Available in PDF, EPUB and Kindle. Book excerpt: An immunological approach to the treatment of cancer has many theoretical features to commend it. There should be specificity, so that tumour cells alone are destroyed whilst normal tissues are unaffected. Provided the tumour is uni form and all of the cells have appropriate antigens, every malignant cell should be destroyed and even distant metastases dealt with. So far these speculative advantages are unfulfilled and the initial optimism that surrounded im munotherapy has not been sustained. Acceptance of the precepts of tumour im munology continues but these disappointing observations had led to increasing scrutiny of certain aspects. The purpose of this chapter is to review the prin ciples which underly tumour immunology and immunotherapy, so that the more detailed studies that follow can be considered in perspective. TUMOUR ANTIGENS (Chapter 2) For a tumour to initiate an immunological response, it must possess distinctive antigens. Much of the early work in tumour immunity was confused because it was not appreciated that tumours, like other tissues, exhibit transplan tation antigens. Only when syngeneic tumours are used can tumour antigens alone be studied and it was the introduction of inbred mouse strains which allowed Foley in 1953' to produce the first evidence for specific an tigenicity of experimental tumours. Demonstration of these antigens requires that pretreatment with syngeneic tumour will influence the growth of a sub sequent challenge with the same neoplastic cells.

Available in PDF, EPUB and Kindle. Book excerpt: In this current volume of Contemporary Topics in Immunobiology we have chosen to continue with the multiple-theme approach that was developed in Volumes 1, 3, and 5 of this series. Immunobiology still shows little sign of decreasing its active growth rate, but rather is continuing to broaden its range of interests and applications, particularly as new techniques and methods are adapted from other fields of medical research. This present volume reflects both several of the more classical areas of immunology now addressed in the light of contemporary immunology, and several newer directions that have been taken in other fields. The general subject of T-cell heterogeneity and functions of T-cell subpop ulations is addressed in Chapters 1 and 2. The potential role of genes of the major histocompatibility complex in controlling the immune functions of T lymphocytes still remains a major unresolved issue in immunogenetics, and the current status of this problem is excellently reviewed by J. F. A. P. Miller. The further elucidation of functional subpopulations of human T lymphocytes has been particularly hampered by the lack of available markers for character izing and isolating such sUbpopulations. A major step in this direction has been made by L. Moretta, M. Ferrarini, and M. D. Cooper, who review their ex perience with Fc-receptor-bearing human T-Iymphocyte populations.

Book excerpt: For many years, experiments using chimpanzees have been instrumental in advancing scientific knowledge and have led to new medicines to prevent life-threatening and debilitating diseases. However, recent advances in alternate research tools have rendered chimpanzees largely unnecessary as research subjects. The Institute of Medicine, in collaboration with the National Research Council, conducted an in-depth analysis of the scientific necessity for chimpanzees in NIH-funded biomedical and behavioral research. The committee concludes that while the chimpanzee has been a valuable animal model in the past, most current biomedical research use of chimpanzees is not necessary, though noted that it is impossible to predict whether research on emerging or new diseases may necessitate chimpanzees in the future.

The book is a compilation of his published work, but in fact it is much more than that. Whether the reader wants to read the book systematically, or only browse, it opens a fascinating world of hypotheses, theories, facts and vistas. His selection theory, his view of how immunological diversity is created, and his concept of lymphocytes interacting as a network, reveals Jerne's revolutionary spirit. The book ought to be a rich source of inspiration for everyone interested in science and how science is made.

Available in PDF, EPUB and Kindle. Book excerpt: The rapid and continuous upsurge of interesting data in the subject of tumor immunology necessitates the publication of an annual series to furnish the updated materials to the students, researchers, and clinicians in this rapidly advancing field. Concepts and methodologies are ever changing. Also, current research in tumor immunology promises to offer breakthroughs in the future. Important is the need to communicate to the right people the exact role of immunodiagnostic methods and immunolog ical intervention in cancer prevention and treatment. The role of immuno therapy in combination with conventional modalities of treatment needs in its proper perspective. Oncogene, interferon, lympho to be understood kines, monoclonal antibodies, natural killer cells, platelet-mediated cyto toxicity of antibody-coated target cells, suppressor cells, platelet-derived factors, plasma-blocking factors, control of suppressor cell function, ab rogation of plasma-blocking factors, etc. , are some of the areas that are continually advancing. Progress in these areas will have implication in cancer therapy. Further, it is already understood that if immunocompe tence of the host can be maintained at a reasonably good level, there exists the potential to increase the therapeutic indexes of conventional modalities of treatment. This series will attempt to present updated infor mation in all these areas based on contributed and solicited articles. P. K.
