Book excerpt: Chronic heavy alcohol consumption results in intestinal injury characterized by increased permeability, dysbiosis, nutrient malabsorption, increased risk of colorectal cancer, and functional alterations in mucosal immune cells. Studies have implicated dysregulation of tight junction proteins as well as quantitative and qualitative changes in gut microbiota as key contributors to gut injury. Recently, we showed using a macaque model of voluntary ethanol self-administration that moderate consumption resulted in a more robust T-cell and antibody vaccine response to Modified Vaccinia Ankara (MVA), while heavy drinkers generated blunted T-cell and antibody response compared to controls. First, we show that chronic alcohol consumption modulates host defense and response to booster vaccination by altering gene expression in circulating leukocytes. Using this model, we previously examined the effect of chronic ethanol consumption on cytokine production using intracellular cytokine staining, and found that colonic T cells exhibited a disruption of inflammatory cytokine production. Next, we show the modulation of transcriptome and microbiota profiles by chronic alcohol consumption within the different segments of the gastrointestinal tract. Lastly, we report alterations in the transcriptional profiles of intestinal lamina propria lymphocytes following chronic alcohol consumption. Since our animals lack overt organ damage, the alterations in gene expression and microbial composition precede clinical disease and could potentially serve as early biomarkers before detrimental effects occur. These findings provide novel insight into the mechanisms of ethanol-induced mucosal damage and lay the foundation for follow up studies to develop new interventions to reverse mucosal damage and improve immunity in heavy alcohol abusers.

Thus, in some parts of the world, population groups previously spared have become involved, which is also re flected in the increasing number of breweries and distilleries in the developing countries. Social, religious, and gender-related barriers to alcohol consumption are loosening, and the financial improvement of some segments of populations now enable them to buy alcoholic beverages. Thus the greatest percentage rise in the United States has recently been in black women. Adolescents and young people drink more alcoholic beverages than ever, and growing alcohol abuse by pregnant women has let to an increase of the incidence of the fetal alcohol syndrome. While the social and behavioral, including psychiatric, consequences of alcoholism are staggering, the gastrointestinal and, particularly, hepatic manifestations are the most widespread somatic effects, and chronic hepatic disease in alcoholics appears to cause the greatest cost to society. Indeed, mortality from liver cirrhosis is considered a reliable index of alcohol consumption in a country.

These chemical networks regulate interactions like metabolic exchange, antibiosis and symbiosis, and communication. The National Academies of Sciences, Engineering, and Medicine's Chemical Sciences Roundtable organized a series of four seminars in the autumn of 2016 to explore the current advances, opportunities, and challenges toward unveiling this "chemical dark matter" and its role in the regulation and function of different ecosystems. All these factors are a challenge to assess the impact (present and future) of parasitic diseases on public health. The intestine is a major target of these infections; it is a nutrient-rich environment harbouring a complex and dynamic population of 100 trillion microbes: the microbiome. Most researches on the microbiome focus on bacteria, which share the gut ecosystem with a population of uni- and multi cellular eukaryotic organisms that may prey on them. Our interest focuses on the families of eukaryotic microbes inhabiting the intestine, called “intestinal eukaryome”, that include fungi, protists and helminths. Knowledge on the reciprocal influence between the microbiome and the eukaryome, and on their combined impact on homeostasis and intestinal diseases is scanty and can be considered as an important emerging field. Furthermore, the factors that differentiate pathogenic eukaryotes from commensals are still unknown. This book presents an overview of the science presented and discussed in the First Eukaryome Congress held from October 16th to 18th, 2019 at the Pasteur Institute in Paris. This book covers the following topics: Phylogenetic, prevalence, and diversity of intestinal eukaryotic microbes; and their (still enigmatic) historical evolution and potential contributions to mucosal immune homeostasis. Globally alcohol misuse is the fifth leading risk factor for premature death and disability, and accounts for ~3.3 million deaths annually. Chronic alcohol use deleteriously affects both normal behavior (e.g., depression, anxiety, and alcohol craving) and physiology (e.g., oxidative stress, intestinal hyperpermeability, immune dysfunction, and organ damage). Both heavy and binge drinking patterns alter immune frequencies, compromise immune cell function, resulting in increased morbidity and mortality. Alcohol misuse can damage barrier functions in vital organs such as the lungs, gut, increase susceptibility to both bacterial and viral infections but delay wound healing. Chronic alcoholics are also at a greater risk of developing Acute respiratory distress syndrome (ARDS), alcoholic liver disease (ALD), and certain cancers. A lot of progress has been made in elucidating the relationship between alcohol consumption and immune function and how it affects human health. However, the mechanisms by which alcohol and its metabolites interacts with peripheral and tissue-resident immune systems remain poorly defined. Defining specific systemic adaptations and molecular mechanisms is important for understanding the long-term consequences of chronic alcohol use. These studies will help devise new interventions or refine existing ones to target the immune system in mitigating alcohol-related diseases. The aim of this Research Topic is to bring together new research in the alcohol field exploring the broad molecular and cellular adaptations of binge and heavy drinking on peripheral and tissue immunity. Studies would ideally include a combination of in vitro cell line models and in vivo animal models, and/or human studies of alcohol-induced end-organ damage mediated by dysregulated immune responses. Papers in this Research Topic could be used to identify complementary mechanisms and pathways that may be the focus of future therapeutic strategies. We encourage the submission of Original Research articles, as well as Reviews, Mini-Reviews, and Perspectives covering the following or related topics: 1.
