Book excerpt: Chronic systemic immunosuppression in pancreatic islet transplantation restricts its clinical application. This study aims to explore the potential of cell-mediated immune-modulation as an alternative to conventional immunosuppressive regimens; specifically investigating the innate immunosuppressive properties of human amniotic epithelial cells (AEC). Cell constructs composed of human islets and AEC (islet:AEC) were bio-engineered in rotational culture. Insulin secretory capacity and immuno-modulatory potential were characterised using appropriate in vitro assays. Fluorescence immunocytochemistry and multiplex arrays was used to identify putative mediators of the immunosuppressive response in isolated AEC monocultures. Islets and islet:AEC constructs demonstrated sustained, physiologically-appropriate insulin secretion. Resting peripheral blood mononuclear cells (PBMC) were activated on exposure to human islets but this response was significantly (p

Book excerpt: Autoimmune diseases are characterized by the occurrence of antibodies reacting with self-constituents of the body. The fully updated third edition of Autoantibodies is an in-depth review of the main autoantibodies identified up to now, with particular emphasis on those that display a diagnostic or prognostic clinical value. The new edition covers recent scientific advances, diagnostic techniques, and therapeutic technologies. Each chapter is focused on a single family of autoantibodies. This important reference contains historical notes, definitions, origins and sources of antigens recognized genetic associations, mediated pathogenic mechanisms, methods of detection, as well as clinical utility (disease prevalence and association, diagnostic value, sensitivity and specificity, prognostic value). This is an ideal reference for anyone involved in the field of autoimmune diseases. Presents all known, important autoantibodies in a single source, focusing on the antibodies needed for autoimmune disorder diagnosis Includes clinical applications for each autoantibody along with general information Organized by disease and disorder type, by autoantibody family, and completely cross-referenced

The chapters examine applications in autoimmune diseases such as lupus, rheumatoid arthritis and multiple sclerosis; blood cancers such as leukemia and lymphoma; and reproductive complications, specifically pre-term labor and use of MSCs in vitro and in animal models to discover methods of suppressing the causal inflammatory response. It also further defines the methodologies required to develop research on MSCs in vitro into established preclinical animal models including those which are proven replicas of autoimmunity and pre-term labor, to name but two. Mesenchymal Stem Cells and Immunomodulation, part of Springer’s Stem Cell Biology and Regenerative Medicine, is an invaluable resource for researchers and clinicians working with stem cells, autoimmune disease, oncology, and reproductive medicine.

This book was released on 2004 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Success of transplantation of pancreatic islets as a promising therapeutic method for restoring efficient regulated insulin secretion in type 1 diabetes depends on lifelong use of immunosuppressive drugs. With the goal of eliminating the necessity of systemic immunosuppressive agents after islet transplantation, in this doctoral research project we hypothesized that a novel non-rejectable islet graft through employing a local immunosuppressive factor, indoleamine 2, 3 dioxygenase (IDO) can be developed and applied. IDO is a tryptophan degrading enzyme and functions as a potent immunomodulatory factor. To address this hypothesis, we engineered a three-dimensional composite islet graft equipped with IDO expressing bystander cells as local immunosuppressive system. In this composite graft, expression of IDO in syngeneic fibroblasts provided a low tryptophan microenvironment within which T-cells could not proliferate and infiltrate islets. Three specific aims were accomplished in this study. We first showed that local expression of IDO in syngeneic bystander fibroblasts efficiently suppressed in vitro proliferation of lymphocytes stimulated with allogeneic pancreatic islets. In the next phase, the impact of IDO on viability and function of mouse islets embedded within IDO-expressing fibroblast-populated collagen matrix was investigated. The result showed no reduction in islet cells viability and comparable insulin content and secretion in IDO-expressing versus control preparations. In contrast to lymphocytes, a nutrient deficiency stress-responsive pathway was not activated in islets co-cultured with IDO-expressing fibroblasts confirming the selective suppressive effect of IDO on immune versus islet cells. Furthermore, when allogeneic immune response was eliminated by using a syngeneic transplant model, IDO-expressing composite islet grafts were functional in vivo for up to 100 days. Finally, to address the last specific aim, composite allogeneic islet grafts were trans.

Book excerpt: According to the World Health Organization, the epidemic of global obesity has nearly tripled since 1975. In 2016, more than 1.9 billion adults were overweight, over 650 million of which were obese. Being overweight and obese has been linked to a number of non-communicable, chronic diseases. Pathophysiology of Obesity-Induced Health Complications is a compilation of review articles dedicated to describe co-morbidities associated with obesity. The wide range that is covered is of significant interest to basic research scientists, clinicians and graduate students who are engaged in studying obesity-induced health complications. Furthermore, this book highlights the potential of novel approaches for the prevention and treatment of obesity and its related illnesses. Nineteen articles in this book are organized in four sections that are designed to provide an overview of obesity-induced health complications. The first section serves as an introductory section on the prevalence, causes, consequences, treatments and preventive approaches for obesity. Section two covers the metabolic disturbances and inflammation due to obesity. The third section is focused on neurological and visceral complications as a consequence of obesity. The final section covers strategies for the prevention of obesity-induced complications. The book illustrates that obesity can result in a diverse range of pathophysiological conditions that adversely affect health.

Book excerpt: This book is a compilation of the bench experience of leading experts from various research labs involved in the cutting edge area of research. The authors describe the use of stem cells both as part of the combinatorial therapeutic intervention approach and as tools (disease model) during drug development, highlighting the shift from a conventional symptomatic treatment strategy to addressing the root cause of the disease process. The book is a continuum of the previously published book entitled "Stem Cells: from Drug to Drug Discovery" which was published in 2017.
