Book excerpt: Ce travail présente la mise au point de deux microscopes à effet tunnel STM fonctionnant l'un dans l'air et en solution, et l'autre dans l'ultravide. Nous nous posons le problème de la signification des images et des différents essais d'interprétation. Dans le domaine de la biologie, nous montrons l'importance des différents paramètres (nature des sondes, nature des substrats, environnement) et présentons quelques méthodes de préparation utilisables pour deux échantillons: l'ARN et l'ADN. Trois méthodes ont été utilisées pour déposer l'ADN: microgoutte, dépôt par pulvérisation, électrodéposition. Ces expérimentations conduites dans l'ultravide ont démontré que l'observation STM (topographie et spectroscopie dI/dV) de molécules biologiques passait par la caractérisation des substrats et par la suppression des processus de formations d'agrégats. La seconde étude concerne l'observation par microscopie à effet tunnel en solution des structures secondaires et tertiaires de molécules d'acide ribonucléique (ARN). La réalisation d'un dispositif d'étude électrochimique comprenant des pointes isolées et une cellule spécifique, nous a permis d'éviter le problème de dénaturation et d'agrégation et nous a montre qu'il était possible de continuer l'observation des molécules en solution aqueuse. Nous avons pu caractériser les molécules d'ARNr par leurs paramètres structuraux et comparer ces valeurs a celles obtenues par EM et STEM et mis en évidence l'altération du substrat et le phénomène de conductance ionique en solution.
