Book excerpt: This detailed volume explores methods and protocols that aim to increase our understanding of how cells enter a quiescent state during homeostasis and how cells exit quiescence and re-enter differentiating cell divisions to restore damaged tissues, essential for developing new approaches in regenerative medicine in the future. The chapters in this book were designed to address cellular quiescence in prokaryote and eukaryote organisms, detection of quiescence (Hoechst/pyronin Y, FUCCI, CFSE, BrdU, H2B-GFP, CyTOF), quiescence in stem cells (skin, intestinal, neuronal, hematopoietic), genomic regulation (gene expression, transcription factors, lncRNA, RNA methylation), as well as analysis of the heterogeneity of quiescence by computer modeling. Written for the highly successful Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Cellular Quiescence: Methods and Protocols offers a broad view of basic and cutting-edge technology to inspire research in this emerging field of cell biology.

This is due to changes in dietary habits and the decrease in physical activity. Exercise is usually part of the prescription, the first line of defense, to prevent or treat metabolic disorders. However, we are still learning how and why exercise provides metabolic benefits in human health. This open access volume focuses on the cellular and molecular pathways that link exercise, muscle biology, hormones and metabolism. This will include novel “myokines” that might act as new therapeutic agents in the future.

Book excerpt: This volume describes the latest findings on transcriptional and translational regulation of stem cells. Both transcriptional activators and repressors have been shown to be crucial for the maintenance of the stem cell state. A key element of stem cell maintenance is repression of differentiation factors or developmental genes – achieved transcriptionally, epigenetically by the Polycomb complex, and post-transcriptionally by RNA-binding proteins and microRNAs. This volume takes two approaches to this topic – (1) illustrating the general principles outlined above through a series of different stem cell examples – embryonic, iPS and adult stem cells, and (2) describing several molecular families that have been shown to have roles in regulation of multiple stem cell populations.

Book excerpt: Given this pervasiveness and importance of miRNA-mediated gene regulation, it should come as little surprise that miRNAs themselves are also highly regulated. However, the recent explosion of knowledge on this topic has been remarkable, providing a primary motivation for publication of this book. As miRNAs are transcribed by RNA polymerase II, the enzyme that also generates mRNAs, it was perhaps not unexpected that miRNA transcription would be subject to regulation, and we have willfully mitted this aspect from this monograph. However, what has been unexpected is the extent of post-transcriptional regulation of miRNAs that is illustrated in this book.

Book excerpt: This book summarizes the latest findings on the functions of microRNAs in the regulation of plant development and responses to the surrounding environment. MicroRNAs are an important class of molecules that can be found in diverse groups of organisms, including plants and animals, and the investigation of their roles is a highly dynamic and “hot” research topic. The respective chapters address four main aspects, namely: microRNA investigation and annotation, the regulatory roles of microRNAs in various developmental processes, in response to abiotic factors, and in the context of biotic stress response regulation. Systematically reviewing the most important findings in this field, the book offers an essential guide for undergraduate and graduate students, teachers, and plant science researchers. Due to the potential applications of microRNAs in crop breeding and plant protection, it also represents a valuable resource for scientists in academia and the private sector alike.

Book excerpt: This reference work presents the origins of cells for tissue engineering and regeneration, including primary cells, tissue-specific stem cells, pluripotent stem cells and trans-differentiated or reprogrammed cells. There is particular emphasis on current understanding of tissue regeneration based on embryology and evolution studies, including mechanisms of amphibian regeneration. The book covers the use of autologous versus allogeneic cell sources, as well as various procedures used for cell isolation and cell pre-conditioning , such as cell sorting, biochemical and biophysical pre-conditioning, transfection and aggregation. It also presents cell modulation using growth factors, molecular factors, epigenetic approaches, changes in biophysical environment, cellular co-culture and other elements of the cellular microenvironment. The pathways of cell delivery are discussed with respect to specific clinical situations, including delivery of ex vivo manipulated cells via local and systemic routes, as well as activation and migration of endogenous reservoirs of reparative cells. The volume concludes with an in-depth discussion of the tracking of cells in vivo and their various regenerative activities inside the body, including differentiation, new tissue formation and actions on other cells by direct cell-to-cell communication and by secretion of biomolecules.

Available in PDF, EPUB and Kindle. Book excerpt: Our highly seasonal world restricts insect activity to brief portions of the year. This feature necessitates a sophisticated interpretation of seasonal changes and enactment of mechanisms for bringing development to a halt and then reinitiating it when the inimical season is past. The dormant state of diapause serves to bridge the unfavourable seasons, and its timing provides a powerful mechanism for synchronizing insect development. This book explores how seasonal signals are monitored and used by insects to enact specific molecular pathways that generate the diapause phenotype. The broad perspective offered here scales from the ecological to the molecular and thus provides a comprehensive view of this exciting and vibrant research field, offering insights on topics ranging from pest management, evolution, speciation, climate change and disease transmission, to human health, as well as analogies with other forms of invertebrate dormancy and mammalian hibernation.

Available in PDF, EPUB and Kindle. Book excerpt: Geneticists and molecular biologists have been interested in quantifying genes and their products for many years and for various reasons (Bishop, 1974). Early molecular methods were based on molecular hybridization, and were devised shortly after Marmur and Doty (1961) first showed that denaturation of the double helix could be reversed - that the process of molecular reassociation was exquisitely sequence dependent. Gillespie and Spiegelman (1965) developed a way of using the method to titrate the number of copies of a probe within a target sequence in which the target sequence was fixed to a membrane support prior to hybridization with the probe - typically a RNA. Thus, this was a precursor to many of the methods still in use, and indeed under development, today. Early examples of the application of these methods included the measurement of the copy numbers in gene families such as the ribosomal genes and the immunoglo bulin family. Amplification of genes in tumors and in response to drug treatment was discovered by this method. In the same period, methods were invented for estimating gene num bers based on the kinetics of the reassociation process - the so-called Cot analysis. This method, which exploits the dependence of the rate of reassociation on the concentration of the two strands, revealed the presence of repeated sequences in the DNA of higher eukaryotes (Britten and Kohne, 1968). An adaptation to RNA, Rot analysis (Melli and Bishop, 1969), was used to measure the abundance of RNAs in a mixed population.
