Sur ce dernier point, la plupart des travaux portent sur des nanoparticules à base d'or ou d'oxyde de fer. Une alternative originale aux nanoparticules dejà étudiées, pour combiner diagnostic et thérapie, pourrait concerner des nanoparticules hybrides à base d'oxyde de gadolinium : Gd203@SiOx[fluo]. Ces particules peuvent être détectées à la fois par luminescence grâce aux fluorophores organiques liés au réseau de polysiloxane enrobant le coeur et par des méthodes magnétiques, attendu que le gadolinium est largement utilisé comme agent de crontraste en IRM. De plus, son isotope 157 est cartaérisé par une forte section efficace de capture des neutrons, ces particules d'oxyde de gadolnium apparaissent comme un substitut très prometteur aux agents neutrophages actuellement utilisé en neutronthérapie. Les études ont montrées que ces nanoparticules pouvaient être suivis par des techniques d'imagerie optique et par résonance magnétique ; en in vitro, où les nanoparticules hybrides se sont relevées être des marqueurs cellulaire ; en in vivo, où, grâce à des modifications de la couche externe, ces nanoparticules ont pu être injectées par voie intraveineuse et dont leur suivis n'a pas révélé d'accumulation indésirable. Il ressort également de cette étude que les nanoparticules de Gd203@SiOx permettent d'améliorer la thérapie par capture neutronique

La tendance actuelle consiste à combiner plusieurs techniques d'imagerie complémentaires pour exploiter les avantages de chacune d'elles tout en surmontant leurs limites. Parmi les nombreuses possibilités, la combinaison de l'imagerie par résonance magnétique (IRM) et de la tomographie par émission de positon (TEP) semble très intéressante, car elle allie la haute résolution de l'IRM à la sensibilité exceptionnelle de la TEP. Si le développement de ce dispositif constitue en soi un défi de taille, la conception de sondes multimodales constitue également une étape essentielle pour l'exploitation d'imageurs intégrés IRM/TEP. Pour réaliser le développement de sondes multimodales pour l'imagerie simultanée IRM/TEP, deux stratégies basées sur le caractère multifonctionnel des nanoparticules ont été explorées, en gardant à l'esprit que ces agents d'imagerie doivent être éliminés par clairance rénale (Dh ˂ 10 nm). La première a consisté en la synthèse de nanoparticules d'or ultrapetites revêtues par deux types différents de chélateur pour une complexation sélective des ions gadolinium (IRM) et des émetteurs de positons (TEP). La deuxième stratégie a reposé sur la réduction d'un sel d'or en présence de molécules dithiolées contenant deux sites de complexation spécifiques. La réduction du sel d'or en présence d'un mélange de deux différents chélateurs ou en présence de molécules dithiolées contenant deux sites de complexation spécifiques a fourni des nanoparticules d'or ultrapetites (taille du cœur d'or (TEM) : 2 à 3 nm et diamètre hydrodynamique (DLS) : 6 - 8 nm) capables d'immobiliser à la fois les ions gadolinium et les ions cuivre-64. En conséquence, la biodistribution de ces nanoparticules a pu être suivie simultanément par IRM et TEP sur un même animal avec le même dispositif d'imagerie IRM/TEP intégré et après une seule injection intraveineuse. Cette étude a également confirmé une libre circulation des nanoparticules et une excrétion rénale sans aucune accumulation indésirable dans l'organisme sain. Ce travail a montré que les nanoparticules d'or ultrapetites revêtues par deux types différents de chélateurs ou des molécules contenant deux sites de complexation spécifiques possèdent un fort potentiel pour l'imagerie bimodale IRM/TEP.
