The public health impact and financial consequences of these diseases are particularly hard on the already overburdened economies of developing countries especially in the tropics. Many of these disease agents utilize insect hosts (vectors) to achieve their transmission to mammals. In the past, these diseases were largely controlled by insecticide-based vector reduction strategies. Now, many of these diseases have reemerged in the tropics, recolonizing their previous range, and expanding into new territories previously not considered to be endemic. Habitat change, irrigation practices, atmospheric and climate change, insecticide and drug resistance as well as increases in global tourism, human traffic and commercial activities, have driven the reemergence and spread of vector borne diseases. While these diseases can be controlled through interventions aimed at both their vertebrate and invertebrate hosts, no effective vaccines exist, and only limited therapeutic prospects are available for their control in mammalian hosts. Molecular technologies such as transgenesis, which is the subject of this book, stand to increase the toolbox and benefit disease management strategies.

Mobile genetic elements (MGEs) constitute much of the repetitive component, particularly the interspersed sequences. This volume of nine chapters addresses MGEs that have colonized the genomes of medically important metazoan parasites and their vectors. These MGEs are parasites of parasites. These mobile elements are compared and contrasted with those from better-studied model organisms. The authors have included plentiful, relevant, and informative illustrations and references. Several chapters focus on mobile elements of the human schistosomes. Gerardo Ferbeyre focuses on the putative ribozyme activities on the SM alpha elements, SINE-like elements, including trans reactions catalyzed by these retroposons. Craig Coates provides an expert article on transposon mediated transgenesis of mosquitoes, in particular dealing with his pioneering studies on the deployment of binary systems with the Mos1 mariner transposon. MGEs from the genomes of these metazoan parasites have practical uses in transgenesis and epidemiological studies and also are valuable for annotation of the genomes of these vectors and pathogens. These issues and future directions are well covered in these chapters.

Even though most of our understand ing about the factors that contribute to a successful tagging experiment has been accumulated from a limited number of experiments using different transposable elements in different genetic backgrounds, it is still possible to draw some conclusions regarding the best experimental strategies for gene tagging. In our experience, Spm has proved to be a good element for transposon tagging. The frequency of recovering mutable alleles in duced by Spm is not significantly different from that for Ac-Ds or for Mu 6 (summarized in Ref. 22) and varies from about 10- to 10=zr:-8pm has the unique advantage, however, in that all of the members of thiSfumily that have been examined thus far are homologous to each other at the DNA level. Therefore, by combining molecular analysis with genetic segre gation, it is possible to identify and isolate alleles that are due to insertions of either autonomous or nonautonomous Spm elements. There are definite steps one can take to increase the chances of de tecting a transposition into the gene of interest. The most important step is to select a genetic background in which the desired phenotype will be easy to screen. If the phenotype is not likely to be mutable, then tester lines should be constructed so as to contain flanking markers that can aid in subsequent segregation analyses.
