Il est synthétisé dans le rumen par isomérisation de l'acide linoléique (C18:2), et dans la mamelle par désaturation de l'acide trans-vaccénique (C18:1t11), provenant aussi de la biohydrogénation ruminale des acides gras polyinsaturés. De nombreux facteurs peuvent faire varier sa teneur dans le lait de vache, mais seule l'alimentation est de manipulation facile et efficace. Plusieurs expériences ont été réalisées afin de mieux maîtriser l'influence de trois grands facteurs de variation de la production ruminale de CLA ou de C18:1t11 liés à l'alimentation : le pH ruminal, la quantité d'acides gras insaturés, et la forme de présentation de la matière grasse. Un pH ruminal bas augmente la production ruminale de CLA, mais diminue sa production mammaire. Un pH proche de la neutralité permet une accumulation ruminale de C18:1t11 puis de CLA. L'apport d'une quantité importante de C18:2, et plus particulièrement de graines de soja extrudées, renforce cette accumulation. CLA et C18:1t11 ainsi produits dans le rumen vont augmenter les teneurs en CLA dans le lait.

Book excerpt: L'ajout de certains acides linoléiques conjugués (ALC) dans l'alimentation diminue la synthèse du gras du lait chez la vache. Les ALC alimentaires peuvent cependant être hydrogénés par les microorganismes du rumen. La formation de sels de calcium est considérée comme une méthode pour prévenir l'hydrogénation des ALC au cours de leur passage dans le rumen. Afin de déterminer les effets des ALC sous forme inerte sur la production et la composition du lait et sur le profil métabolique sanguin en conditions commerciales, 240 vaches laitières provenant de 8 troupeaux québécois ont été réparties selon un dispositif en blocs, selon la date de vêlage. Les traitements consistaient en quatre différentes doses d'ALC (0, 8, 16 et 32 g/j) distribuées de façon aléatoire à l'intérieur de chaque bloc. La production de gras a été diminuée de 11, 20 et 28 %, et la teneur en gras diminuée de 13, 22 et 28 % par rapport au témoin lorsque les animaux ont reçu 8, 16 et 32 g/j d'ALC (effet linéaire : P 0,001). La production laitière, la production et la teneur en protéine du lait et le profil métabolique des animaux n'ont pas été affectés. Les sels de calcium d'ALC pourraient être utilisés en conditions commerciales pour contrôler efficacement la teneur en gras du lait. Il a été démontré que l'isomère d'ALC 18:2 trans-10, cis-12 réduit l'expression de plusieurs gènes codant pour des enzymes lipogéniques et inhibe la désaturation des acides gras dans le tissu mammaire. De récentes études suggèrent aussi que le 18:2 trans-10, cis-12 stimule l'activité d'enzymes proinflammatoires, qui elles entraînent une diminution de l'adipogenèse et une résistance du tissu adipeux à l'insuline. L'obectif du deuxième volet de ce programme de recherche consistait à vérifier si les effets du 18:2 trans-10, cis-12, qui sont rapportés dans la littérature pour le tissu adipeux, peuvent être partiellement responsables de l'inhibition de la synthèse de la matière grasse laitière chez la vache en lactation. Quatre vaches Holstein ont été réparties selon un dispositif en chassé-croisé. Les animaux recevaient une perfusion intraveineuse de 18:2 ds-9, cis-12 (témoin) ou de 18:2 trans-10, cis-12, à raison de 10 g/j, sur une période de 5 jours. Au dernier jour de perfusion, des biopsies du tissu mammaire étaient prélevées et soumises à une analyse de l'abondance de l'ARNm codant pour différentes enzymes associées au métabolisme lipidique des tissus mammaires et adipeux. Suite à la perfusion intraveineuse de 18:2 trans-10, cis-12, la teneur et la production de gras du lait ont diminué respectivement de 46 et 38 % (P 0,05). Aucun effet de traitement n'a été observé sur la consommation volontaire de matière sèche, la production laitière ou la teneur en protéines du lait La perfusion intraveineuse de 18:2 trans-10, cis-12 a respectivement diminué l'ARNm de l'acétyl-coenzyme A carboxylase et de l'acide gras synthase de 46 et 57 % (P
