This book was released on 1991-02-22 with total page 136 pages. Available in PDF, EPUB and Kindle. Book excerpt: This monograph describes the determination by X-ray methods and the structural analysis of one of the largest single polypeptide chain proteins, glycogen phosphorylase. It is shown that the large protein conforms to the rules deduced from smaller protein molecules and provides examples of almost every structural feature. The structure is analysed with special reference to the hydrogen bonding pattern of secondary structural elements, the distributions and interactions of the buried amino acids, the role of the water structure and the details of the subunit interface of the dimer. The results show some unusual secondary structural elements and environments for some side chain atoms. As methods for prediction of protein structures from amino acid sequences advance from assignments of secondary structural elements to predicted tertiary assemblies, knowledge of the preferred and exceptional detailed interactions of the amino acid side chains will need to be incorporated. Glycogen phosphorylase provides a wealth of examples of these interactions. This monograph is a useful reference on protein structure and function.

Book excerpt: This book compiles detailed information concerning a dozen of the best known allosteric enzymes, and so allows the comparison of their regulatory mechanisms and the confrontation of these mechanisms with the theoretical models. Stimulating and unexpected ideas emerge from these comparisons and emphasize the importance of developing various methods of investigation such as crystallography, X-ray solution scattering, and the study of fast movements in proteins and site-directed mutagenesis. This book is addressed to students and researchers interested in structure-function relationship in proteins, enzymology and metabolic regulation. It is also a basis for teaching.

Available in PDF, EPUB and Kindle. Book excerpt: This second edition of Medical Biochemistry is supported by more than 45 years of teaching experience, providing coverage of basic biochemical topics, including the structural, physical, and chemical properties of water, carbohydrates, lipids, proteins, and nucleic acids. In addition, the general aspects of thermodynamics, enzymes, bioenergetics, and metabolism are presented in straightforward and easy-to-comprehend language. This book ties these concepts into more complex aspects of biochemistry using a systems approach, dedicating chapters to the integral study of biological phenomena, including cell membrane structure and function, gene expression and regulation, protein synthesis and post-translational modifications, metabolism in specific organs and tissues, autophagy, cell receptors, signal transduction pathways, biochemical bases of endocrinology, immunity, vitamins and minerals, and hemostasis. The field of biochemistry is continuing to grow at a fast pace. This edition has been revised and expanded with all-new sections on the cell plasma membrane, the human microbiome, autophagy, noncoding, small and long RNAs, epigenetics, genetic diseases, virology and vaccines, cell signaling, and different modes of programmed cell death. The book has also been updated with full-color figures, new tables, chapter summaries, and further medical examples to improve learning and better illustrate the concepts described and their clinical significance. Integrates basic biochemistry principles with molecular biology and molecular physiology Illustrates basic biochemical concepts through medical and physiological examples Utilizes a systems approach to understanding biological phenomena Fully updated for recent studies and expanded to include clinically relevant examples and succinct chapter summaries

Book excerpt: The Carbohydrate Bioengineering Meeting held in Elsinore, Denmark, April 23-26, 1995, gathered 230 scientists, mostly from Europe, with interest in carbohydrate analysis and structure; carbohydrates in medicine and glycopathology; structure, function, application, and protein engineering of carbohydrate active enzymes; oligo- and polysaccharides of industrial interest; and production of carbohydrate containing new materials. The first chapters address glycoconjugates as modulatory and recognition molecules, structure determination using NMR and mass spectrometry, and microdialysis-chip enzyme-based sensors. Active site mutations coupled with crystal structures and synthetic substrate analogue interactions as well as new three-dimensional structures and binding domains for biotechnological applications are included in the chapters. Carbohydrate active enzymes turned out to be a predominant topic. The rapid development in glycobiology and glycotechnology has resulted in an enormous increase in our knowledge on the structure conversion, and application of carbohydrates in industry and medicine.

Book excerpt: Post-Translational Modifications of Proteins discusses several important topics of interest to researchers and students in protein chemistry and biochemistry, including the occurrence and function of hydroxylated residues and the three enzymes required for their formation; the damaging effects of reactions between sugars and proteins; ADP-riboosylation of proteins outside the nucleus; and Monod, Wyman, and Changeux's concerted model for allosteric control of enzyme activity exemplified by studies on glycogen phosphorylase. The application of Fast Atom Bombardment Mass Spectometry (FAB-MS) to studies on the structure and biosynthesis of various oligosaccharide moieties in protein is examined, and the understanding of the structural diversity and function of glycoprotein oligosaccharides is discussed in this volume.
