Book excerpt: The aim of MHC Protocols is to document protocols that can be used for the analysis of genetic variation within the human major histocompatibility complex (MHC; HLA region). The human MHC encompasses approximately 4 million base pairs on the short arm of chromosome 6 at cytogenetic location 6p21. 3. The region is divided into three subregions. The telomeric class I region contains the genes that encode the HLA class I molecules HLA-A, -B, and -C. The centromeric class II region contains the genes encoding the HLA class II molecules HLA-DR, -DQ, and -DP. In between is the class III region, originally identified because it contains genes encoding components of the complement pathway. The entire human MHC has recently been sequenced (1) and each subregion is now known to contain many other genes, a number of which have immunological functions. The study of polymorphism within the MHC is well established, because the region contains the highly polymorphic HLA genes. HLA polymorphism has been used extensively in solid organ and bone marrow transplantation to match donors and recipients. As a result, large numbers of HLA alleles have been identified, a process that has been further driven by recent interest in HLA gene diversity in ethnic populations. The extreme genetic variation in HLA genes is believed to have been driven by the evolutionary response to infectious agents, but relatively few studies have analyzed associations between HLA genetic variation and infectious disease, which has been difficult to demonstrate.

Available in PDF, EPUB and Kindle. Book excerpt: Mitogen-activated protein kinase (MAPK) pathways are modules involved in the transduction of extracellular signals to intracellular targets in all eukaryotes. Distinct MAPK pathways are regulated by different extracellular stimuli and are implicated in a wide variety of biological processes. In plants, there is evidence for MAPKs playing a role in the signaling of abiotic stresses, pathogens, plant hormones, and cell cycle cues. The large number and divergence of plant MAPKs indicates that this ancient mechanism of bioinformatics is extensively used in plants and may provide new molecular hands on old questions.

Available in PDF, EPUB and Kindle. Book excerpt: Multidetector-row computed tomography (MDCT) has become a fundamental imaging technique for the study of many anatomical districts in different clinical situations, as it provides a fast, reliable, and accurate simultaneous evaluation of different organs, including parenchyma, hollow viscera, vessels, and bony structures. It has also equipped the radiologist with the ability to explore areas that in the last decade were largely ignored by CT, especially the coronary arteries and the colon, and to limit invasive diagnostic tests (e.g., catheter angiography) to those cases in which interventional procedures are required. The examination quality and the consequent diagnostic accuracy of MDCT are the results of an optimized study technique, which nonetheless needs to be adapted to the particular clinical situation of the patient, while bearing in mind the radiation exposure. Another fundamental parameter in the optimization of MDCT is the protocol for the intravenous injection of iodinated contrast material; in these studies, multiple variables, some modifiable by the operator and others patient-dependent, must be considered based on the aims of maximizing arterial and venous enhancements whilst minimizing the dose of iodine injected, saving money, and increasing patient safety. Through the presentation of cases addressing different anatomical regions and various clinical indications, including emergency and neuro-imaging, this volume seeks to provide the general radiologist and trainee specialist with a guide to the main study protocols to be implemented in order to optimize examination quality and, consequently, facilitate the diagnostic process.

Book excerpt: After the discovery of milk fat globule-epidermal growth factor-factor 8 (MFG-E8) about two decades ago, a new era of delineating its potential beneficial role in several inflammatory diseases has begun to spout from the bench to translational research. In MFG-E8 and Inflammation, the editor and contributors have gathered a remarkable collection covering novel discoveries on the rapidly growing field of MFG-E8 and Inflammation which includes not only the findings from their individual lobotomies, but also from a host of pioneering researchers of this field. MFG-E8 and Inflammation starts by describing the origin, structure, expression, functions and regulation of MFG-E8, and then continues thoughtfully exploring its potentiality as a marker for apoptotic, stressed and activated cells. The topics cover the cellular and physiological function of MFG-E8, especially its role in efficient phagocytosis of apoptotic cells, intestinal barrier function, blood cell homeostasis and coagulation, and in the maintenance of the intact vascular system. The role of MFG-E8 in macrophages, neutrophils, lymphocytes, dendritic cells, platelets, as well as non-hematopoietic cells is adequately described in the book. The chapters also contain several lucid discussions on the recent discoveries of the roles of MFG-E8 in the autoimmune diseases, sepsis, tissue ischemia-reperfusion, hemorrhage, inflammatory bowel diseases, acute lung injury, asthma, lung fibrosis, stroke, prion diseases and Alzheimer’s diseases with the potential focus on elucidating novel mechanistic pathways. MFG-E8 and Inflammation is an indispensable resource for scientists and clinical researchers working on fundamental or applied aspects of MFG-E8 pathobiology. This book explores, dissects and reviews several noteworthy findings and striking future perspectives which not only rewrite the disease pathophysiology, but also update our understanding towards attaining novel therapeutic potentials against various inflammatory diseases.

Available in PDF, EPUB and Kindle. Book excerpt: This book focusses on evolutionary, structural and functional aspects of pore-forming proteins, bringing together prominent researchers in the fields of structural biology and cellular and biophysical techniques. The focus is on the MACPF/CDC protein super family that was originally discovered because of unexpected structural similarity between a domain present in bacterial cholesterol-dependent cytolysins (CDC) and proteins of the membrane attack complex/perforin (MACPF) family. Members of the MACPF/CDC super family are crucial for many biological processes, being efficient agents of development, defence, attack and invasion of cells and tissues. However, their best-known role is in bacterial pathogenesis and the proper functioning of the vertebrate immune system, via formation of transmembrane pores in target cell membranes. The book contains chapters on the distribution of MACPF/CDC proteins and on aspects of their evolution and structural properties, the similarities between different super family members and functional properties of some of the best known examples. The book also contains an overview of biophysical approaches that may be used in the future to provide further insights into how these interesting proteins function.
