The lack of Ado kinase activity (ADK) in Arabidopsis thaliana adk1 adk2 double mutants results in embryonic lethality; reduction of ADK expression causes a pleiotropic phenotype due to the accumulation of Ado inhibiting transmethylation activities. The phenotype of ADK mutants shows that this enzyme plays a critical role in Ado salvage but the functional significance of the other putative Ado recycling enzymes Ado deaminase (ADA) and Ado nucleosidase (ADN) in Arabidopsis thaliana have yet to be elucidated. ADA catalyzes the irreversible deamination of Ado to inosine. The locus At4g04880 (AtADA) of A. thaliana is annotated as encoding a putative ADA, based on its amino acid sequence similarity and the presence of important, conserved catalytic residues. However, indirect and direct spectrophotometric activity assays of the recombinant enzyme demonstrated that the gene product of this locus does not possess ADA activity; complementation experiments to test for the functionality of the AtADA product in A. thaliana and E. coli confirmed its lack of ADA activity. Instead, phylogenetic analysis revealed that AtADA belongs to the group of ADA-like (ADAL) proteins, a group closely related to ADAs that to date have not been shown to have ADA activity. AtADA is no exception as it also lacks ADA activity based on the in vivo and in vitro experiments outlined in this thesis. Thus, the locus At4g04880 should be re-annotated as ADAL. The question of the function of AtADAL cannot be answered as of yet; in general, the knockout of ADA gene product demonstrated that At4g04880 is not essential for Arabidopsis growth. Since no further ADA-related genes exist in the genome of Arabidopsis it is concluded that ADA activity is not present in this plant. ADN catalyzes the conversion of purine and pyrimidine ribosides to their corresponding bases; although it prefers Ado as a substrate it also acts on cytokinins. The activity of this enzyme has been described in several plant species but no corresponding genes have been identified to date. The genome of Arabidopsis was screened for ADN genes using an inosine-uridine nucleoside hydrolase sequence from the protozoa Crithidia fasciculata. Two genes, annotated as ADN1 and ADN2 were identified and their gene products were studied using a spectrophotometric assay. The substrate spectrum of ADN2 includes both purine and pyrimidine nucleosides but it prefers to utilize uridine. Thus, ADN2 is proposed to be involved in the purine and pyrimidine salvage in Arabidopsis but predominantly in uridine recycling. Recombinant ADN1 did not show activity on any of the tested substrates. Even though the in vivo role of both ADNs is still uncertain, due to their lack or low activity on Ado there may yet be the ADN gene in the Arabidopsis genome which likely acts on both adenosine and cytokinin ribosides.

Book excerpt: The cereal endosperm is an excellent system to study cell differentiation and fate specification. The aleurone of maize is one cell layer and requires continuous positional cues to specify and maintain its identity. A genetic hierarchy is implicated in this process. CRINKLY4 (Cr4), a receptor-like kinase, belongs to cell fate-specifying genes and is required to specify aleurone cell identity. Viviparousl (Vpl), a B3 domain-containing transcription factor, functions late in development to regulate the expression of Cl in aleurone cells. It also plays an important regulatory role in late embryogenesis and seed maturation with abscisic acid (ABA). The Arabidopsis genome encodes ACR4, an ortholog of CR4, and four AtCRRs. ACR4 and AtCRRl and 2 are mainly expressed in developing young tissues. The kinase domain of ACR4 was shown to be a serine/threonine kinase and had ability to phosphorylate the non-functional kinase domain of AtCRR2. T-DNA insertion mutants of acr4 showed a phenotype restricted to the integument and seed coat, while mutants of atcrrs did not show obvious phenotype and triple mutants of acr4, atcrrl and 2 had similar phenotype as acr4, suggesting that Arabidopsis might have a redundant function and the hypothetical redundant function is not encoded by AtCRRs. Vpl functions in ABA regulated processes. Its expression is regulated by ABA, high salinity and osmoticum. The induction of Vpl expression by ABA does not require de novo protein synthesis. The sequence analysis of 958 bp of the Vpl promoter identified a potential ABRC (ABA response complex), which consists a predicted ABRE (ABA response element) and a CEIL (coupling element 1-like) element. Electrophoretic mobility shift assay (EMSA) confirmed that the ABRE and CEIL specifically bound proteins in 20 DAP embryo nuclear protein extracts. ABA or osmoticum induce nuclear protein interaction with the ABRE. The ABRE was also shown to be critical for the induction of Vpl expression by ABA in a transient gene expression experiment. Sequence analysis and EMSA experiments also identified several other binding sites such as: VPP1, AtMYB2 and ZIP1. This provides a starting point to further study the regulation of Vpl expression.

Book excerpt: The addition of sugar to cell wall polysaccharides and proteins is mainly catalyzed by glycosyltransferases (GTs). Plant O-fucosyltransferases (O-FuTs) are a type of GTs that catalyze fucose transfer from the donor, guanosine diphosphate fucose (GDP-Fuc), to various acceptor molecules, including oligosaccharides, glycoproteins and glycolipids. Previous microarray analysis identified a group of genes that were highly expressed in embryogenic B. napus microspore culture cells in as early as 3 days after microspore embryogenesis induction. Of special interest was the Brassica transcript homologous to the Arabidopsis gene AT2G44500 since it was consistently upregulated (threefold increase) in embryogenic cells compared to pollen-like/non-responsive cells. This Arabidopsis gene contains a GDP-O-fucosyltransferase (O-FuT) domain. In the present study, the complete genomic and coding sequences of At2G44500 homologs were isolated and sequenced from B. napus cv. Topas. Sequence analyses of At2G44500 and its B. napus homolog, BnAt2G (1) suggest that they encode type II-membrane proteins that contain conserved key amino acids essential for O-FuT catalytic activity. BnAt2G(1)/At2G44500 are differentially expressed throughout plant development. In particular, At2G44500 was shown to be highly expressed in the shoot apical meristem (SAM) and young vegetative tissues in A. thaliana. BnAt2G(1) was also shown to be expressed at higher levels in B. napus embryogenic microspore cultures as well as in vegetative tissues. The putative transcription control elements identified in the in silico promoter analysis of At2g44500/BnAt2G(1) suggest that their gene expression is controlled by various transcription factors that are involved in hormone-regulated stress responses. At2G44500 has also been shown to be co-expressed with genes that have known functions in cell wall biosynthesis, embryogenesis and signal transduction. The sub-cellular localization of a GFP.
