PD models based on the manipulation of PD genes should prove valuable in elucidating important aspects of the disease, such as selective vulnerability of substantia nigra dopaminergic neurons to the degenerative process.

Book excerpt: Neurofibrillary tangles (NFTs) composed of intracellular aggregates of tau protein are a key neuropathological feature of Alzheimer’s Disease (AD) and other neurodegenerative diseases, collectively termed tauopathies. The abundance of NFTs has been reported to correlate positively with the severity of cognitive impairment in AD. However, accumulating evidences derived from studies of experimental models have identified that NFTs themselves may not be neurotoxic. Now, many of tau researchers are seeking a “toxic” form of tau protein. Moreover, it was suggested that a “toxic” tau was capable to seed aggregation of native tau protein and to propagate in a prion-like manner. However, the exact neurotoxic tau species remain unclear. Because mature tangles seem to be non-toxic component, “tau oligomers” as the candidate of “toxic” tau have been investigated for more than one decade. In this topic, we will discuss our consensus of “tau oligomers” because the term of “tau oligomers” [e.g. dimer (disulfide bond-dependent or independent), multimer (more than dimer), granular (definition by EM or AFM) and maybe small filamentous aggregates] has been used by each researchers definition. From a biochemical point of view, tau protein has several unique characteristics such as natively unfolded conformation, thermo-stability, acid-stability, and capability of post-translational modifications. Although tau protein research has been continued for a long time, we are still missing the mechanisms of NFT formation. It is unclear how the conversion is occurred from natively unfolded protein to abnormally mis-folded protein. It remains unknown how tau protein can be formed filaments [e.g. paired helical filament (PHF), straight filament and twisted filament] in cells albeit in vitro studies confirmed tau self-assembly by several inducing factors. Researchers are still debating whether tau oligomerization is primary event rather than tau phosphorylation in the tau pathogenesis. Inhibition of either tau phosphorylation or aggregation has been investigated for the prevention of tauopathies, however, it will make an irrelevant result if we don’t know an exact target of neurotoxicity. It is a time to have a consensus of definition, terminology and methodology for the identification of “tau oligomers”.

Book excerpt: "Progressive and stereotypical spread of aggregated, misfolded proteins is a common feature of many neurodegenerative diseases, including Parkinson's disease (PD). Animal models, where misfolded proteins are intracerebrally injected, robustly show a similar spread of pathology to anatomically connected regions. Neuroimaging studies have also shown that neurodegenerative diseases preferentially attack different, intrinsic brain networks. Computational models that conceptualize the spread of misfolded proteins as the underlying cause of those imaging patterns further complement the animal model observations. Yet, to date, there has been little attempt to combine these two approaches. The overarching objective of this project was to evaluate a computational "Epidemic Spreading Model" (ESM) using the [alpha]-synuclein ([alpha]syn) inoculation mouse model for PD, as it is well-suited for spatial and temporal control over the spread of misfolded proteins. We injected misfolded [alpha]syn pre-formed fibrils (PFFs) into the anterior olfactory nucleus (AON) of wild-type (WT) and M83 (hemi- and homozygous) transgenic (TG) mice. After 4 months, the PFF-inoculated mice all developed [alpha]syn pathology and an olfactory deficit. WT mice were further assessed with longitudinal, in vivo MRI, and showed region-specific volume changes compared to control-injected mice. From these same mice, we generated whole brain, 3D quantitative immunohistochemistry (qIHC) maps of [alpha]syn pathology burden that were used to perform ESM. Using connectivity measures defined from publicly-available tract tracer studies, and also defined from an advanced diffusion MRI protocol we acquired on WT and TG mice, the ESM successfully reproduced the pattern of [alpha]syn pathology in WT mice. In TG mice, the model was successful after applying a simple correction to account for pathology influenced by transgene overexpression. Across behavioral measures, qIHC analyses, and computational modeling, results of WT and TG mice were complementary. We have demonstrated that a computational model works well to describe pathology in a mouse model for PD, and their combined use could be valuable in evaluating potential therapies aimed at slowing or halting the aggregation and spread of misfolded [alpha]syn. " --
