Book excerpt: The general process of lipid peroxidation consists of three stages: initiation, propagation, and termination. The initiation phase of lipid peroxidation includes hydrogen atom abstraction. Several species can abstract the first hydrogen atom and include the radicals: hydroxyl, alkoxyl, peroxyl, and possibly HO* 2. The membrane lipids, mainly phospholipids, containing polyunsaturated fatty acids are predominantly susceptible to peroxidation because abstraction from a methylene group of a hydrogen atom, which contains only one electron, leaves at the back an unpaired electron on the carbon. The initial reaction of *OH with polyunsaturated fatty acids produces a lipid radical (L*), which in turn reacts with molecular oxygen to form a lipid hydroperoxide (LOOH). Further, the LOOH formed can suffer reductive cleavage by reduced metals, such as Fe++, producing lipid alkoxyl radical (LO*). Peroxidation of lipids can disturb the assembly of the membrane, causing changes in fluidity and permeability, alterations of ion transport and inhibition of metabolic processes. In addition, LOOH can break down, frequently in the presence of reduced metals or ascorbate, to reactive aldehyde products, including malondialdehyde (MDA), 4-hydroxy-2-nonenal (HNE), 4-hydroxy-2-hexenal (4-HHE) and acrolein. Lipid peroxidation is one of the major outcomes of free radical-mediated injury to tissue mainly because it can greatly alter the physicochemical properties of membrane lipid bilayers, resulting in severe cellular dysfunction. In addition, a variety of lipid by-products are produced as a consequence of lipid peroxidation, some of which can exert beneficial biological effects under normal physiological conditions. Intensive research performed over the last decades have also revealed that by-products of lipid peroxidation are also involved in cellular signalling and transduction pathways under physiological conditions, and regulate a variety of cellular functions, including normal aging. In the present collection of articles, both aspects (adverse and benefitial) of lipid peroxidation are illustrated in different biological paradigms. We expect this eBook may encourage readers to expand the current knowledge on the complexity of physiological and pathophysiological roles of lipid peroxidation.

Book excerpt: The adduction of proteins and other biomolecules by electrophilic lipid peroxidation products such as 4-hydroxy-2-nonenal (HNE), 4-oxo-2-nonenal (ONE), malondialdehyde (MDA) or acrolein (ACR) is thought to be an initiating and/or propagating factor in the pathophysiology of several diseases such as atherosclerosis, diabetes, Alzheimer's, Parkinson's and other age-related disorders. The identification of protein sites modified by oxylipids is of key relevance for advancing our understanding how oxidative damage affects structure and function of proteins. Here, the use of MALDI tandem mass spectrometry with high energy collision-induced dissociation (CID) on a TOF/TOF instrument for sequencing oxylipid-peptide conjugates was systematically studied. Three synthesized model peptides containing one nucleophilic residue (i.e. Cys, His or Lys) were reacted with MDA, HNE, ONE and ACR. MALDI-MS analysis and MS/MS analysis were performed to confirm the adduct type and the modification sites. Michael adducts and Schiff bases were the predominant products under pH 7.4 within 2 hours. All MS/MS spectra of Michael adducts show the neutral loss of the oxylipid moiety ions. MS/MS spectra of Cys-containing peptide oxylipid conjugates exhibit additional characteristic neutral loss of HS-oxylipid moiety ions. MS/MS spectra of His-containing peptide oxylipid conjugates show characteristic oxylipid-containing His immonium ions. Spectra of Lys-containing peptide oxylipid conjugates (Schiff base) also show oxylipid-containing Lys immonium ions. However, there is no neutral loss of the oxylipid moiety ion for these Schiff bases. Determining the extent or relative amounts of the oxidative damage in cells could provide valuable insights into the molecular mechanisms of the diseases caused by oxidative stress. Relative quantitation of oxylipid-modified proteins in biological samples is a challenging problem because of the complexity and extreme dynamic range that characterize these samples. In this study, the reagents, N'-aminooxymethylcarbonylhydrazino-D-biotin (ARP) and iodoacetyl-PEO2- biotin (IPB), were used to enrich acrolein-modified Cys-containing peptides and the corresponding unmodified ones from subsarcolemmal mitochondria (SSM). The ratios between them were determined by nanoLC-SRM analysis. Model Cys-containing peptides labeled with ARP-acrolein and IPB were employed to demonstrate this method. Seven acrolein-modifed Cys-containing peptides from five mitochondrial proteins were quantified. The ratios for those seven peptides from the CCl4-treated rats are higher than the control ones indicating that the ratios of acrolein-modified peptides to unmodified ones are potential markers of oxidative stress in vivo. Age-dependent changes of protein carbonyls were investigated in subsarcolemmal mitochondria by using LC-SRM analysis of distinct ACR-modified Cys-containing peptides. Immunochemical analysis using an anti-ACR monoclonal antibody supported an increase of proteins modified by acrolein with age. However, total protein carbonyls measurement using ARP in Western blot analysis did not conform to this change suggesting that age-related changes in protein carbonyls are complex and would benefit from more specific measurement protocols.

Elucidating the processes leading to DNA damages, from the production of a simple radical entity to deleterious lesions, as well as the opportunities of repair by devoted enzymes, is a cornerstone towards the development of more efficient protection strategies. Sensitization and selective production of DNA lesions can also be exploited to induce the selective apoptosis of cancer cells upon exposition to radiation or to oxidative stress, for instance in the field of photodynamic therapy. The importance and relevance of the field is witnessed by the impressive amount of high-level papers dealing with this complex subject, and notably tackling the structural elucidation of DNA and DNA-drug adducts, the mechanisms of formation of DNA lesions (including the precise detection of the final lesion products), as well as the influence of the lesions on the DNA stability and dynamics and the consequences on the ease of repair. Book excerpt: Plasmodesmata (PD) are plant-specific intercellular nanopores defined by specialised domains of the plasma membrane (PM) and the endoplasmic reticulum (ER), both of which contain unique proteins, and probably different lipid compositions than the surrounding bulk membranes. The PD membranes form concentric tubules with a minimal outer diameter of only 50 nm, and the central ER strand constricted to ~10-15 nm, representing one of the narrowest stable membrane tubules in nature. This unique membrane architecture poses many biophysical, structural and functional questions. PM continuity across PD raises the question as to how a locally confined membrane site is established and maintained at PD. There is increasing evidence that the PM within PD may be enriched in membrane ‘rafts’ or TET web domains. Lipid rafts often function as signalling platforms, in line with the emerging view of PD as central players in plant defense responses. Lipid-lipid immiscibility could also provide a mechanism for membrane sub- compartmentalisation at PD. Intricate connections of the PM to the wall and the underlying cytoskeleton and ER may anchor the specialised domains locally. The ER within PD is even more strongly modified. Its extreme curvature suggests that it is stabilised by densely packed proteins, potentially members of the reticulon family that tubulate the cortical ER. The diameter of the constricted ER within PD is similar to membrane stalks in dynamin-mediated membrane fission during endocytosis and may need to be stabilised against spontaneous rupture. The function of this extreme membrane constriction, and the reasons why the ER is connected between plant cells remain unknown. Whilst the technically challenging search for the protein components of PD is ongoing, there has been significant recent progress in research on biological membranes that could benefit our understanding of PD function. With this Research Topic, we therefore aim to bring together researchers in the PD field and those in related areas, such as membrane biophysics, membrane composition and fluidity, protein-lipid interactions, lateral membrane heterogeneity, lipid rafts, membrane curvature, and membrane fusion/fission. We wish to address questions such as: - What mechanisms restrict lateral mobility of proteins and lipids along the PD membranes? - How can specific proteins be targeted to and turned over from membrane domains with restricted lateral access? - What elements (lipids, proteins, membrane curvature, packing order, thickness etc.) may contribute to the identity of PD membranes? - How do the structural and functional features of PD compare to other ER-PM contact sites? - How is the high curvature of the PD ER stabilised and what are possible functions of such a tightly constricted membrane tubule? - Do PD need to be prevented from spontaneous collapse and sealing? - What technologies are available to address these questions that can underpin PD research? While many of the processes are carried out intentionally, there are also certain unwanted changes that naturally occur in food and might have to be controlled. Therefore, efforts are made to devise processing technologies in which desirable attributes of foods are retained and their deleterious ef fects are minimized. While proteins, lipids and carbohydrates are the main nutrients of food that are affected by processing, it is their interaction with one another, as well as in volvement oflow-molecular-weight constituents that affects their flavor, color and overall acceptability. Thus, generation of aroma via thermal processing and bioconversion is of utmost importance in food preparation. Furthermore, processing operations must be opti mized in order to eliminate or reduce the content of antinutrients that are present in foods and retain their bioactive components. Therefore, while novel processing technologies such as freezing, irradiation, microwaving, high pressure treatment and fermentation might be employed, control process conditions in a manner that both the desirable sensory attributes and wholesomeness of foods are safeguarded is essential. Obviously, method ologies should also be established to quantitate the changes that occur in foods as a result of processing. This volume was developed from contributions provided by a group of internation ally-recognized lead scientists.
