Book excerpt: Abstract: Microglia are part of the innate immune system and the tissue resident macrophages of the CNS. Important functions of these immune cells are maintaining tissue homeostasis and defending the CNS against pathogens. Within brain aging microglia change to a more activated phenotype associated with hyper-responsiveness to inflammatory stimuli, which is called 'microglial priming'. This causes a heightened inflammation in the aging brain. In the recent years it has become evident that extrinsic factors like the gut microbiota and sex differences have an impact on microglial physiology. Aging is a main risk factor for neurodegenerative diseases, in which microglia as the immune cells of the CNS play an important role. Therefore, the aim of this investigation is the characterization of the impact of external factors like gut microbiota and sex on the aging process of microglia. For this purpose, we investigated male and female SPF (specific pathogen free) and GF (germfree) mice at 6-10, 50-54 and 96-104 weeks of age. Main features of microglial aging are the increase in oxidative stress and mitochondrial functional decline, as well as increasing cellular senescence and DNA-damage. In this study electron microscopy and immunohistochemical analysis was used to investigate iNOS as a marker for oxidative stress, mitochondrial ultrastructure for mitochondrial function, as well as p16INK4a and LaminB1 as markers for cellular senescence and 8OHdG as marker for DNA-damage. We could show that iNOS increased within aging in both investigated groups. This increase however was significantly lower in GF mice. Furthermore the investigation of mitochondrial ultrastructure revealed significantly higher percentage of damaged mitochondria in old SPF mice (Mossad et al., 2022). Analysis of cellular senescence and DNA-damage revealed significantly higher values in middle-aged female mice than in their male counterparts. In conclusion these results indicate that the absence of the gut microbiome has a protective effect on the increasing oxidative stress and mitochondrial functional decline within aging. Additionally, sex differences might be one factor driving microglial aging, as we detected that females display enhanced microglial aging at an earlier age than males. Future investigations should prioritize identifying mechanisms for these phenomena in order to find potential approaches for prevention and treatment of immune and neurodegenerative diseases
