Available in PDF, EPUB and Kindle. Book excerpt: Since their development a decade ago, human induced pluripotent stem cells (iPSC) have revolutionized the study of human disease, given rise to regenerative medicine technologies, and provided exceptional opportunities for pharmacologic research. These cells provide an essentially unlimited supply of cell types that are difficult to obtain from patients, such as neurons or cardiomyocytes, or are difficult to maintain in primary cell culture. iPSC can be obtained from patients afflicted with a particular disease but, in combination with recently developed gene editing techniques, can also be modified to generate disease models. Moreover, the new techniques of 3 Dimensional printing and materials science facilitate the generation of organoids that can mirror organs under disease conditions. These properties make iPSC powerful tools to study how diseases develop and how they may be treated. In addition, iPSC can also be used to treat conditions in which the target cell population has been lost and such regenerative approaches hold great promise for currently untreatable diseases, including cardiac failure or photoreceptor degenerations.

State-of-the-art updates have highlighted the feasibility of hPSCs for the large-scale preparation of diverse kinds of stem cells and functional cells, such as mesenchymal stem/stromal cells (MSCs), hematopoietic stem cells (HSCs), neural stem cells (NSCs), natural killer (NK) cells, and chimeric antigen receptor-transduced T cells (CAR-Ts). With the aid of preclinical investigations and clinical practice, hPSCs have been recognized as promising therapeutic cell sources with excellent properties for treating a variety of refractory and recurrent diseases. This book provides a comprehensive overview of advances in pluripotent stem cells.

From chronic diseases to complex diagnostic procedures, the industry stands at the crossroads of technological innovation and a burgeoning demand for more efficient, precise interventions. Patient expectations are soaring, and the deluge of medical data is overwhelming traditional healthcare systems. It is within this challenging environment that AI-Driven Innovations in Digital Healthcare: Emerging Trends, Challenges, and Applications emerges as a beacon of insight and practical solutions. The traditional healthcare framework is struggling to keep pace with the diverse demands of patients and the ever-expanding volume of medical data. As diseases become more intricate, attempts to provide timely identification and precise treatment of ailments become increasingly elusive. The urgency for a paradigm shift in healthcare delivery is emphasized by the critical need for early interventions, particularly in disease prediction. This challenge necessitates a holistic approach that harnesses the power of artificial intelligence (AI) and innovative technologies to steer healthcare toward a more responsive and patient-centric future.

Available in PDF, EPUB and Kindle. Book excerpt: Phenotyping of Human iPSC-derived Neurons: Patient-Driven Research examines the steps in a preclinical pipeline that utilizes iPSC-derived neuronal technology to better understand neurological disorders and identify novel therapeutics, also providing considerations and best practices. By presenting example projects that identify phenotypes and mechanisms relevant to autism spectrum disorder and epilepsy, this book allows readers to understand what considerations are important to assess at the start of project design. Sections address reproducibility issues and advances in technology at each stage of the pipeline and provide suggestions for improvement. From patient sample collection and proper controls to neuronal differentiation, phenotyping, screening, and considerations for moving to the clinic, these detailed descriptions of each stage of the pipeline will help everyone, regardless of stage in the pipeline. In recent years, drug discovery in the neurosciences has struggled to identify novel therapeutics for patients with varying indications, including epilepsy, chronic pain, and psychosis. Current treatment options for such patients are decades old and offer little relief with many side effects. One explanation for this lull in novel therapeutics is a lack of novel target identification for neurological disorders (and target identification requires exemplar preclinical data). To improve on the preclinical work that often relies on rodent modeling, the field has begun utilizing patient-derived induced pluripotent stem cells (iPSCs) to differentiate neurons in vitro for preclinical characterization of neurological disease and target identification. Discusses techniques and new technology for iPSC culturing and neuronal differentiation to establish best practices in the lab Outlines considerations for phenotypic assay development Provides information about the successes, failures, and implications of phenotyping and screening with iPSC-derived neurons Describes how human iPSC-derived neurons are being used for preclinical discovery research as well as the development of therapeutics utilizing hiPSC-derived neurons

Book excerpt: The aim of this collection of articles is to assemble advances in stem cell-based approaches and their application to the study of parasitic diseases. Tropical parasites, including unicellular protozoan organisms and helminths, represent a major public health burden, particularly in tropical regions of the world. The study of these organisms is significantly hampered by the lack of effective in vitro/ ex vivo culture systems that mimic natural conditions and facilitate a thorough understanding of parasite development and host-parasite interactions. The advent of stem cell technology offers the opportunity to derive the right cell types to culture these parasites. Moreover, stem cell-derived organoids accurately reproduce the particular niche in which the parasites grow, develop, interact with host tissues and reproduce. In addition, particularly for helminths (i.e., multicellular parasites), the identification and characterisation of the parasite’s stem cell system, will be critical to complement our current and future understanding of fundamental biological processes, such as worm maturation, and interaction with the host immune system and microbiota.
