Book excerpt: The first edition of this manual appeared in 1992 and was entitled ECAT Assay Procedures. It was the result of a unique cooperation between experts brought together by the European Concerted Action on Thrombosis and Disabilities (ECAT). The Concerted Action was at that time under the auspices of the Commission of the European Union. The second edition, like the first edition, deals with diagnostic tests within the field of thrombosis. However, the second edition has a broader scope because it is no longer limited by the frontiers of ECAT. Experts allover the world, in and outside ECAT, have contributed to this edition. The editors are very grateful for their contributions. The need for a new edition is obvious. Since 1992 new assays have been introduced for research, diagnosis, and therapy of thrombosis; for other assays improvements have been suggested, while a few others became redundant. The editors waived the radioimmunoassays of ~-thrombog1obulin and platelet factor 4 due to the fact that the kits required for these assays are rarely, or no longer, available. Also the PAI-1 activity assay was waived as it is liable to many inconsistencies and to large variations. A list of names and addresses of manufacturers marketing the kits and reagents has been compiled, together with a list of the recommended nomenclature of quantities in thrombosis and haemostasis, in order to facilitate the use of the updated version. These lists have been carefully compiled by Johannes J. Sidelmann, PhD, Department of Clinical Biochemistry in Esbjerg, Denmark.

Book excerpt: This book offers a description of current and recently developed laboratory assays in the field of haemostasis and thrombosis. It is the result of a unique cooperation between experts from more than 60 institutes in 12 European countries, brought together by the ECA T (European Concerted Action on Thrombosis and Disabilities) under the auspices of the Commission of the European Communities in Brussels, Belgium. The ECAT, which was initiated in 1981, designed and performed three prospective clinical studies to establish haemostatic factors as risk indicators of thrombosis. Included were patients with angina pectoris at risk from myocardial infarction, patients undergoing angioplasty at risk from re-stenosis, and patients receiving hip replacement at risk from deep venous thrombosis. Assay procedures were chosen, training courses for technicians held, and essential reagents were supplied from a central source. A quality control assessment scheme served to compare assay results both within and between laboratories. In the angina pectoris study, centres determined most of the assays locally; in the other two studies assays were performed centrally. The need for further quality assessment in Europe Dr J. F. Davidson in Glasgow, led to a separate activity coordinated by including coagulation inhibitors and plasminogen as risk factors for familial venous thrombosis. The Editors hope the ECA T Assay Procedures book will contribute to further harmonization ofhaemostasis assays, and ultimately to their standardization.

Book excerpt: Coagulation testing is the basis for the diagnosis of bleeding and thrombotic disorders, as well as the mainstay of anticoagulant monitoring and management. This handbook provides practical information and guidance on topics relevant to directing a coagulation laboratory, filling a void in the literature. Since the first edition, all chapters have been updated and an entirely new chapter is included on pharmacogenomics and pharmacogenetics. The book will aid pathologists, clinical laboratory scientists and other physicians serving as laboratory directors to understand and carry out their responsibilities. It will also assist residents and fellows in learning the basics of coagulation testing and serve as a useful day-to-day reference for coagulation laboratory supervisors, technologists, and technicians. Finally, clinicians may find aspects of the book helpful in understanding the role of the coagulation laboratory in patient evaluation and monitoring.

Available in PDF, EPUB and Kindle. Book excerpt: A practical guide to laboratory diagnosis and treatment of hemostatic disorders. This concise book distils the most clinically up-to-date information on thrombotic and bleeding disorders, including the latest treatment strategies, for key conditions and diseases. Essential Guide to Blood Coagulation covers both the stable and the acute stages of hereditary and acquired bleeding and thrombotic disorders. Faced with a bleeding patient, it may be difficult to determine whether blood loss is due to a local factor, or an underlying hemostatic defect. There are a range of laboratory tests which can be performed to identify the cause of bleeding in a patient. This book highlights the tests that can be used in the laboratory to aid diagnosis. Essential Guide to Blood Coagulation has been updated to include the new anticoagulants and now has a dedicated chapter on antiplatelet drugs. This invaluable guide will help all those treating patients to expand their knowledge of hemostatic disorders. This book presents reviews of the underlying theory, physiology, and biochemistry of hemostasis and thrombosis, including the enzymology of blood coagulation and fibrinolysis. This compilation is divided into three levels of specific purposes. First is to provide the most reliable and widely accepted laboratory assays of undisputed diagnostic clinical value, which provides newcomers in the field and experienced workers in the coagulation laboratory with a reference manual to everyday work in a clinically-oriented environment. Second is to review and sketch in outline the theoretical sections focusing on mechanisms. Finally, this text aims to include a systematic review of the most successful purification techniques for individual coagulation factors and moieties of the fibrinolytic enzyme system. This publication is beneficial to medical students and clinicians concerned with human blood coagulation.

Book excerpt: This second edition volume expands on the previous edition with updates about the latest state-of-the-art techniques used in leading hemostasis and thrombosis laboratories for diagnosis and exclusion of hemorrhagic and thrombotic diseases. The chapters in this book are organized into seven parts. Part One provides a general overview on hemostasis and thrombosis, preanalytical issues in testing, and routine hemostasis assays. Part Two covers laboratory testing for thrombophilia, including reviews for activated protein C resistance, protein C, lupus anticoagulant testing, and antiphospholipid antibodies. Part Three addresses monitoring continuous anticoagulant infusions and measuring the effects of oral anti-thrombotic therapy. Part Four talks about heparin induced thrombocytopenia and vaccine induced immune thrombotic thrombocytopenia. Part Five and Six cover ADAMTS13 activity testing and new information on bleeding disorders such as chromogenic factor VIII assays, measurement of emicizumab, and treatment of hemophilia A and B. Finally, Part Seven discusses global assays, research applications, and postanalytical considerations. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Cutting-edge and comprehensive, Hemostasis and Thrombosis: Methods and Protocols, Second Edition is a valuable resource for scientists and researchers struggling to identify the appropriate methods for hemostasis and thrombosis testing, or who seek additional expert guidance on such testing.

Eric Preston, Emeritus Professsor of Haematology, University of Sheffield and Director, WHO and WFH International External Quality Assessment Programs for Blood Coagulations, Sheffield, UK The hemostasis laboratory has a vital role in the diagnosis and management of patients with familial and acquired hemorrhagic and thrombotic disorders. Rapid changes in the number and complexity of tests in this discipline have presented challenges for laboratories, as they develop quality programs for the oversight of this testing. Quality in Laboratory Hemostasis and Thrombosis has been completely revised and updated to reflect the changing process of managing quality. The second edition provides information on all aspects of testing, from pre-analytic to analytic and result reporting, in addition to external quality assurance. Chapters throughout the book include the development of global guidelines for methods, as well as the preparation of international standard plasmas and reagents. Designed to capture the elements of quality at all levels of the practice of laboratory hemostasis and thrombosis, this book will guide the reader through the development of a quality program to support all activities in the hemostasis laboratory, both simple and complex.
