These inefficient multiplication methods have long been a serious problem in rose breeding program in Pakistan. Plant Tissue Culture offers an alternative method of multiplication. Significant features of in vitro propagation are its enormous multiplication capacity in relatively shorter span of time, production of disease free and premium quality plating material and ability to regenerate planting material round the year. Hence, optimization of culture conditions for micropropagation of Roses for commercial exploitation to get great number of disease free plants in shorter period of time with high uniformity is inevitable.

Shoot tips with axillary buds were used as explant. The explants were surface sterilized with 70% alcohol for 4 minutes and 5% bleach for 4 minutes followed by three washing with autoclaved double distilled water After that the explants were cultured on MS media (Murashige and skoog,1962) supplemented with different concentration of BAP (Benzyladenine) and NAA (Naphthalene acetic acid). Effect of different concentrations of BAP (1.5 mg/l, 3 mg/l) alone and in combination with NAA (0.5 mg/l) for shoot induction. BAP and NAA (3.0 mg/l +1.5 mg/l respectively) gives the good results for shooting. the effect of NAA (1.5 mg/l, 3 mg/l) alone and in combination with BAP (0.5 mg/l) for root induction was examined. NAA and BAP (3.0 mg/l +1.5 mg/l respectively) gives the good results for rooting. After rooting plants were acclimatized and successfully transferred to the greenhouse. To control the oxidative browning in Rosa centifolia different treatments (charcoal, charcoal + running water, stirring in antioxidant solution of citric acid and ascorbic acid) were used. But the charcoal with the running water gives the good results.

Book excerpt: "Protocols for the successful micropropagation of 'Queen Elizabeth' ('Q.E.') and 'Dick Koster' ('D.K.') roses were established, yielding a seven-ten fold multiplication rate per month. The effects of mechanically induced stress (MIS) (shaking stress) were evaluated on early establishment of greenhouse-grown 'Q.E.' and 'D.K.' rose cuttings and the ex vitro survival and hardiness of micropropagated 'Q.E.' plantlets. Shaking 'Q.E.' rose cuttings at 200 rpm for 30 min daily for 4 weeks during the rooting stage increased root length, dry weight and the root:shoot dry weight ratio. Similar shaking of 'D.K.' rose at 200 rpm for 15 min increased shoot fresh and dry weight and root length and dry weight. Prior to ex vitro acclimatization, plantlets shaken at 150 rpm for 15 min had reduced leaf dry weights. Those shaken at 200 rpm for 15 min had lower specific root water content but greater percent root dry matter. MIS was not directly implicated in improving ex vitro survival and hardiness of 'Q.E.' rose. (Abstract shortened by UMI.)" --
