Book excerpt: While seed priming (controlled hydration followed by drying) enhances seed germination rates, the longevity of primed seeds in storage is often reduced. Post-priming treatments involving a slow drying process to cause a partial reduction in seed moisture content prior to complete drying can restore longevity in several species compared to rapid drying after priming. The goal of this project is to develop solid matrix priming and post-priming protocols that improve seed germination performance without penalizing potential seed quality or longevity after storage. A solid matrix-based priming protocol (10 g of seeds incubated for 2 days with 12 mL of water and 20 g of AGSORBR® clay matrix followed by rapid drying) was developed for two tomato seed lots that advanced germination timing, although it accelerated loss of seed quality during storage. Similar experiments with pepper and onion seeds encountered some initial complications, including low seed viability and susceptibility to fungal growth, so experiments were continued only with tomato seeds. For both tomato seed lots (T1 and T2), two days of priming advanced germination timing but had the maximum adverse effect on subsequent storage life. After the priming but before drying, post-priming treatments were conducted on the two tomato seed lots by adding additional amounts of clay matrix (2, 4, or 6 g) into the priming treatment to reduce seed moisture content and incubating for two additional days before drying to 8% seed moisture content (dry weight basis). Controlled deterioration tests (47% RH and 50°C) were conducted on control seeds and seeds exposed to different priming and post-priming treatments and seed longevities were compared. In this study, ASTEQ Q2 Oxygen Sensing Technology was used to measure the oxygen consumption of individual seeds at frequent intervals during the priming and aging process in lieu of manual radicle emergence observations. The results for determination of priming conditions showed a high correlation between respiration rates and radicle emergence rates in the same conditions. Potential maximum longevity (Pmax) values were estimated from the aging-time model using respiration time course data. Generally, the results of post-priming treatments did not accord with our expectations, for the post-priming treatments did not extend tomato seed longevity to match that of the unprimed seeds. Although some post-priming treatments slightly increased P[subscript max] values for the T2 seed lot compared to the conventional priming treatment, the differences were small. For the T1 seed lot, the restoration treatments did not result in any improvement in longevity compared with the priming-only treatment. Curiously, in some cases there was not a clear decline in germination rates or viability with increasing aging time. Thus, further optimization is needed for post-priming treatments of tomato seeds to restore longevity in storage.

Book excerpt: Seed priming can markedly increase the germination rate of carrot seeds, but has not always been accompanied by improvements in field emergence. The objectives of this study were to evaluate the influence of seed priming, size and GA levels on laboratory germination and emergence of high and low quality carrot varieties. Seeds of different size were primed by incubating in a solid media of Micro-Cell E, with or without addition of GA. Control and primed seeds were germinated at 20°C and planted in peatmoss under two alternating temperatures and light of 20°C/light and 10°C/dark or 15°C/light and 10°C/dark. Priming slightly reduced final laboratory germination percentages of the high quality variety. In contrast, it increased percent germination of the low quality variety. Priming also increased speed of germination index (SEI) of both varieties. Emergence percentages and SEI were generally unaffected by priming, but emergence rates were increased. There was a strong interaction between seed lots and priming response, with the low quality variety exhibiting the greatest benefit from priming. Conductivity was a good indicator of seed quality and was reduced by seed priming. Protein analysis by gel electrophoresis showed that seed priming had no effect on storage protein mobilization. The lack of significant interaction between priming and seed size indicated that the beneficial effect of priming was most pronounced for seeds of low quality with poorer seed benefiting more from priming than high quality seed. Effect of GA on emergence and germination was variable, however, its beneficial effect on fresh weight should be considered as one way of improving early plant establishment and productivity. It is concluded that priming will enhance field performance by increasing the uniformity and rapidity of emergence, and that its effects are further enhanced by other treatments, such as sizing.

The methodology was applied both to regions such as Africa, where the crop is widely cultivated but where experimental evidence is limited, and to new regions that have not previously been associated with bambara groundnut but where environmental factors are conductive for productive growth. A weather generator and a crop simulation model of bambara groundnut (BAMnut) were incorporated into a Geographical Information System (GIS) to predict, for the first time, bambara groundnut production for the world. BAMnut is a process-based model that uses physiological principles to describe the capture and use of environmental resources principally solar radiation and soil moisture. This mechanistic approach allows crop growth and yield to be predicted for regions beyond those used in the development of the model or regions where bambara groundnut is currently cultivated. The preliminary identification of potentially suitable areas for production, based on the agro-ecological requirements of the crop, serve as a useful prelude to detailed field investigations on bambara groundnut to identify appropriate management practices and may provide a basis for a similar assessment on many other underutilized crops.--Publisher's description.
