Available in PDF, EPUB and Kindle. Book excerpt: A large number of chemical compunds are constantly being introduced and produced, both to assist and ease modern human life. Among these chemicals, industrial compounds represent a particular fraction of chemicals which are not intended for use in biological systems, but to which humans may be accidently exposed, whether in the workplace, by product application, or via the environment. Industrial chemicals, as opposed to pharmaceuticals and agrochemicals, are in many cases subjected only to a very basic examination of handling safety, and may lack further toxicity testing. This implies that essentially nothing is known about their bioavailability, metabolism, excretion and toxicological properties, unless problems arise.; Covering the investigation of industrial chemicals, this book focuses on their individual structure, biological fate, potential toxicity to mammals and the molecular mechanisms possibly underlying their adverse effects by highlighting the use and significance of experimental toxicology. Special emphasis is placed on mechanistic aspects in the safety assessment of industrial compounds, in addition to current regulatory and legal considerations.

Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

This book was released on 2000-09-07 with total page 92 pages. Available in PDF, EPUB and Kindle. Book excerpt: Manufactured vitreous fibers (MVF), also known as synthetic vitreous fibers, are considered to be less hazardous than asbestos to human health. They are used in many thermal- and acoustical-insulation applications as an asbestos substitute or as a filtration medium. The Navy uses MVF in shipboard and onshore applications. To protect Navy personnel from harmful exposures to MVF, the U.S. Navy Environmental Health Center (NEHC) developed occupational exposure standards. The documentation assists industrial hygienists, occupational medicine physicians, and other Navy health professionals in assessing and controlling the health hazards linked with exposure to MVF. In 1997, the National Research Council (NRC) was asked to conduct an independent review of the Navy's toxicological assessment of MVF and to evaluate the scientific validity of its exposure standard of 2 fibers per cubic centimeter of air (f/cm3). The NRC assigned the task to the Committee on Toxicology, which established the Subcommittee on Manufactured Vitreous Fibers, a multidisciplinary group of experts, to determine whether all relevant toxicological and epidemiological data were correctly considered in developing the exposure standard; and to examine the uncertainty, variability, and quality of data and the appropriateness of assumptions used in the derivation of the exposure standard. The subcommittee was also asked to identify deficiencies in the MVF database and, where appropriate, to make recommendations for future research and data development. Review of the U.S. Navy's exposure Standard for Manufactured Vitreous Fibers represents the subcommittee's final report. The committee had expanded its review when in January 1999, the Navy revised its Occupational Safety and Health Program Manual (CNO 1999), changing the occupational exposure limit for MVF to the American Conference of Governmental Industrial Hygienists (ACGIH) threshold limit value (TLV) of 1 f/cm3. The report features recommendations by the subcommittee as well as information gaps found throughout investigation. Overall, the subcommittee found that the Navy made a good start in assessing the health effects of MVF, but needed further research.

Asbestos is a group of 6 different fibrous minerals that occur naturally in the environment. All forms of asbestos are hazardous, and all can cause cancer. This profile includes: (1) The exam¿n. and interpretation of toxicologic info. and epidemiological eval¿s. on asbestos to ascertain the levels of human exposure for the substance and its health effects; (2) A determination of whether adequate info. on the health effects of asbestos is available or in the process of development to determine levels of exposure that present a significant risk to human health; and (3) Where appropriate, identification of toxicologic testing needed to identify the types or levels of exposure that may present significant risk of adverse health effects in humans. Book excerpt: Advances in Molecular Toxicology features the latest advances in all of the subspecialties of the broad area of molecular toxicology. Toxicology is the study of poisons, and this series details the study of the molecular basis by which a vast array of agents encountered in the human environment and produced by the human body itself manifest themselves as toxins. Not strictly limited to documenting these examples, the series is also concerned with the complex web of chemical and biological events that give rise to toxin-induced symptoms and disease. The new technologies that are being harnessed to analyze and understand these events will also be reviewed by leading workers in the field. Advances in Molecular Toxicology will report progress in all aspects of these rapidly evolving molecular aspects of toxicology with a view toward detailed elucidation of both progress on the molecular level and on advances in technological approaches employed. - Cutting-edge reviews by leading workers in the discipline - In-depth dissection of molecular aspects of interest to a broad range of scientists, physicians and any student in the allied disciplines - Leading edge applications of technological innovations in chemistry, biochemistry and molecular medicine
