensions of a complex waveform are elementary waveforms from which that waveform can be built by simple addition. Any finite single-valued function of time is admissible. They are called basis functions (lO, 15), and they can be expressed in numeric as well as geometric form.
