Book excerpt: Molecular and Cellular Enzymology addresses not only experienced enzymologists but also students, teachers and academic and industrial researchers who are confronted with enzymological problems during their fundamental or applied research. In this field there is an urgent need for training in order to meet the requirements of both research and industrial endeavours. This book consists of several levels. Practical aspects and elementary explanations are given for the benefit of non-specialists’ and students’ understanding. In order to facilitate the task of students, two typographies have been adopted. The main text corresponds to basic knowledge, whereas text in a smaller font provides more specialised information. Specialists will also find topics more deeply expounded with the principal bibliographic references cited. The bibliography, however, is not exhaustive; the choice includes general books and review articles as well as some specialised articles. In this book, for the first time, the different molecular and cellular aspects of enzymology are presented together. Until now, there has been no book available in which these different aspects are treated in the same volume. In addition, besides the theoretical developments, this book provides a wealth of practical information for experimentalists.

Hence, understanding the way they act is a prerequisite for understanding how a cell functions and how life evolves. Aspects such as the protein-ligand relationship, recognition, protein evolution by point mutation, enzyme-substrate interactions, behaviour of an enzyme in a living cell, control and dynamics of enzyme networks as well as the physico-chemical background of enzyme actions and multi-enzyme complexes are comprehensively treated in this volume.

Book excerpt: This book covers important advances in enzymology, explaining the behavior of enzymes and how they can be utilized to develop novel drugs, synthesize known and novel compounds, and understand evolutionary processes. Advances in Enzymology focuses on enzymes, the primary catalysts of life processes. The explanation of the behavior of enzymes can be found via studies of their chemical mechanisms and can be utilized to develop novel drugs, synthesize known and novel compounds, and understand evolutionary processes. The transglutaminases, first described in 1957, are a large, widely-distributed family of enzymes canonically responsible for the amidation/transamidation of protein side chains. The extraordinary diversity of names associated with various enzymatic activities now recognized and aggregated as transglutaminase bears witness to the remarkable diversity of biological roles associated with the activity, including myriad human diseases.

Book excerpt: The last fifteen years have witnessed the birth and maturation of many original methods and the development of protocols specific to single molecule measurements and their analysis, including techniques involving optical imaging, electron microscopy, optical and magnetic trapping, and developments in atomic force microscopy. In Single Molecule Enzymology: Methods and Protocols, experts in the field provide procedures which enable the extraction of detailed information about enzyme work cycles, their static and kinetic properties, and information about their location and activity within cells. The detailed volume offers practical advice on many aspects of single molecule enzymology and includes strategic overviews of interconnected methods involved in sample preparation, single molecule measurements, and data analysis. Written in the highly successful Methods in Molecular BiologyTM series format, chapters contain introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and up-to-date, Single Molecule Enzymology: Methods and Protocols is intended for use within the diverse community of molecular biologists, biochemists, and biophysicists studying enzymes in detail and can be used by researchers planning their first single molecule study or to aid more experienced researchers in further developing their existing studies.

Book excerpt: Prior to the start of the eighth meeting, I had the good sense to ask Professor Rosa Angela Canuto of Turin, Italy if she would help me organize the ninth meeting. She quickly suggested that both she and Dr. Guiliana Muzio, also of Turin, help plan the meet ing. Each of our previous eight meetings was a unique experience for the participants. The science was always outstanding and the presentations and discussions were excellent. By moving each meeting to a different part of the world we were able to experience exciting foods and cultural aspects of the world in addition to the science. The ninth meeting was no exception. We met from June 18 to 22 in the small mountain city of Varallo, Italy, the birth place of Dr. Canuto. Holding the scientific sessions in a several-hundred-year-old converted mansion and having an afternoon trip to either Lago Maggiore or Monte Rosa made some aspects of this meeting extremely memorable. An additional unique aspect of the social portion of the meeting was our ability to invite the townspeople to share with us a concert performed in an old church. Though the social and cultural aspects of the meeting were outstanding, the pur pose of the meeting was to exchange scientific information about the status of the three enzyme systems.
