Book excerpt: The identity and characteristics of aquatic soluble phosphorus (SUP) present in a small mid-western mesotropic lake were examined with 31-Phosphorus Fourier Transform Nuclear Magnetic Resonance Spectroscopy $rm (sp{31}P$ FT-NMR) and High Performance Liquid Chromatography (HPLC). Samples were concentrated and fractionated according to molecular size using a series of ultrafiltration (UF) and reverse osmosis (RO) membranes. The UF membrane pore sizes were 30,000 Da and 1,000 Da while the RO membrane had a 95% NaCl rejection rating. The SUP was analyzed as a function of apparent molecular size and season. In the 30,000 Da retentate, monoester phosphates, diester phosphates, and phophonates were detected. In the 1,000 Da retentate, monoester and diester phosphates were seen, and in the reverse osmosis retentate, orthophosphate and monoester phosphates were detected. Seasonal changes in the monoester phosphates of the 1,000 Da retentate were detected with $rmsp{31}P$ FT-NMR and HPLC. Enhancement of the $rmsp{31}P$ FT-NMR spectral resolution and sample characterization was achieved with the use of $rm Tsb1$ relaxation agents and aqueous soluble lanthanide shift reagents. A Mississippi River sample, fractionated into a 1,000K Da retentate and a RO retentate sample, was examined with $rmsp{31}P$ FT-NMR and HPLC. The SUP in the 1,000 Da retentate sample was found to be different from the lake samples in that it contained polyphosphates and possibly phosphonates, while the RO retentate was very similar to the lake RO retentate sample. This research has also demonstrated that changes occur with the SUP in the sample during concentration by UF and RO methods. It appears the phosphorus compounds are becoming incorporated into an aggregate structure which forms during the concentration procedure. Possible identities of this aggregate structure were examined using a variety of extraction and degradation methods. Extraction of SUP with ion pairing reagents and hydrophobic resins was started to be examined as a method for further enhancing SUP characterization ability by obtaining samples free from $rmsp{31}P$ FT-NMR and HPLC interferents.

Book excerpt: The concentration and distribution of soluble phosphorus (P) forms were determined in compartments of a hydrologic pathway in a forested watershed (Walker Branch, Tennessee). Rainfall, throughfall, soil water, groundwater, stream water, and water from two sites in Melton Hill reservoir downstream of Walker Branch were examined for soluble reactive and total soluble phosphorus (SRP and TSP). Soluble unreactive P (SUP) was determined from their difference. An increase of TSP from rainfall to throughfall indicated leaching or wash off of P from the canopy. SRP and SUP decreased markedly as water percolated through the soil, suggesting biological uptake and/or geochemical adsorption of phosphate groups on soil particles. Changes in soluble P. concentrations within the stream channel supported previous evidence for biological control of P dynamics in Walker Branch. Overall, SUP (an estimate of soluble organic P) constituted a significant fraction of the total soluble P present in each compartment of the flowpath. An analytical technique using high-performance liquid chromatography (HPLC) to separate the inositol phosphates (IP's) was developed and used in characterizing organic P fractions of natural systems. Commercial orthophosphate, inositol monophosphate (IMP), and inositol hexaphosphate (IHP) were adequately separated from each other on Aminex A-27 resin using a sodium chloride/tetrasodium EDTA gradient elution. The technique was used to separate an enzyme hydrolysate mixture of IP's into five components. IHP was separated from PO4 and IMP in a wheat bran extract using the HPLC method. Alkaline bromination was used to extract IP's from a Walker Branch soil sample and HPLC was used to examine the extract; at least three IP peaks were recognized. Using the HPLC technique, an attempt was made to detect the presence of IP's in a Walker Branch groundwater sample concentration by ultrafiltration. The concentration process was unsuccessful possibly due to filtration membrane leakage, so no peaks were detected. The HPLC developmental work indicated the potential usefulness of this technique in characterizing soluble organic P compounds in natural waters, leading to the identification of the inositol phosphates.

Response to phosphorus in the form of phosphoric acid was linear from 0.5 to 16 ppm phosphorus. The relative standard deviation was approximately constant at 20 percent over the range. River water and municipal sewage effluent were analyzed after the addition of phosphoric acid (1.8 ppm P) and filtration through a series of microporous membranes. Recovery of the added phosphorus averaged 70 percent for the river water and 95 percent for the sewage effluent after treatment with cation exchange resin. There was no clear relation to filter pore size in the range 5 to 0.2 micrometers. Analyses of the higher range EPA Nutrient Reference Samples (approximately 0.5 ppm P) agreed within one standard deviation with the reference values, both for inorganic and total phosphorus. The lower concentration range samples (approximately 0.1 ppm P) gave barely detectable signals. Suggestions are given for further development of the instrument. -- Abstract
