Presently to some extent it is also being used in Commercial Bovine Frozen Semen Production Laboratories of the world for evaluating the quality of spermatozoa before use in artificial breeding services. It is also made mandatory on such laboratories under minimum standard protocols of concerned Governments. The system provides useful information in numerical as well as percentage values on sperm motility and velocity parameters such as progressive sperm motility, average path velocity, straight line velocity, curvilinear velocity, straightness, linearity, elongation as well as static, slow, medium and rapidly motile sperm proportion of a given semen sample of ox- and buffalo-bulls, both fresh and frozen-thawed semen. There were very good interrelationships among various parameters tested, particularly with progressive motile and hypo-osmotic swelling reactive spermatozoa.

Book excerpt: Master's Thesis from the year 2015 in the subject Biology - Zoology, , language: English, abstract: In the present study semen samples from NRF bulls were analysed for sperm viability, acrosome integrity and ATP content. The effect of incubation at 37 °C on sperm viability, acrosome integrity and ATP content was investigated on fresh and frozen semen samples from 20 NRF. Semen was incubated at 37 °C and the sperm quality parameters were analysed at 0 hr and after 6 and 24 hrs of incubation for both fresh and frozen samples. Sperm viability and acrosome integrity was assessed with flow cytometry and ATP content with luminometer. The incubation time at 37 °C had a significant effect on all studied parameters. However, ATP content was significantly more affected than sperm viability and acrosome integrity. After 24 hrs of incubation ATP content of frozen samples decreased approximately to 0 for all tested samples, while percentage of viability and acrosome integrity decreased by 78 ± 11.9 %. Percentage of AIL sperm cells of frozen samples decreased at corresponding rate as in fresh samples. Concerning ATP content there was however, a more marked decline in frozen samples compared to fresh samples during incubation. Sperm samples from eight NRF bulls with high and low fertility were also analysed for sperm viability, acrosome integrity and ATP content at 0 hr (right after thawing), 3, 6 and 24 hrs incubation at 37 °C. The ATP content adjusted for % AIL at 0 hr was significantly correlated with fertility measured as 56 days NRR, while % AIL or ATP content analysed separately were not correlated to field fertility.

From the earliest works to the present day, a considerable number of analytical techniques have been developed. These tests study many aspects of the morphology and physiology of the spermatozoan; however, their ability to predict male fertility remains low. Some of these techniques have been automated, which can make the test results more objective, but others still require subjective evaluation. The introduction of computer-assisted sperm analysis (CASA) systems and flow cytometry has revolutionized sperm quality analysis in recent decades. However, the use of these advanced techniques remains experimental, with only a few of them having successfully led to practical applications in routine commercial semen evaluation. Future sperm biology research should focus on developing analytical techniques that have a greater capacity to predict male fertility and can be used in both the laboratory and the field. Field adaptation will require progress in automation and simplification, to produce precise, economical and efficient techniques. Nevertheless, as technology advances in these analytical tests and research continues, our greater understanding of male fertility will aid the development of new methods of sperm evaluation.
