Book excerpt: In Molecular Diagnostics for Melanoma: Methods and Protocols, expert researchers and clinicians in the field of melanoma provide updated information on biomarkers and assays for diagnosis, prognosis, and assays predicting response to treatment for routine testing. The focus of the volume is on biomarkers with established clinical validity rather than those on early discovery stage. With additional in-depth discussion of the molecular biology and pathology of melanoma, treatment options in adjuvant and metastatic setting, and implications of biomarker testing for clinical management of melanoma patients. Written in the highly successful Methods in Molecular Biology series format, chapters include extensive introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and key tips on troubleshooting and avoiding known pitfalls. Comprehensive and practical, Molecular Diagnostics for Melanoma: Methods and Protocols seeks to provide both clinicians and scientists with technical information and extensive background information on the wide ranging approaches available in the field of diagnostics of melanoma.

Available in PDF, EPUB and Kindle. Book excerpt: This book aims to bring together a broad variety of examples of the role of pharmacogenomics in current drug development, uncovering dynamic concentration-dependent drug responses on biological systems to understand pharmacodynamics responses in human cancer where genetic lesions serve as tumor markers and provide a basis for cancer diagnosis. The book describes methods and protocols applied in molecular diagnostics. It offers pathologists and researchers providing molecular diagnostic services an array of the most recent and readily accessible reference to compare methods and techniques. Highlights include the molecular diagnosis of genetic aberrations by quantitative polymerase reaction (qPCR), sequence-specific oligonucleotide arrays, next-generation sequencing (NGS), CGH arrays-and methodologies directed at the detection of epigenetic events, high-throughput nucleic acid and protein arrays, direct sequencing and FISH-based methodologies, currently used in the diagnosis of solid tumors. The book also includes an innovative line of treatment in relation to the molecular prognosis, diagnosis and pharmacogenomics in the actual practice of clinical findings at molecular levels. The book covers the applications of numerous genetic testing methodologies; in approximately the chronological order of discovery and high-throughput diagnosis using advanced genomic approaches to identify such genes, in the search for novel drug targets and/or key determinants of drug reactions. It also promotes a wider understanding of molecular diagnostics among physicians, medical students, and scientists in academics, industry and corporate world.

Book excerpt: Molecular diagnostics is an exploding field, and recent advances in our understanding of the molecular basis of disease have provided a platform for the development of new diagnostic tests as well as tests to predict tumor behavior and potential response to targeted therapy. This textbook provides a reference and practical guide to molecular diagnostics for dermatologists and dermatopathologists. It outlines our current understanding of the molecular underpinnings of dermatologic disease, describes the appropriate use of currently available molecular tests, and explains the interpretation of these tests in the context of diagnosis and management. Tests relating to various disorders are covered, including but not confined to melanoma, genodermatoses, and infectious disease. Pitfalls are highlighted and user-friendly algorithmic approaches, presented.

Book excerpt: This book is about melanoma—its biology, immunology, and pathology, as well as the initial use of powerful genomic tools to study its fundamental mole- lar and genetic characteristics. The study of cancer will be profoundly impacted by the Human Genome Project. I would like to discuss some of these changes. The first draft of the human genome sequence was announced in June 2000, and we have just scratched the surface of the changes it will engender in medicine. A relevant question is what are the long-term effects of the Human Genome Project for medicine? I would argue that there are three, and each of these three point toward the view that systems biology will dominate biology and medicine of the 21st century. First, the Human Genome Project introduced a new type of s- ence—discovery science. Discovery science takes a biological system (e. g. , the genome) and defines all of its elements (e. g. , the sequences of the 24 human ch- mosomes). Thus, it creates a rich infrastructure from which the classical hypo- esis-driven science can be done more effectively. The effective integration of discovery- and hypothesis-driven science is a key for systems approaches to bi- ogy and medicine. Second, the Human Genome Project has provided a “periodic table of life.

In addition, new molecular techniques are entering the clinical arena for their value in predicting therapy response and tumor prognosis. This book provides a guide for the practicing pathologist and for both pathology residents and fellows during the daily sign-out of challenging cases. The book is organized by anatomical systems and provides a detailed description of molecular tests that may help in the diagnosis. Furthermore, a description of the current molecular tests required to identify patients for treatment is offered. The application of molecular pathology techniques to the clinical practice has already shown its usefulness and the number of such tests is growing exponentially as more molecular targets are discovered. Molecular Pathology and Diagnostics of Cancer will give practicing and training pathologists an up-to date resource to guide the correct management of pathology cases requiring molecular testing.

Book excerpt: Molecular Diagnostics in Dermatology and Dermatopathology presents the basics of molecular biology and molecular diagnostic methods most commonly used in the clinical laboratory, with an emphasis on the concepts and testing most relevant to dermatological diseases. Topics include the integration of newer diagnostic and prognostic techniques with ‘traditional’ histologic approaches, and discussions of regulatory, ethical, legal, economic issues and ‘newer’ technologies. This important diagnostic tool outlines the clinically relevant uses (i.e.; diagnostic, staging and/or prognostic) applications of these techniques in the field of dermatology. Molecular studies that investigate the pathogenesis of skin diseases will be excluded, unless they also have a direct diagnostic utility. The book will be of interest to practicing pathologists, dermatology and pathology residents, dermatologists, and dermatopathologists.

It aims to address the practical questions frequently encountered in the molecular oncology practice, as well as key points and pitfalls in the clinical interpretation of molecular tests in guiding precision cancer management. The text uses a Q&A format and case presentations, with emphasis on understanding the molecular test methods, diagnosis, classification, risk assessment and clinical correlation. Starting with an update on the molecular biology of cancer, the book focuses on the topics related to molecular diagnostics and genetics-based precision oncology. Separate chapters are dedicated to discussion of the bioinformatics for the analysis of genetic/genomic data generated from molecular assays, and quality control (QC)/quality assurance (QA) programs in the clinical laboratories; both are critical in producing high quality results for clinical care of cancer patients. These are followed by organ system-based reviews and discussions on the molecular genetic abnormalities and related tests covering diverse types of common to rare malignant neoplasms. This book also provides up-to-date knowledge related to malignant neoplasms, discusses the established as well as evolving requirements for pathologic diagnosis of these malignancies. It also discusses the cost effective utilization of molecular tests in clinical oncology. Written by experts in the field, Practical Oncologic Molecular Pathology serves as a valuable reference for practicing pathologists, fellows, residents and other health care professionals.

Book excerpt: This book describes both the technologies used in the discovery of melanoma biomarkers and the clinical application of these biomarkers for diagnosis and staging of disease, determination of prognosis, treatment planning, monitoring of response to therapy, identification of novel therapeutic targets and drug development. A broad range of biomarkers (DNA/chromosomal, mRNA, microRNA, mitochondrial DNA, epigenetic and protein) is outlined. As therapies for melanoma become increasingly more target specific, the identification, validation and use of biomarkers will invariably play a greater role in the management of patients with this disease. Diagnostic and Prognostic Biomarkers and Therapeutic Targets in Melanoma is an essential resource for oncologists, dermatologists, dermatopathologists, general pathologists with an interest in melanoma, and melanoma researchers.
