Book excerpt: Excerpt from Germination of Treated Shortleaf Pine Seed The study was made in greenhouse facilities of the University of Missouri, using 1957 Missouri seed that had been stored in sealed containers at 32 to 36 F. Moisture content of this seed before stratification was about 22 percent. Treatments studied were stratification for 20, 40, and 60 days at 38 to 40 F. Followed by storage for 2, 15, and 30 days at 38 and 60 F. Some seed lots received no coating, others the sticker only, and the rest were treated with both sticker and a mixture of two repellents. All treatment combinations were replicated four times. There were 220 sample lots of 100 seeds each. The polyethylene-plastic-bag method of seed stratification was used in this study. Seed soaked in water at 60 to 70 F. For 24 hours was drained, sealed in plastic bags, and placed in a refriger ator (fig. Repellents and sticker were applied after stratification but before storage. The sticker used was Dow Latex si2r; repellents were Endrin 25w, a rodent repellent that also possesses insecticidal properties, and sublimed anthraquinone, a bird repellent. Two pounds of Endrin 25w (25 percent concentration) and 15 pounds of anthraquinone were applied per 100 pounds of seed. Book excerpt: Excerpt from Preliminary Investigations on Dry, Cold Storage of Southern Pine Seed Early attempts at the Southern Forest Experiment Station to store south ern pine seed made use of a variety of containers, such as sealed glass jars, paper bags, and even sealed wax - paper bags. Some seed lots were disinfected with formaldehyde, other lots were untreated. No record was made of the moisture content of the seed at time of storing, nor was the germination per centage (measure of initial seed quality) always determined for the fresh seed. Temperatures during the storage period were disregarded. A concise, quantita tive summary of experiments during this early period cannot be made, but the nature of the results is readily apparent when the data are grouped loosely by species and by treatment previous to storage. The results were highly erratic and generally unsatisfactory (see table The viability of longleaf seed was usually greatly reduced within a year. For instance, the germination per centage of a longleaf seed lot dropped in 1 year from an initial value of 49 percent to 14 percent when stored, without disinfection, in sealed glass; yet the following season, the germination percentage of a lot under comparable storage conditions with 77 percent initial germination dropped in 1 year to percent. On the other hand, longleaf seed from the 1925 crop, with a ger mination of 75 percent when fresh, when stored in sealed glass after treatment with formaldehyde kept remarkably well for 1 year (germination percent) and moderately well for 2 years more percent and percent germina tion after 2 and 3 years, respectively). Since the longleaf seed were usually dried by air currents from an electric fan after treatment with formaldehyde, it was thought that this thorough drying might have been the factor responsible for the good preservation of these seed lots. This led to measurements of seed moisture content before storage, and in 1935 a cautious surmise was expressed that moisture content was important in the storage of longleaf pine seed. Germinated seeds were counted on a variable schedule, beginning after 1 day and ending after 80 days in the germinator. Seeds were considered to have germinated when their radicle appeared millimeter long. Counts were more frequent at the beginning of the incubation period when germination was peaking; nearly half of the 50 total counts were made in the first week. Some seeds germinated during the 120 day stratification period, and they were included in the first count. Seeds destroyed by fungi were recorded and removed. Ungerminated seeds were cut open at the end of incubation; full seeds, considered potentially viable, and fungi-infected seeds were counted. The mean and standard deviation of embryo development rate towards germination were estimated for each seed lot by using a procedure suggested by Campbell and Sorensen Rate is the reciprocal of days to germination if a seed requires, for example, 4 days to germinate, its embryo development rate is units per day. Mean rate is the reciprocal of days to 50 percent germination, and standard deviation of rate is the standard deviation around mean rate. We analyzed germina tion data on the rate scale rather than the day scale because the distribution was closer to normal on the rate scale.
