Book excerpt: It has recently become clear that the excitatory amino acids and their receptors are critically linked to normal processes of development and synaptic transmission, and to learning and memory, as well as to identifiable disease processes such as Alzheimer's disease, epilepsy, and cortical damage due to stroke/ischemia. Moreover, the pharmacological nature and chemical structures of many of the receptors and binding sites for these synaptic mediators and their modulators are becoming known, thereby enabling the cloning of each receptor subtype. Such advances may help immeasurably in the identification of endogenous ligands for the amino acid receptors and the development of pharmaceuticals and other therapeutic interventions in coming years.

Despite this fact, no validated biological markers, or biomarkers, currently exist for measuring glutamate pathology in CNS disorders or injuries. Glutamate dysfunction has been associated with an extensive range of nervous system diseases and disorders. Problems with how the neurotransmitter glutamate functions in the brain have been linked to a wide variety of disorders, including schizophrenia, Alzheimer's, substance abuse, and traumatic brain injury. These conditions are widespread, affecting a large portion of the United States population, and remain difficult to treat. Efforts to understand, treat, and prevent glutamate-related disorders can be aided by the identification of valid biomarkers. The Institute of Medicine's Forum on Neuroscience and Nervous System Disorders held a workshop on June 21-22, 2010, to explore ways to accelerate the development, validation, and implementation of such biomarkers. Glutamate-Related Biomarkers in Drug Development for Disorders of the Nervous System: Workshop Summary investigates promising current and emerging technologies, and outlines strategies to procure resources and tools to advance drug development for associated nervous system disorders. Moreover, this report highlights presentations by expert panelists, and the open panel discussions that occurred during the workshop.

Book excerpt: This past decade has led to many significant advances in the understanding of the function of excitatory amino acids in synaptic transmission. The cloning of the ionotropic and metabotropic glutamate receptor families of receptor proteins has produced new strategies for the pharmacological modulation of glutamate transmission. The engineering of transgenic animals with modified expression of receptor proteins has created new insights into the function, dysfunction and possible pathology causally related to glutamate receptors. Advances in the pharmacology of glutamate receptors has led to clinical research addressing multiple therapeutic applications of drugs that act on excitatory amino acid systems. A number of NMDA receptor anatagonists have now been studied in humans. AMPA/kainate and metabotropic receptor active compounds have left the preclinical realms of research and have moved towards or are in the clinic.

Book excerpt: Glutamate is the major excitatory neurotransmitter in the mammalian central nervous system (CNS). It regulates normal CNS function, is a major participant in pathology, and serves learning, memory, and higher cognitive functions. The 12 chapters of this book provide the first comprehensive coverage of all the major features of glutamate as excitatory neurotransmitter. The book begins with a valuable historical backdrop. Building from a chapter on the common structure of glutamate receptors, several others cover the major ionotropic receptors, their structure, function, and pharmacology. A follow-up chapter discusses the metabotropic receptors that are directly coupled to second messenger metabolism. A central theme of the book is the capacity of the excitatory amino acid system to contribute to the diverse array of signaling systems in the CNS as a direct result of the large assortment of receptors (including, for the ionotropic receptors, various subunits) the combination of which determine distinct functional properties. A recent development in the field discussed in several chapters is the biochemical characterization of a supermolecular protein complex, the post-synaptic density, that makes the unique structure of excitatory synapses. This complex subserves the experience-dependent modulation of synaptic strength and synaptic plasticity, and gives the synapse the capacity to change dynamically in both structure and receptor composition. Drawing on the individual properties of the receptors, transporters, and functional architecture of the synapse, the concluding chapters describe the functional integration of these components in the more complex physiological processes of plasticity and pathology. Recognition that the regulation of excitatory amino acid receptor activity underlies the pathology of many neurological diseases, including stroke, Alzheimer's disease and schizophrenia, has opened up an exciting frontier that will allow the translation of our understanding of these basic mechanisms into new concepts of pathology and new therapeutic strategies. This book will be invaluable for neuroscientists, pharmacologists, neurologists, and psychiatrists, and for their students and trainees.

Available in PDF, EPUB and Kindle. Book excerpt: Epilepsy is a devastating group of neurological disorders characterized by periodic and unpredictable seizure activity in the brain. There is a critical need for new drugs and approaches given than at least one-third of all epilepsy patients are not made free of seizures by existing medications and become "medically refractory". Much of epilepsy research has focused on neuronal therapeutic targets, but current antiepileptic drugs often cause severe cognitive, developmental, and behavioral side effects. Recent findings indicate a critical contribution of astrocytes, star-shaped glial cells in the brain, to neuronal and network excitability and seizure activity. Furthermore, many important cellular and molecular changes occur in astrocytes in epileptic tissue in both humans and animal models of epilepsy. The goal of Astrocytes and Epilepsy is to comprehensively review exciting findings linking changes in astrocytes to functional changes responsible for epilepsy for the first time in book format. These insights into astrocyte contribution to seizure susceptibility indicate that astrocytes may represent an important new therapeutic target in the control of epilepsy. Astrocytes and Epilepsy includes background explanatory text on astrocyte morphology and physiology, epilepsy models and syndromes, and evidence from both human tissue studies and animal models linking functional changes in astrocytes to epilepsy. Beautifully labelled diagrams are presented and relevant figures from the literature are reproduced to elucidate key findings and concepts in this rapidly emerging field. Astrocytes and Epilepsy is written for neuroscientists, epilepsy researchers, astrocyte investigators as well as neurologists and other specialists caring for patients with epilepsy. - Presents the first comprehensive book to synthesize historical and recent research on astrocytes and epilepsy into one coherent volume - Provides a great resource on the field of astrocyte biology and astrocyte-neuron interactions - Details potential therapeutic targets, including chapters on gap junctions, water and potassium channels, glutamate and adenosine metabolism, and inflammation

Book excerpt: This is the second time that I have had the honor of opening an interna tional symposium dedicated to the functions of the hippocampus here in Pecs. It was a pleasure to greet the participants in the hope that their valuable contributions will make this meeting a tradition in this town. As one of the hosts of the symposium, I had the sorrowful duty to remind you of the absence of a dear colleague, Professor Graham God dard. His tragic and untimely death represents the irreparable loss of both a friend and an excellent researcher. This symposium is dedicated to his memory. If I compare the topics of the lectures of this symposium with those of the previous one, a striking difference becomes apparent. A dominating tendency of the previous symposium was to attempt to define hippocam pal function or to offer data relevant to supporting or rejecting existing theoretical positions. No such tendency is reflected in the titles of the present symposium, in which most of the contributions deal with hip pocampal phenomena at the most elementary level. Electrical, biochemi cal, biophysical, and pharmacological events at the synaptic, membrane, or intracellular level are analyzed without raising the question of what kind of integral functions these elementary phenomena are a part of.

Book excerpt: The thirty original contributions in this book provide an up-to-date, interdisciplinary account of current research activity in all aspects of excitatory amino acid transmitters, particularly glutamate and aspartate, in the context of the structure and function of the cerebral cortex in health and in disease. It has recently become clear that the excitatory amino acids and their receptors are critically linked to normal processes of development and synaptic transmission, and to learning and memory, as well as to identifiable disease processes such as Alzheimer's disease, epilepsy, and cortical damage due to stroke/ischemia. The topics are dealt with from electrophysiological, biochemical, anatomical, pharmacological, clinical, and behavioral perspectives to provide a thorough account of the functional roles played by these compounds in the part of the brain dealing with higher processes. The material is broad-based yet presented in a style that allows clinical and basic scientists alike easy access to integrated material across chapters and between sections.
