Smith Intracellular Perfusion of Excitable Cells Edited By P.G. Kostyuk and O.A.Krishtal With this Handbook Series, IBRO aims to fill a major gap in the world literature. The neuroscientist needs to be able to turn to which ever specialized method is most suited to the problem he is currently studying. Methods in the Neurosciences will help to provide expert advice on exactly how to carry out the experiments, on what difficulties can occur and on the limitations of the method. Intracellular Perfusion of Excitable Cells is the fifth book in the series. It describes in detail methods of replacing the internal medium of excitable cells with artificial solutions. The transmembrane ionic gradients and intracellular metabolic systems can then be precisely controlled in experiments investigating the electrical properties of the cellular membrane. The principles of the techniques were discovered over twenty years ago, and the first chapters discuss classic work on giant nerve and muscle fibres. However, the book compares and contrasts work on many different types of excitory cell, including isolated nerve cells and oocytes. New data are presented concerning the excitability of the somatic neural membrane. The final chapters analyse some common methodological problems, such as the isolation of cells from surrounding tissue and how to obtain reliable electrical measurements from perfused cell membranes. Enough experimental detail is given for the book to be of immediate practical use to research workers in all areas of neuroscience, including membrane biophysics, cell physiology, pharmacology, oncology and toxicology. Available in PDF, EPUB and Kindle. Book excerpt: A wealth of information has been accumulated about the function of ion channels of excitable cells since the extensive and pioneering voltage clamp studies by Hodgkin, Huxley, and Katz 36 years ago. The study of ion chan nels has now reached a stage at which a quantum jump in progress is antici pated. There are many good reasons for this. Patch clamp techniques origi nally developed by Neher and Sakmann 12 years ago have made it possible to study the function of ion channels in a variety of cells. Membrane ionic currents can now be recorded practically from many types of cells using the whole-cell patch clamp technique. The opening and closing of individual ion channels can be analyzed using the single-channel patch clamp method. Techniques have also been developed to incorporate purified ion channels into lipid bilayers to reconstitute an "excitable membrane. " Advanced tech niques developed in molecular biology, genetics, and immunology, such as gene cloning and the use of monoclonal antibodies, are now being applied to the study of ion channels. A variety of drugs have now been found or are suspected to interact with ion channels to exert therapeutic effects. In addition to the classical exam ples, as represented by local anesthetics, many other drugs, including cal cium antagonists, psychoactive drugs, cardiac drugs, and anticonvulsants, shown to alter ion channel function. For certain pesticides such as have been pyrethroids and DDT, sodium channels are clearly the major target site.

Available in PDF, EPUB and Kindle. Book excerpt: The Textbook of Ion Channels is a set of three volumes providing a wide-ranging reference source on ion channels for students, instructors and researchers. Ion channels are membrane proteins that control the electrical properties of neurons and cardiac cells; mediate the detection and response to sensory stimuli like light, sound, odor, and taste; and regulate the response to physical stimuli like temperature and pressure. In non-excitable tissues, ion channels are instrumental for the regulation of basic salt balance that is critical for homeostasis. Ion channels are located at the surface membrane of cells, giving them the unique ability to communicate with the environment, as well as the membrane of intracellular organelles, allowing them to regulate internal homeostasis. Ion channels are fundamentally important for human health and diseases, and are important targets for pharmaceuticals in mental illness, heart disease, anesthesia, pain and other clinical applications. The modern methods used in their study are powerful and diverse, ranging from single ion-channel measurement techniques to models of ion channel diseases in animals, and human clinical trials for ion channel drugs. Volume I, Part 1 covers fundamental topics such as the basic principles of ion permeation and selectivity, voltage-dependent, ligand-dependent, and mechano-dependent ion channel activation mechanisms, the mechanisms for ion channel desensitization and inactivation, and basic ion channel pharmacology and inhibition. Volume I, Part 2 offers a practical guide of cardinal methods for researching ion channels, including heterologous expression and voltage-clamp and patch-clamp electrophysiology; isolation of native currents using patch clamping; modeling ion channel gating, structures, and its dynamics; crystallography and cryo-electron microscopy; fluorescence and paramagnetic resonance spectroscopy methods; and genetics approaches in model organisms. All three volumes give the reader an introduction to fundamental concepts needed to understand the mechanism of ion channels; a guide to the technical aspects of ion channel research; a modern guide to the properties of major ion channel families; and includes coverage of key examples of regulatory, physiological and disease roles for ion channels.

Each chapter presents important discoveries in calcium entry pathways, specifically dealing with the molecular identification of store-operated calcium channels which were reviewed by earlier volumes in the Methods in Signal Transduction series. Crystallographic and pharmacological approaches to the study of calcium channels of epithelial cells are also discussed. Calcium ion is a messenger in most cell types. Whereas voltage gated calcium channels have been studied extensively, the non-voltage gated calcium entry channel genes have only been identified relatively recently. The book will fill this important niche.

Technological advance ment in biophysics, molecular biology, and immunology has been greatly accelerated, making it possible to conduct experiments that were deemed very difficult if not impossible in the past. For example, patch-clamp tech niques can now be used to measure ionic currents generated by almost any type of cell, thereby allowing us to analyze single-channel events. It is now possible to incorporate purified ion channel components into lipid bilayers to reconstitute an "excitable membrane." Gene cloning and monoclonal antibody techniques provide us with new approaches to the study of the molecular structure of ion channels. A variety of drugs have now been found or are suspected to interact with ion channels to exert therapeutic effects. In addition to the classical exam ples, as represented by local anesthetics, many other drugs, including cal cium antagonists, psychoactive drugs, cardiac drugs, and anticonvulsants, have been shown to alter the ion channel function. For certain pesticides such as pyrethroids and DDT, sodium channels are clearly the major target site. Many diseases of excitable tissues are known to be associated with, if not caused by, dysfunction of ion channels; these include cardiac ar rhythmias, angina pectoris, cystic fibrosis, myotonia, and epilepsies, to men tion only a few. Channel dysfunction can now be studied due to theoretical and technological developments in this area.

The key approach in the text is to model excitable tissue by representing the individual cells constituting the tissue. This is in stark contrast to the common approach where homogenization is used to develop models where the cells are not explicitly present. The approach allows for very detailed analysis of small collections of excitable cells, but computational challenges limit the applicability in the presence of large collections of cells.

Available in PDF, EPUB and Kindle. Book excerpt: A number of techniques to study ion channels have been developed since the electrical basis of excitability was first discovered. Ion channel biophysicists have at their disposal a rich and ever-growing array of instruments and reagents to explore the biophysical and structural basis of sodium channel behavior. Armed with these tools, researchers have made increasingly dramatic discoveries about sodium channels, culminating most recently in crystal structures of voltage-gated sodium channels from bacteria. These structures, along with those from other channels, give unprecedented insight into the structural basis of sodium channel function. This volume of the Handbook of Experimental Pharmacology will explore sodium channels from the perspectives of their biophysical behavior, their structure, the drugs and toxins with which they are known to interact, acquired and inherited diseases that affect sodium channels and the techniques with which their biophysical and structural properties are studied.
