One subgroup, the mitogen-activated protein kinase (MAPK) family, are Ser/Thr protein kinases responsible for regulating cellular processes such as division, differentiation, survival, and death. The c-Jun N-terminal kinase (JNK) MAPK pathway is primarily responsive to stress signals like UV radiation and inflammatory cytokines and its activation can be pro-survival or pro-apoptotic depending on cell type, stimulus, duration of activation, and/or crosstalk with other signaling pathways. Its deregulation has been implicated in diseases like cancer, obesity, Parkinson's, muscular dystrophy, rheumatoid arthritis and aging. How do MAPKs recognize their substrates and regulators? A recognition mechanism, characterized by specific interaction at a so-called docking site (D-site), exists for JNK interacting proteins. The linear JNK docking sites share a core consensus sequence and due to the conservation of the D-site sequence and its importance in mediating interactions with the MAPKs, a protein database was searched for novel JNK D-sites using a hybrid algorithm called D-finder. Following in silico validation, in vitro validation by peptide macroarray, and biochemical workup, functional D-sites were identified in multiple proteins. GLI3 and its human hedgehog transcription factor paralog, GLI1, were identified as JNK substrates where the D-site mediated phosphoacceptor sites were also characterized. These findings are the first to identify a link between the Hedgehog and MAPK signaling pathways which has implications in various types of cancer. The functionally uncharacterized protein Smoothelin Like 2, SMTNL2, also had a D-finder predicted JNK D-site that was biochemically validated. Mass Spectrometry and subsequent biochemical validation were able to identify a subset of these residues that underwent D-site mediated phosphorylation. Furthermore, in a cell based assay, these sites also underwent D-site mediated phosphorylation. Based on high throughput datasets and RT-PCR analysis SMTNL2 is expressed in adult skeletal muscle as well as a mouse skeletal muscle cell line. The potential JNK interaction with SMTNL2 in skeletal muscle has implications for understanding the mechanism of myogenesis as well as muscle associated diseases such as muscular dystrophy and insulin resistance associated type 2 diabetes.
