Book excerpt: O gergelim (Sesamum indicum L.) é um vegetal que apresenta ampla adaptabilidade, sendo encontrada em regiões de clima tropical, subtropical e em zonas temperadas. O conhecimento e a quantificação da diversidade genética entre genótipos são de grande importância para o melhoramento do gergelim, tendo em vista a ausência de informações genéticas sobre a maioria do germoplasma disponível. Este trabalho teve por objetivo proceder a uma caracterização dos acessos de gergelim disponíveis da coleção de germoplasma da Embrapa Algodão, baseada em dados morfológicos e agronômicos. Para a realização desse trabalho, foi conduzido um ensaio com genótipos oriundos do BAG do gergelim, que foram semeados e avaliados na Estação Experimental de Veludo, em Itaporanga, PB, sob regime de irrigação. Os dados morfológicos e agronômicos foram obtidos de acordo com uma tabela de descritores. Para análise dos dados foram utilizados procedimentos multivariados hierárquico UPGMA e otimização de Tocher. Para obtenção da matriz de distância foi utilizado o complemento aritmético do coeficiente de Cole-Rodger. A representação gráfica simplificada das distâncias foi feita através de um dendrograma obtido pelo método UPGMA. Concluí-se que genótipos mais divergentes foram BRA 28151 e 24414 e os mais similares foram BRA 2879 E BRA 27855. Houve a variabilidade dos acessos avaliados para fins de obtenção de genótipos superiores com características de produtividade, número de frutos por planta, comprimento do fruto, cor das sementes, ciclo de maturação e número de ramificações laterais que podem ser usadas na seleção de genótipos superiores para o melhoramento genético da espécie. Dentre os acessos, os genótipos BRA 28771, BRA 28401 destacaram-se com alto potencial para produção de sementes.

Book excerpt: O gergelim (Sesamum indicum L.) apresenta centenas de variedades e linhagens com ampla variação de caracteres, dos quais a deiscência dos frutos recebe destaque atualmente. Marcadores moleculares têm sido utilizados de várias formas na análise genética e em programas de melhoramento das espécies. Este trabalho objetivou caracterizar a diversidade genética de acessos de gergelim por marcadores RAPD, agrupar os acessos avaliados de acordo com o grau de similaridade genética entre eles e identificar, pelo menos, uma marca molecular associada à característica de indeiscência dos frutos. Para tanto, foram utilizados 100 acessos da coleção de germoplasma de gergelim da Embrapa Algodão. A extração do DNA e a amplificação de fragmentos polimórficos por PCR, usando primers RAPD para análise da diversidade genética, seguiram metodologias do protocolo CTAB, com modificações. Cada produto de amplificação do PCR foi considerado um marcador RAPD, os quais foram visualizados com presença ou ausência de bandas em um gel de eletroforese. Os acessos foram agrupados pelo método hierárquico aglomerativo da média aritmética entre pares não ponderados (UPGMA) e a representação simplificada das distâncias genéticas foi feita através de um dendrograma. Todos os genótipos com frutos indeiscentes foram comparados a uma cultivar deiscente, para se identificar uma marca relacionada àquela característica. A partir de 47 primers, foi gerado um total de 130 bandas polimórficas, as quais foram utilizadas no cálculo das distâncias genéticas e na construção do dendrograma. Os genótipos de maior similaridade genética correspondeu aos pares 18-19 e 53-54, ambos com divergência aproximada de 0,08. A maior divergência detectada ocorreu entre os genótipos 18 e 69. Apesar do agrupamento da maioria dos acessos com características de indeiscência, a distribuição dos genótipos aparentemente não se mostrou diretamente associada a tal característica, fato provavelmente relacionado ao caráter aleatório do marcador utilizado. Entretanto, polimorfismo foi detectado entre genótipos indeiscentes e a testemunha utilizada, tornando-se possível relacionar uma marca que provavelmente esteja associada a tal característica. Os marcadores RAPD mostraram-se eficientes para detectar polimorfismo em gergelim, podendo ser utilizados como um poderoso instrumento na obtenção de informações úteis para o manejo de coleções de germoplasma e para o direcionamento de programas de melhoramento genético.
