Book excerpt: The central theme of this dissertation is represented by the versatility of mesoporous silica nanomaterials in various applications such as catalysis and-applications, with main focus on biological applications of Mesoporous Silica Nanospheres (MSN). The metamorphosis that we impose to these materials from catalysis to sensing and to drug and gene delivery is detailed in this dissertation. First, we developed a synthetic method that can fine tune the amount of chemically accessible organic functional groups on the pores surface of MSN by exploiting electrostatic and size matching between the cationic alkylammonium head group of the CTAB surfactant and various anionic organoalkoxysilane precursors at the micelle-water interface in a base-catalyzed condensation reaction of silicate. Aiming nature imitation, we demonstrated the catalytic abilities of the MSNs. We utilized an ethylenediamine functional group for chelating Cu2 as a catalytic functional group anchored inside the mesopores. Thus, a polyalkynylene-based conducting polymer (molecular wire) was synthesized within the Cu-functionalized MSNs silica catalyst. For sensing applications, we have synthesized a poly(lactic acid) coated mesoporous silica nanosphere (PLA-MSN) material that serves as a fluorescence sensor system for detection of amino-containing neurotransmitters in neutral aqueous buffer. We exploited the mesoporosity of MSNs for encapsulating pharmaceutical drugs. We examined bio-friendly capping molecules such as polyamidoamine dendrimers of generations G2 to G4, to prevent the drug leaching. Next, the drug delivery system employed MSNs loaded with Doxorubicin, an anticancer drug. The results demonstrated that these nano-Trojan horses have ability to deliver Doxorubicin to cancer cells and induce their death. Finally, to demonstrate the potential of MSN as an universal cellular transmembrane nanovehicle, we anchored positively charged dendrimers on the surface of MSN and utilize them to complex cationic DNA. The p-EGFP-C1 gene-coated MSN nanocomposite was able to transfect cancer cell lines, such as human HeLa and CHO cancer cell lines. The gene carrier ability of MSNs was further proved by transfecting primary cells and cotransfecting of two different genes in cancer cell lines. In sum, MSN are versatile partners in several types of applications, as demonstrated in the 164 pages of my dissertation.

This book was released on 2004 with total page 286 pages. Available in PDF, EPUB and Kindle. Book excerpt: Structurally well-defined mesoporous silica materials with tunable pore size and narrow pore-size distribution have attracted much attention for their potential applications in sensing, drug delivery and gene transfer. A series of mesoporous silica nanosphere-based (MSN) nano-devices were synthesized and utilized for the aforementioned applications. For sensing application, we provide a first example in utilization of a multifunctionalized mesoporous silica material to selectively detect dopamine. A second application, rooted in this work, is the selective detection of amino-containing neurotransmitters under physiological condition. The drug delivery applications have been built around a controlled release drug delivery device. In the original conception, the system is using surface-derivatized cadmium sulfide (CdS) nanocrystals as chemically removable caps to encapsulate several drug molecules and neurotransmitters inside the organically functionalized MSN mesoporous framework. To study the cap-dependence release profile, adenosine 5-triphosphate (ATP) molecules were encapsulated in the MSNs followed by capping of the mesopores with chemically removable caps, such as CdS nanoparticles and poly(amido amine) dendrimers (PAMAM). A luciferase chemiluminescence imaging method was employed to investigate the kinetics and mechanism of the ATP release with various disulfide-reducing agents as uncapping triggers in real time. By varying the chemical nature of the "cap" and "trigger" molecules in our MSN system, we observed that the release profiles could be regulated. Further, the PAMAM dendrimers of low generations were utilized as MSN caps for drug delivery. The biocompatibility and delivery efficiency of the later MSN system in interaction with HeLa cervical cancer cells were demonstrated. The system renders the possibility to serve as a universal transmembrane carrier for intracellular drug delivery and imaging applications. This is the first uptake study of MCM-41 type mesoporous silicas into eukaryotic cells. The last application presented in the current dissertation is gene delivery. A MSN based gene transfection system was developed, where second generation (G2) PAMAMs were covalently attached to the surface of MSN. The G2-PAMAM-capped MSN material (G2-MSN) was able to complex with a plasmid DNA (pEGFP-C1). The gene transfection efficacy, uptake mechanism, and biocompatibility of the G2-MSN system with various cell types were investigated.

Book excerpt: Tiny Tech for Targeted Treatment: Mesoporous Silica and Gold Nanoparticles in Biomedicine Imagine microscopic delivery vehicles carrying potent drugs directly to diseased cells, minimizing harm to healthy tissue. This exciting field is explored in "Mesoporous Silica and Gold-Based Nanodevices: New Controlled Delivery Platforms for Biomedical Applications." This doctoral thesis delves into the design and application of innovative nanomaterials for targeted drug delivery, particularly in the fight against breast cancer. The thesis focuses on two promising nanomaterials: mesoporous silica and gold nanoparticles. Mesoporous silica boasts a sponge-like structure, ideal for encapsulating drugs and releasing them in a controlled manner. Gold nanoparticles, on the other hand, offer unique properties like biocompatibility and the ability to attach targeting molecules, guiding the nanodevice to specific cells. By combining these materials, researchers create "hybrid nanodevices" with remarkable abilities. One example involves a nanodevice loaded with two anti-cancer drugs and equipped with a "molecular gate" that unlocks only upon reaching cancer cells. This targeted approach promises to overcome drug resistance and reduce side effects. Another exciting application involves a pH-responsive nanodevice designed for gene editing. This tiny carrier delivers the CRISPR/Cas9 gene editing machinery directly to diseased cells, offering a potential future for gene therapy.
