Abstract: Abstract of thesis entitled MOLECULAR CLONING AND FUNCTIONAL CHARACTERIZATION OF GOLDFISH ALPHA-2 ADRENERGIC RECEPTORS Submitted by Chan Hoi Yan for the degree of Master of Philosophy at The University of Hong Kong in September 2004 In mammals, adrenergic stimulation at the hypothalamic level through alpha-2 adrenergic receptors can induce growth hormone secretion from the pituitary. However, the alpha-2 receptor subtype(s) responsible for this stimulatory action has not been elucidated. In the goldfish, we have previously shown that alpha-2 activation at the pituitary level can suppress growth hormone release and that removal of alpha-2 inhibition can trigger a rebound of growth hormone secretion. These findings have prompted us to speculate that alpha-2 adrenergic receptors should be expressed in goldfish pituitary cells and be responsible for the growth hormone regulation by adrenergic input at the pituitary level. As a first step to study the alpha-2 receptor subtype(s) expressed in the goldfish pituitary, molecular cloning was performed to establish the structural identity of the goldfish alpha-2A, alpha-2B, and alpha-2C adrenergic receptors. By library screening and 3'/5' RACE, the complementary DNAs (cDNAs) for alpha-2A, alpha-2B, and alpha-2C adrenergic receptors with the sizes of 2.72 Kb, 2.86 Kb, and 1.72 Kb, respectively, have been isolated from the brain-pituitary axis of the goldfish. The open-reading frames of these cDNAs encode proteins of 383 to 505 amino acids in size with seven transmembrane domains typical of those of the G protein-coupled receptors. Sequence analysis also reveals that these goldfish alpha-2A, alpha-2B, and alpha-2C adrenergic receptors are structurally homologous to the corresponding receptor subtypes in mammals. By reverse transcription polymerase chain reaction (RT-PCR), the transcripts of alpha-2A and alpha-2B adrenergic receptors were identified in many tissues in the goldfish, including the heart, liver, gill, intestine, kidney, muscle, spleen, pituitary, and various brain segments. A similar expression pattern was also noted for alpha-2C adrenergic receptor, except that its mRNA was not detected in the intestine and its expression in the brain was restricted to the brain stem and spinal cord. To characterize the pharmacological and biochemical properties of these newly cloned receptors, functional expression of goldfish alpha-2A, alpha-2B, and alpha-2C adrenergic receptors was conducted in Chinese hamster ovary (CHO) cells. In this case, the receptor binding properties of these goldfish adrenergic receptors were tested using a radioreceptor binding approach. Although the order of affinity of the respective alpha-2A, alpha-2B, and alpha-2C adrenergic receptors is different from that reported in mammals, these receptors consistently exhibited a high affinity for ligands selective for alpha-2 but not for alpha-1 and beta adrenergic receptors. Furthermore, activation of these receptors by norepinephrine or the alpha-2 agonist + + clonidine could trigger rapid activation of the Na /H exchanger, an inhibition of 2+ cyclic AMP production and a decrease in intracellular Ca levels. Abstract: Abstract of thesis entitled CLONING AND CHARACTERIZATION OF PAC1 RECEPTOR SPLICE VARIANTS IN GOLDFISH (CARASSIUS AURATUS) Submitted by Kwok Yuen Yuen for the degree of Master of Philosophy at The University of Hong Kong in August 2004 It has already been demonstrated in several differnet vertebrates that alternative splicing of pituitary adenylate cyclase-activating polypeptide (PACAP) receptors can generate different types of variants. Through various signal transduction pathways, these isoforms regulate the release of neuroendocrine hormones, like growth hormone and gonadotropin. Since PACAP and its receptors are highly conserved through vertebrate evolution, it would be interesting to characterize PACAP receptor splice variants in lower vertebrates. Goldfish PACAP type I receptor (goldfish PAC -R-short) has been cloned previously. Based on this previous information, as well as knowledge on the splice variants of other species, goldfish PAC -R-hop1 variant was characterized in the present study. This hop1 cassette shares 71.4% homology to its counterpart in mammalian species. Functional expression of goldfish PAC -R-short and goldfish PAC -R-hop1 in mammalian 1 1 Chinese hamster ovary (CHO) cells showed that the intracellular cAMP production was elevated with EC values of 8.7 1.5 nM and 8.8 1.9 nM, respectively, upon 50 stimulation by ovine PACAP . Quantitative RT-PCR revealed that both goldfish 38 PAC -R-short and goldfish PAC -R-hop1 were highly expressed in the brain, heart, 1 1 pituitary and male gonad, with goldfish PAC -R-short as the predominant form. In situ hybridization of goldfish PAC -R in the pituitary revealed its prevalence in the pars distalis. These distribution studies presented an expression profile of the receptor which would be useful for elucidating the pleiotropic effect of PACAP. Together, characterization of the splice variants would provide some clues to the physiological roles, as well as the evolution of the receptor in vertebrates. Part of this mechanism is mediated via steroid/ thyroid hormone modulation of growth hormone (GH) synthesis at the pituitary level. In this study, using a static incubation approach, a systematic survey was conducted in vitro to examine the effects of sex steroids, adrenocorticoids, and thyroid hormone on GH mRNA expression in grass carp (Ctenopharyngodon idella) pituitary cells. Sex steroids, including estradiol, testosterone, and the estrogen agonist diethylstilbestrol, increased GH mRNA levels in grass carp pituitary cells. These stimulatory actions reached their peaks at submicromolar concentrations. In a similar dose range, progesterone did not affect basal GH mRNA expression and a slight inhibition was observed when the doses were increased to micromolar levels. Glucocorticoids, including hydrocortisone, corticosterone, and the synthetic analog dexamethasone, suppressed GH mRNA levels when tested at high micromolar doses. Unlike glucocorticoids, a slight elevation in GH mRNA levels was noted when pituitary cells were exposed to submicromolar doses of aldosterone. In parallel studies, retinoic acid and thyroid hormone did not alter basal GH mRNA expression in grass carp pituitary cells. Data provides evidence that steroid hormones can differentially regulate GH gene expression in grass carp by acting directly at the pituitary cell level. Among the hormones tested, estradiol was a potent stimulant for GH mRNA expression. Molecular cloning of grass carp estrogen receptors (ER) was also performed. Using 5' and 3' RACE, three isoforms of grass carp ERs, namely ERα, ERβ-S and ERβ-L, were cloned. They were highly homologous to the ERα and ERβ reported in other fish species. The full-length cDNAs of grass carp ERα (1692 bp), ERβ-S (2091 bp) and ERβ-L (2707 bp) carried the open reading frames of ERs composing of 564, 605, and 697 amino acids, respectively. RT-PCR also revealed that transcripts for ERα and ERβs could be identified in a variety of tissues in the carp, including the brain, pituitary, muscle, gonad, gill, heart, spleen, liver, and intestine. To characterize the pharmacological properties of the cloned ERs, functional expression of grass carp ERα, ERβ-S and ERβ-L was conducted in CHO-K1 cells. In the presence of estradiol, the ERs expressed were effective in activating a luciferase reporter gene with estrogen-responsive promoter elements. These stimulatory actions were specific for estradiol and could not be mimicked by parallel treatments with testosterone, progesterone, hydrocortisone, or thyroid hormone. Furthermore, estradiol-stimulated luciferase expression in CHO-K1 cells with expressed ERα, ERβ-S and ERβ-L could be partially inhibited or abolished by a simultaneous treatment with the estrogen antagonists, tamoxifen and ICI182780. In the past decade, extragonadal expression of LHR has been reported with exciting findings on the novel functions of LH in non-gonadal tissues. In our recent studies with grass carp as a model for modern day bony fish, endogenous LH was found to act locally at the pituitary level to induce GH secretion and GH gene expression. To further extend our understanding on this unique paracrine interaction within the carp pituitary, the structural identity of carp LHR was established by molecular cloning. The receptor specificity of this newly cloned LHR was confirmed by functional expression in CHO cells. Using RT-PCR, LHR expression was detected in mixed populations of carp pituitary cells as well as in immuno-identified somatotrophs isolated by laser capture microdissection. During our cloning exercise, a splice variant of the full-length LHR (LHR-L) with LRR truncation in N-terminal 1-3 ectodomain was isolated. This LHR variant, namely LHR-S, is unique and has never been reported in other vertebrates. In contrast to LHR-L, which was shown to be 2+ 2+ 2+ functionally coupled to cAMP synthesis, [Ca ]e entry via Ca channels, and [Ca ]i mobilization by IP R and RyR activation, LHR-S stimulation did not trigger cAMP 2+ production nor Ca mobilization. LHR-S co-expression, however, was effective in 2+ attenuating the cAMP responses and Ca signals induced by LHR-L activation, suggesting that this novel variant of LHR may act as a dominant negative LHR in fish model. Based on the LHR sequence obtained, a real-time PCR system was set up to monitor LHR mRNA expression in grass carp pituitary cells. In these studies, dopaminergic stimulation could up-regulate transcript expression of LHR via pituitary D1 receptors coupled to the AC/cAMP/PKA pathway. Furthermore, LH produced locally in carp pituitary cells could suppress basal levels of LHR mRNA whereas exogenous LH was effective in blocking D1-stimulated LHR gene expression. The inhibitory effect of LH on dopamine D1 stimulation was found to occur selectively in the enriched fraction of carp somatotrophs, suggesting that the functional interactions between dopamine and LH may play a role in modulating the sensitivity of somatotrophs to local stimulation by endogenous LH. To shed light on the signaling mechanisms responsible for LH-induced GH gene expression, GH cells with transient expression of carp LHR was co-transfected with luciferase-expressing constructs carrying the 5' promoter of grass carp GH gene. The cell model was used to study the signaling pathways involved in GH gene transcription induced by LHR activation. Using 5' deletion analysis, the LH responsive sequence was mapped to position -616 and -572 of the grass carp GH promoter. Book excerpt: Spermatogenesis is a process highly conserved throughout vertebrate species and is mainly under hypothalamic-pituitary control. It occurs in the testis in a stepwise fashion so that committed spermatogonia develop into spermatocytes and enter meiosis to produce round spermatids. These undergo a morphological transformation (spermiogenesis) into mature spermatids (i.e.: spermatozoa), which are differentially released from Sertoli cells (spermiation) depending on the species. In mammals, further transformations are necessary to form mature spermatozoa, suitable for fertilization. Gonadotropins, mainly responsive to gonadotropin-releasing hormone, control spermatogenesis through specific receptors located at the gonadal level. However, besides the endocrine route, the chemical mediators may also act locally in the gonad. Indeed, it is documented that testis physiology, including steroidogenesis and spermatogenesis, does not fully account for traditional endocrine control but an intragonadal network of autocrine and/or paracrine regulators also exists, whose activity, via cell-to-cell communication, regulates germ cell progression and development of qualitatively mature spermatozoa. Of note, a number of testicular modulators, such as gonadotropin releasing hormone, Kiss-peptin, endocannabinoids, has been early isolated in the brain and latest in the gonads. To fully understand precise mechanisms underlying the functional interaction of this intricate network, needless to say, it is crucially required to have detailed information about modulators and target cells. Through synergy between the respective specializations of all the authors, this topic reviewed emerging knowledge about neuroendocrine and local mediators controlling germ cell progression and maturation.
