Book excerpt: Talin is an essential cytoskeletal protein present in cell-matrix type adherens junctions where it helps in attachment of the actin cytoskeleton to the plasma membrane. The nature of the interaction between talin and actin was examined in this dissertation by investigating three actin binding sites (ABS1-3), with ABS1 located in the 47 kDa N-terminal head and ABS2 and ABS3 present in the 190 kDA C-terminal tail of the talin molecule. Cosedimentation of expressed GST-fusion deletion constructs from the talin head with F-actin showed that actin binding activity of the talin head is contained within residues 271-320. Talin constructs containing the narrowed ABS1 exhibited less pH dependence compared to those containing ABS3. Both ABS1 and ABS3 showed high dependence on ionic strength in ability to bind actin. Nearly all of the N-terminal head fragment purified from calpain-cleaved talin cosedimented with F-actin under conditions (pH 6.4 and low ionic strength) found to be optimal for the talin-actin interaction. EGFP-fusion talin constructs containing ABS1 were primarily colocalized with actin stress fibers when transfected into COS cells, whereas EGFP-ABS3 was predominantly colocalized with actin-rich membrane ruffles. The ABS2 and ABS3 within the C-terminal tail were further defined to residues 1051-1250 (ABS2) and 2331-2530 (ABS3), respectively. ABS2 exhibited high dependence on both pH and ionic strength in interacting with actin, which is similar to that found with ABS3. Individual transfections of EFGP-ABS2 and EGFP-ABS3 into COS cells showed that ABS2 was primarily colocalized with actin stress fibers, whereas ABS3 was colocalized with actin filaments at both membrane ruffles and microspike-like cell extensions. Site-directed mutagenesis studies on ABS1 showed that substitution of alanine for the five positively charged residues K295, R297, K300, R303 and K306, or for all six residues within 295-300 resulted in significant reduction in actin binding. Taken together, these results suggest that the ABS1 located within the head domain, and the ABS2 and ABS3 present in the C-terminal tail domain, of talin each may play distinct roles in the interaction of talin with actin. These differences may be important in regulating integrin-mediated cell adhesion and migration.

Book excerpt: The regulation of filamentous actin (F-actin) production from the polymerization of globular actin (G-actin) within the cell is critical for many cell functions. Since actin is found in all cells, understanding how actin-binding-proteins (ABPs) bind and how their regulating mechanisms work is not only important to the basics of cytoskeletal pathways, but also to understanding associated diseases and creating possible therapeutics to combat them. Cofilin is an ABP that plays an important part in the regulation process and in recent times, has come to be known as a player in maintaining a cell's homeostasis. It's activity has been shown to have implications in many diseases, such as Alzheimer's and certain cancers. Cofilin binds and severs actin filaments, leading to depolymerization as well as the creation of new barbed ends. Although some of the details of cofilin's interaction with G-actin have been illuminated through a range of experimental studies, the specific interactions with F-actin have remained much more elusive. As of yet, there are only cryoEM models of cofilin-bound F-actin (where the binding occurs at a 1:1 ratio), which are not high enough resolution and do not show molecular interactions. The focus of this research is to build a model of how cofilin binds F-actin and understand the mechanism of severing. Computational methods, such as protein-protein docking, all atom molecular dynamics (AA MD) simulations, and Coarse Grain MD (CG MD) can help in understanding the interactions between cofilin and F-actin. Iteratively combining these methods with biochemical and mutagenesis experiments to reach a consensus offer a guide towards a more cogent answer. Here in this dissertation, I describe how I built a cofilin and F-actin binding model, with the aid of empirical data. This work allowed me to create several filament models with varying number of bound cofilin, which replicates different binding states of the filament. I also simulated the dynamics of these filaments models to gain insight into filament behavior, particularly twist.

I was tempted to write back in my rebuttal to the reviewer that I didn’t realise it had been lost. Uncharacteristic maturity prevailed and I resisted the temptation, but I was struck by the underlying observation that research on the str- ture and function of tropomyosin has been somewhat invisible, particularly in terms of the cytoskeleton isoforms. So, how can it be that one of the two major components of the actin filament has been so thoroughly overlooked? I suspect that the answer is disappointingly pedestrian. Whereas the biochemistry of the 1980s revealed the potential of tropomyosin isoforms to diversify the function of actin filaments, the subsequent disenchantment with isoform biology in general in the 1990s inhibited growth of this field. With the development of more sophisticated experimental - proaches we are now seeing a growing realisation of the importance of tropomyosin in regulating actin filaments beyond its pivotal role in muscle contraction. The opportunity to edit this book came at a time when we had written several reviews on different aspects of tropomyosin function and I had just finished the background reading for a comprehensive review of tropomyosin biology. I realised that the field was simply beyond the capacity of any one person to do the field j- tice.

Sliding filaments and the working-stroke mechanism provide the framework for individual myosin motors to act in parallel, generating tension and loaded shortening with an efficient use of chemical energy. Our knowledge of this exquisitely structured molecular machine has exploded in the last four decades, thanks to a bewildering array of techniques for studying intact muscle, muscle fibres, myofibrils and single myosin molecules. After reviewing the mechanical and biochemical background, this monograph shows how old and new experimental discoveries can be modelled, interpreted and incorporated into a coherent mathematical theory of contractility at the molecular level. The theory is applied to steady-state and transient phenomena in muscle fibres, wing-beat oscillations in insect flight muscle, motility assays and single-molecule experiments with optical trapping. Such a synthesis addresses major issues, most notably whether a single myosin motor is driven by a working stroke or a ratchet mechanism, how the working stroke is coupled to phosphate release, and whether one cycle of attachment is driven by the hydrolysis of one molecule of ATP. Ways in which the theory can be extended are explored in appendices. A separate theory is required for the cooperative regulation of muscle by calcium via tropomyosin and troponin on actin filaments. The book reviews the evolution of models for actin-based regulation, culminating in a model motivated by cryo-EM studies where tropomyosin protomers are linked to form a continuous flexible chain. It also explores muscle behaviour as a function of calcium level, including emergent phenomena such as spontaneous oscillatory contractions and direct myosin regulation by its regulatory light chains. Contraction models can be extended to all levels of calcium-activation by embedding them in a cooperative theory of thin-filament regulation, and a method for achieving this grand synthesis is proposed.
