In the wake of materialism and social change, man had been described in the early years of our century as the “dead end” of nature. Freud had redefined culture as illness—a result of drive sublimation; Klages had called the spirit (and he was surely speaking of the hypertrophied intellect) the “adversary of the soul,” propounding a return to a life like that of the Pelasgi, the aboriginal inhabitants of Greece; and Spengler had declared the “Demise of the West” during the years following World War I. The consequences of such pessimism continued to proliferate long after its foundations had been superseded. It was with these foundations—the natural sciences—that Gebser began. As early as Planck it was known that matter was not at all what materialists had believed it to be, and since 1943 Gebser has repeatedly emphasized that the so-called crisis of Western culture was in fact an essential restructuration.… Gebser has noted two results that are of particular significance: first, the abandonment of materialistic determinism, of a one-sided mechanistic-causal mode of thought; and second, a manifest “urgency of attempts to discover a universal way of observing things, and to overcome the inner division of contemporary man who, as a result of his one-sided rational orientation, thinks only in dualisms.” Against this background of recent discoveries and conclusions in the natural sciences Gebser discerned the outlines of a potential human universality. He also sensed the necessity to go beyond the confines of this first treatise so as to include the humanities (such as political economics and sociology) as well as the arts in a discussion along similar lines. Paul Philibert, OP, has taught pastoral theology in the United States and abroad. He is a Dominican friar of the Southern Province. His translation of a collection of Congar's essays on the liturgy has recently been published by Liturgical Press under the title At the Heart of Christian Worship. His book The Priesthood of the Faithful: Key to a living Church (Liturgical Press, 2005) reflects the ecclesiology of Yves Congar and his Vision of the apostolic life of the faithful."

It compares our life experience with the attempts to grasp it by astrologers, eugenicists, psychologists, neuroscientists, social scientists, and philosophers. The main opposition among these specialties lies between understanding and misunderstanding. The book also addresses the central methodological difficulty of capturing a human life. It is first examined how certain approaches may lead to a misunderstanding of human life. The book contrasts the example of astrology—an accepted practice in ancient civilizations, but now classified among the pseudosciences—with astronomy, a full-fledged science since Galileo’s time. Another, more recent approach regards human life as predetermined by genes: the methods used by eugenicists, and later by political regimes under the name of hereditarianism, came to compete with genetics. A broader analysis shows how astrology and eugenicism are not truly scientific approaches. Next, the book looks at the ways of capturing an imaginary or real human life story. A comprehensive approach will try to fully understand their complexity, while a more explanatory approach considers only certain specific phenomena of human life. For example, demography studies only births, deaths, and migration. Another crucial factor in the collection of life histories is memory and its transmission. Psychology and psychoanalysis have developed different schools to try to explain them. The book concludes with a detailed discussion of the concepts and tools that have been proposed in more recent times for understanding the various aspects of life stories: mechanisms, systems, hermeneutics, and autonomy.

Book excerpt: Ethnographers have observed Muslims nearly everywhere Islam is practiced. This study analyzes four seminal texts that have been read widely outside anthropology. Two are by distinguished anthropologists on either side of the Atlantic, Islam Observed (by Clifford Geertz in 1968) and Muslim Society (by Ernest Gellner in 1981). Two other texts are by Muslim scholars, Beyond the Veil (Fatima Mernissi in 1975) and Discovering Islam (by Akbar Ahmed in 1988). Varisco argues that each of these four authors approaches Islam as an essentialized organic unity rather than letting 'Islams' found in the field speak to the diversity of practice. The textual truths engendered, and far too often engineered, in these idealized representations of Islam have found their way unscrutinized into an endless stream of scholarly works and textbooks. Varisco's analysis goes beyond the rhetoric over what Islam is to the information from ethnographic research about what Muslims say they do and actually are observed to do. The issues covered include Islam as a cultural phenomenon, representation of 'the other', Muslim gender roles, politics of ethnographic authority, and Orientalist discourse.

B Introduction The expression of prokaryotic and eukaryotic genes has been shown most often to be regulated at the level of mRNA synthesis. Thanks to the rapid development of methods for dissecting DNA sequences, cis-acting regulatory elements such as promoters and enhancers have been recognised. More recently, the widely expressed intuition that discrete sequences within these elements constitute binding sites for sequence-specific binding proteins has been confirmed, especially through the use of "footprinting" assays (for examples, Galas and Schmitz, 1978). This and similar assays have already resulted in the recognition, isolation and analysis of DNA-bind ing proteins for several genes. Excellent reviews exist of the structural studies on these transcription regulatory proteins and related DNA elements (for example, Glover, 1989 and Johnson and McKnight, 1989), to which the reader is referred for detailed information. To set the scene for applications of the techniques described in this volume, only the barest outline of previous studies is presented here. Protein-DNA interactions are dependent on very specific tertiary configurations of the binding protein which allow the closest contact with the DNA helix.
