Interferometry allows the manipulation of intensity distributions, and thus, force distributions on a trapped particle. To demonstrate the feasibility of such manipulation, 458 nm light, from an argon-ion laser, was injected into a Mach Zender interferometer. One mirror in the interferometer was oscillated with a piezoelectric phase modulator. The light from the interferometer was then injected into a microscope to trap a 9.75 micron polystyrene sphere. Book excerpt: Particles of nanometer- and micrometer-size, including diverse types of aerosols, dusts, and material powders, pose significant influence on environment, climate, human health, and material properties. Our understanding towards these particles and materials is mainly established from bulk samples and from particles deposited on substrates or dispersed in liquid solutions. However, fundamental properties and interactions of single particles cannot be fully resolved by characterizing bulk materials or deposited particles. Pristine physicochemical properties and rudimentary gas-particle interactions can only be investigated from single particles in their native states. The goal of this dissertation is to study single particles in gaseous environment with minimum external interferences. Diverse optical trapping (OT) methods for trapping and manipulating single airborne particles were explored and summarized. These configurations are categorized into radiation-pressure traps, photophoretic traps, and universal optical traps (UOT), according to particle’s absorbance. The UOT configurations are able to trap and manipulate a wide variety of single particles, including transparent and absorbing, spherical and irregular, inorganic and biological particles. Four key aspects of these OTs, simplicity, robustness, flexibility, and efficiency, are brought up to evaluate OT capabilities, providing a guidance to select the optimal optical configuration for specific applications. With single particles stably trapped, optical manipulations and characterizations can therefore be interrogated. Single-particle manipulations in air from one-dimension to three-dimension are reviewed; pulling and pushing single optically trapped particles along a Gaussian laser beam were explored and explained. Cavity ringdown spectroscopy is integrated with OTs to measure the light extinctions of trapped particles. Microscopic imaging and Raman systems are combined with OTs to examine the trapped particles. A number of application examples of this optical trapping-Raman spectroscopy system are demonstrated, including online characterizing carbon particles, effectively bleaching fluorescence in dye-doped polymer microspheres, acquiring clean Raman spectra of single biological particles, and monitoring chemical reactions in single biological particles. Optical trapping, associated with advanced laser spectroscopic techniques, offer a great flexibility for trapping, manipulating, characterizing, and monitoring single particles in air.

Book excerpt: A tightly focused laser beam can trap and manipulate microscopic objects by exerting force on them. Optical trapping or laser trapping or laser tweezers is a term widely used in describing the trapping and manipulation of these objects by optical gradient forces. Such devices have many uses in biophysics. This thesis describes the reconstruction of a previously existing optical trapping system that had fallen into disrepair and the work done to calibrate that optical trapping system. The pre-existing system was refurbished and aligned so that optical traps could again be generated. The resulting traps were then calibrated in preparation for further improvements to the trapping strength. Micrometer-sized dielectric spheres were trapped by the tweezers. Trap stiffness was measured using a mean square displacement method from data obtained with two particle tracking programs operating on captured microscope image stacks. The trap stiffnesses thus obtained were checked by applying known hydrodynamic forces to the particles and measuring the displacement. The results show that both the tracking programs (Rytrack and Sphere tracker)generated trap stiffness values closer to the results from hydrodynamic displacement. Trap stiffness was also found to increase with increasing laser power, as expected. The stiffness vs power can be fit by a power law with an exponent of 0.73 for Rytrack and 0.66 for Sphere tracker. The trap stiffness was also weakly dependent on height above the sample plane. Surprisingly, it was found that trap strength differed by some significant factors (sometimes as much as a factor of 2) along perpendicular axes. The source of this asymmetrical trap strength has yet to be determined. In addition, the apparent uncertainty in the trap strength data is much larger than the expected uncertainty from the mean square displacement. Mean square displacement seemed to vary with time. This left a source of systematic error seen in the values of trap stiffness. The source of this additional uncertainty is not clear but may lie in time variations in laser power. This will also be a subject of further work.

Book excerpt: We propose and demonstrate a novel approach to measure the size and refractive index of microparticles based on two beam optical trapping, where forward scattered light is detected to give information about the particle. The counter-propagating optical trap measurement (COTM) system exploits the capability of optical traps to measure pico-Newton forces for microparticles refractive index and size characterization. Different from the current best technique for microparticles refractive index measurement, refractometry, a bulk technique requiring changing the fluid composition of the sample, our optical trap technique works with any transparent fluid and enables single particle analysis without the use of biological markers. A ray-optics model is used to explore the physical operation of the COTM system, predict system performance and aid system design. Experiments demonstrate the accuracy of refractive index measurement of triangel n = 0.013 and size measurement of 3% of diameter with 2% standard deviation. Present performance is instrumentation limited, and a potential improvement by more than two orders of magnitude can be expected in the future. With further development in parallelism and miniaturization, the system offers advantages for cell manipulation and bioanalysis compatible with lab-on-a-chip systems.

This work is performed as a precursor to the development of a vacuum based optical tweezers interrogation tool for nanotechnology research. It was decided to first proceed with solving the problem of achieving optical levitation of a micro-scale particle in air before trying the same in a vacuum environment. This successful optical levitation in air confirms the work of Ashkin and Dziedzic. Levitation of 10 and 13.8 microns diameter polystyrene spheres was achieved, as well as the levitation of 10 and 100 microns diameter glass spheres. Particles were raised and lowered. A modicum of success was achieved translating particles horizontally. Trapping of multiple particles in one laser beam has been photographed. Also, it has been observed that particles, that may be conglomerates or irregular in shape, can also be trapped by a focused laser beam. Levitated glass beads were photographed using laser light scattered from the beads. The fact that there is evidence of optical traps in air containing irregular and conglomerate particles provides hope that future tool particles need not be perfect spheres.

Back focal plane interferometry has become a commonly precise method for position detection, but it is limited to single trap calibration. However, the study of complex biological systems requires simultaneous multiple trap manipulation. Time-sharing a trap at kHz by acousto-optic-deflectors produces multiple independent tweezers, but limited to two dimensions. In this work, we aim to combine the versatility of digital holography for simultaneous multiple manipulation with the precision of back focal plane interferometry for force measurement. A holographic optical system set-up has been built and used to generate two traps with orthogonal polarizations: a fixed trap useful for measuring forces, and a holographic tweezer which allows dynamic manipulation. This method represents a step towards simultaneous manipulation with many holographic traps and precise position detection with only one trap.
