In this subchapter, we will delve into the world of epigenetics, demystifying its concepts and shedding light on its profound implications for genetics and genomics. At its core, epigenetics refers to the study of heritable changes in gene expression that do not involve alterations to the underlying DNA sequence. It explores how environmental factors and lifestyle choices can influence gene activity and subsequently affect our health and development. Epigenetic modifications act as switches, turning genes on or off, and thus playing a critical role in determining an individual's traits and susceptibility to diseases. One of the key mechanisms of epigenetic regulation is DNA methylation, a process where methyl groups attach to specific regions of DNA, marking them as "silent" and preventing gene expression. Another mechanism is histone modification, where proteins called histones, around which DNA is wound, undergo chemical changes that either promote or inhibit gene activity. These epigenetic marks can be passed on from one generation to another, leading to the inheritance of certain traits or diseases. Epigenetics also challenges the long-held belief that our genetic destiny is fixed and unchangeable. It reveals that our genes are not our fate, and that our lifestyle choices can have a profound impact on our gene expression and overall health. Studies have shown that factors such as diet, stress, exercise, and exposure to toxins can all influence epigenetic modifications. Understanding epigenetics has far-reaching implications for personalized medicine and disease prevention. By unraveling the epigenetic changes associated with various diseases, scientists can develop targeted therapies and interventions to reverse or mitigate these alterations. Furthermore, epigenetic research has shed light on the interplay between nature and nurture, providing valuable insights into how our environment can shape our genetic potential.

Book excerpt: This book is an introduction to epigenetics, a controversial term that denotes the mechanisms that instruct the genome on how to express the purely genetic information that encodes proteins. Starting with the discovery of repressor proteins in the 1960s, epigenetics evolved into a kind of user manual for the genetic information, telling the genome if, when, how much and in what cells to read genes. Advances in epigenetics in the past 15 years have revealed how it lies at the heart of virtually every branch of biological and medical sciences and an understanding of its basic principles is therefore essential for every student in this field. The field of chromatin and epigenetics has developed very rapidly in the past 15-20 years. The pace in the field of epigenetics has now slowed down, the basic outlines of the mechanisms and implications have solidified and a consensus has been achieved among the researchers in the field. At the same time, these mechanisms and implications are now an integral part of how we think about the genome. Genes are more than just DNA and more than just protein-coding sequences and this realization has revolutionized our understanding of the genome, gene expression and its regulation in development, health and disease. It is time, therefore, for a textbook to help train a new generation of biologists and health scientists as well as providing a basic competence among practitioners in allied fields. The present book has grown out of a course given for the past 13 years to advanced undergraduates at Rutgers University. In keeping with the experience in that course, the book is abundantly illustrated, presents a wealth of specific examples, and includes a chapter describing a number of methods and techniques that have driven the advances in the field.

It explains why mapping an organism’s genetic code is not enough to determine how it develops or acts and shows how nurture combines with nature to engineer biological diversity. Surveying the twenty-year history of the field while also highlighting its latest findings and innovations, this volume provides a readily understandable introduction to the foundations of epigenetics. Nessa Carey, a leading epigenetics researcher, connects the field’s arguments to such diverse phenomena as how ants and queen bees control their colonies; why tortoiseshell cats are always female; why some plants need cold weather before they can flower; and how our bodies age and develop disease. Reaching beyond biology, epigenetics now informs work on drug addiction, the long-term effects of famine, and the physical and psychological consequences of childhood trauma. Carey concludes with a discussion of the future directions for this research and its ability to improve human health and well-being.

Available in PDF, EPUB and Kindle. Book excerpt: The view “It’s all in our genes and we cannot change it” developed in the past 150 years since Gregor Mendel’s experiments with flowering pea plants. However, there is a special form of genetics, referred to as epigenetics, which does not involve any change of our genes but regulates how and when they are used. In the cell nucleus our genes are packed into chromatin, which is a complex of histone proteins and genomic DNA, representing the molecular basis of epigenetics. Our environment and lifestyle decisions influence the epigenetics of our cells and organs, i.e. epigenetics changes dynamically throughout our whole life. Thus, we have the chance to change our epigenetics in a positive as well as negative way and present the onset of diseases, such a type 2 diabetes or cancer. This textbook provides a molecular explanation how our genome is connected with environmental signals. It outlines that epigenetic programming is a learning process that results in epigenetic memory in each of the cells of our body. The central importance of epigenetics during embryogenesis and cellular differentiation as well as in the process of aging and the risk for the development of cancer are discussed. Moreover, the role of the epigenome as a molecular storage of cellular events not only in the brain but also in metabolic organs and in the immune system is described. The book represents an updated but simplified version of our textbook “Human Epigenomics” (ISBN 978-981-10-7614-8). The first five chapters explain the molecular basis of epigenetics, while the following seven chapters provide examples for the impact of epigenetics in human health and disease.

Book excerpt: Epigenetics is defined as the study of modifications of the genome, heritable during cell division that does not involve changes in DNA sequences. Up to date, epigenetic modifications involve at least three general mechanisms regulating gene expression: histone modifications, DNA methylation, and non-coding RNAs (ncRNAs). For the past two decades, an explosion in our interest and understanding of epigenetic mechanisms has been seen. This mainly based on the influence that epigenetic alterations have on an amazing number of biological processes, such as gene expression, imprinting, programmed DNA rearrangements, germ line silencing, developmentally cued stem cell division, and overall chromosomal stability and identity. It has become also evident that the constant exposure of living organisms to environment factors affects their genomes through epigenetic mechanisms. Viruses infecting animal cells are thought to play central roles in shaping the epigenetic scenario of infected cells. In this context it has become obvious that knowing the impact that viral infections have on the epigenetic control of their host cells will certainly lead to a better understanding of the interplay viruses have with animal cells. In fact, DNA viruses use host transcription factors as well as epigenetic regulators in such a way that they affect epigenetic control of gene expression that extends to host gene expression. At the same time, animal cells employ mechanisms controlling transcription factors and epigenetic processes, in order to eliminate viral infections. In summary, epigenetic mechanisms are involved in most virus-cell interactions. We now know that some viruses exhibit epigenetic immune evasion mechanisms to survive and propagate in their host; however, there is still much ambiguity over these epigenetic mechanisms of viral immune evasion, and most of the discovered mechanisms are still incomplete. Other animal viruses associated to cancer often deregulate cellular epigenetic mechanisms, silencing cellular tumor-suppressor genes and/or activating either viral or host cell oncogenes. In addition, in several cancers the down-regulation of tumor suppressor protein-coding genes and ncRNAs with growth inhibitory functions, such as miRNAs, have been closely linked to the presence of cell CpG island promoter hypermethylation. The goal of the aforementioned Research Topic is to bring together the key experimental and theoretical research, linking state-of-the-art knowledge about the epigenetic mechanisms involved in animal virus-cell interactions.

Available in PDF, EPUB and Kindle. Book excerpt: The study of epigenetics, or how heritable changes in gene expression are regulated without modifying the coding DNA sequence, has become an increasingly important field of study in recent years. Rapid developments in our understanding of the way in which gene function is modulated by the environment has revolutionized the way we think about human development and health. Nutrition in Epigenetics reviews the latest research looking at the interaction between genes and nutrients and the role they play together in maintaining human health. Nutrition in Epigenetics is divided into two primary parts. The first part provides key principles such as epigenetic mechanisms, developmental epigenetics, and the role of epigenetics in disease. The second part looks specifically at the application of epigenetics to the field of human nutrition. Chapters review the role of specific nutrients in modulating epigenetic status and the effect on health and disease. Nutrition in Epigenetics is an indispensable resource for researchers, professionals and advanced students with an interest in human nutrition, epigenetics, and biomedical research.

Book excerpt: Your genes play an important role in your health, but so do your behaviors and environment, such as what you eat and how physically active you are. Epigenetics is the study of how your behaviors and environment can cause changes that affect the way your genes work. Unlike genetic changes, epigenetic changes are reversible and do not change your DNA sequence, but they can change how your body reads a DNA sequence. Gene expression refers to how often or when proteins are created from the instructions within your genes. While genetic changes can alter which protein is made, epigenetic changes affect gene expression to turn genes "on" and "off." Since your environment and behaviors, such as diet and exercise, can result in epigenetic changes, it is easy to see the connection between your genes and your behaviors and environment. Researchers studying the microscopic roundworm Caenorhabditis elegans recently discovered a set of mutations that extended the worms' normal 2-3 week lifespan by up to 30%. This was exciting, not least because discoveries in animals such as roundworms can sometimes help us understand processes like ageing in humans. This was not the end of the story though, as the researchers found that the descendants of the long-lived roundworms could also live longer than normal, even if they only inherited the non-mutated version of the genes from their parents. This doesn't seem to make sense at first; surely characteristics such as hair colour, height and even how long we or a microscopic worm could potentially live are carried in the DNA sequence of the genes that we inherit from our parents. So how can we solve the conundrum of how the roundworms inherited the long lived characteristic, without inheriting the DNA sequence that initially caused it? The answer is epigenetics.
