Book excerpt: Metabolic Inhibitors: A Comprehensive Treatise, Volume I focuses on the properties of inhibitors of metabolic processes or enzyme systems. The book explores a wide range of substances that interfere with (and usually retard) metabolic and enzymic processes, emphasizing the inhibitor rather than the metabolic or enzymic system that is affected. Organized into 15 chapters, this volume begins with a historical overview of research on the mode of action of biological inhibitors, touching on the principle of competitive inhibition by structural analogues. The book then turns to the concept of a competitive enzymic relationship by growth inhibitory analogues and their corresponding amino acids; the metabolic inhibitory effects of polypeptides and proteins; and the use of pentoses as analogues in carbohydrate metabolism. The reader is also introduced to the metabolic inhibitory effects of fatty acids; the intermediary metabolism of phospholipids; and the inhibition of purine and pyrimidine analogues. The remaining chapters discuss the role of nucleic acids and nucleoproteins in the regulation of cellular metabolism and the inhibition of amino acid decarboxylases, cholesterol, and steroid biosynthesis. This book will be of value to biochemists, advanced students, medical research workers, and research workers in the various fields of biological chemistry, as well as those working in the microbiological, botanical, and other agricultural fields.

Book excerpt: Metabolic Inhibitors: A Comprehensive Treatise, Volume IV reviews developments in studies of inhibition of metabolic and enzymic processes ranging from photosynthesis and blood clotting to protein synthesis, fatty acid metabolism, and phospholipid metabolism. The book also explores the inhibition of specific enzyme reactions, such as amino acid activation, amino acid hydroxylation, and cyclic AMP formation. Organized into nine chapters, this volume begins with an overview of allosteric inhibition and inhibitors, and then discusses amino acid hydroxylase inhibitors. The reader is also introduced to inhibitors and activators of enzymes that regulate the cellular concentration of cyclic AMP. In particular, the book describes the role of lipids in the activation of adenyl cyclase by hormones; modification of adenyl cyclase in various physiological and pathological conditions; and synthesis of glycerophosphatides as well as phospho- and glycosphingolipids. This book is a valuable source of information for biochemists and medical research workers as well as virologists, microbiologists, plant physiologists, and agronomists.

Book excerpt: Metabolic Inhibitors: A Comprehensive Treatise, Volume II charts the major advances that have been made in understanding metabolic inhibition and inhibitors. The book explores the inhibition of enzymes, such as catechol amines, acetylcholinesterases, and succinic dehydrogenases, as well as inhibitors of processes ranging from gas transport to photosynthesis, nitrogen fixation, nitrification, and oxidative phosphorylation. Organized into 23 chapters, this volume begins with a discussion on the biosynthesis of nicotinamide adenine dinucleotide and dinucleotide analogues. The reader is then introduced to the biochemical significance and mode of action of antibiotics; substances interfering with the biogenesis and metabolism of catechol amines; and inhibition of enzyme activities by thioarsenites. Some chapters focus on mercaptide-forming agents, biological alkylating agents, organophosphates, and carbamates, while others examine the metabolic inhibitory effects of quinones, fungicides, and surface active agents. The book also considers the effects of anesthetics, depressants, and tranquilizers on cerebral metabolism, and then concludes with a chapter on inhibition caused by radiation. This book is a valuable resource for biochemists, advanced students, medical research workers, and research workers in the fields of biological chemistry, microbiology, botany, and agriculture.

The Workshop was attended by 250 scientists from 14 countries including Austria, Australia, Canada, Denmark, England, France, Germany, Holland, Israel, Italy, Japan, Sweden, and the United States of America. The Sixth International Workshop on Phosphate and Other Minerals will be held during June 24-26, 1983 in Verona, Italy. It will be hosted by Professor Giuseppe Maschio, Director, Division of Nephrology, Istituti Ospitalieri, Verona, Italy. The theme of this coming Workshop will continue to focus on the pathophysiology of phosphate homeostasis and the metabolism of other minerals.

This book was released on 2009-12-17 with total page 742 pages. Available in PDF, EPUB and Kindle. Book excerpt: Germination of the thought of "Enzymatic- and Transporter-Based Drug-Drug Interactions: Progress and Future Challenges" Proceedings came about as part of the annual meeting of The American Association of Pharmaceutical Scientists (AAPS) that was held in San Diego in November of 2007. The attendance of workshop by more than 250 pharmaceutical scientists reflected the increased interest in the area of drug-drug interactions (DDIs), the greater focus of PhRMA, academia, and regulatory agencies, and the rapid pace of growth in knowledge. One of the aims of the workshop was to address the progress made in quantitatively predicting enzyme- and transporter-based DDIs as well as highlighted areas where such predictions are poor or areas that remain challenging for the future. Because of the serious clinical implications, initiatives have arisen from the FDA (http://www.fda.gov/cber/gdlns/interactstud.htm) to highlight the importance of enzyme- and transporter-based DDIs. During the past ten to fifteen years, we have come to realize that transporters, in addition to enzymes, play a vital role in drug elimination. Such insight has been possible because of the continued growth in PK-ADME (pharmacokinetics-absorption-distribution-metabolism-excretion) knowledge, fueled by further advances in molecular biology, greater availability of human tissues, and the development of additional and sophisticated model systems and sensitive assay methods for studying drug metabolism and transport in vitro and in vivo. This has sparked an in-depth probing into mechanisms surrounding DDIs, resulting from ligand-induced changes in nuclear receptors, as well as alterations in transporter and enzyme expression and function. Despite such advances, the in vitro and in vivo study of drug interactions and the integration of various data sets remain challenging. Therefore, it has become apparent that a proceeding that serves to encapsulate current strategies, approaches, methods and applications is necessary. As Editors, we have assembled a number of opinion leaders and asked them to contribute chapters surrounding these issues. Many of these are the original Workshop speakers whereas others had been selected specially to contribute on topics related to basic and applied information that had not been covered in other reference texts on DDI. The resulting tome, entitled Enzyme- and Transporter-Based Drug Interactions: Progress and Future Challenges, comprises of four sections. Twenty-eight chapters covering various topics and perspectives related to the subject of metabolic and transporter-based drug-drug interactions are presented.

Book excerpt: Current Topics in Cellular Regulation, Volume 33: From Metabolite, to Metabolism, to Metabolon is a collection of articles on the study of the machinery of the living cell. The book is also a tribute to Paul Srere, an eminent biochemist, on the occasion of his sixty-fifth birthday. The book contains contributions written by students, postdoctoral fellows, and biochemists within Paul Srere's broad sphere of influence. The text provides articles that discuss topics on cellular infrastructure and metabolic organization; how carbohydrates cross the lipid membrane of bacterial cells; and macromolecular processes. The structural aspects of citrate biochemistry; the electron transport systems; nucleotide biosynthesis; enzyme organization and the direction of metabolic flow; and bioengineering applications are tackled as well. Biochemists, cytologists, and cell biologists will find the book very insightful.
