Available in PDF, EPUB and Kindle. Book excerpt: The endothelium, the cell layer that forms the inner lining of blood vessels, is a spatially distributed system that extends to all areas of the human body. Clinical and basic research demonstrates that the endothelium plays a crucial role in mediating homeostasis and is involved in virtually every disease, either as a primary determinant of pathophysiology or as a victim of collateral damage. The endothelium has remarkable, though largely untapped, diagnostic and therapeutic potential. This volume bridges the bench-to-bedside gap in endothelial biomedicine, advancing research and development and improving human health. The book is the first to systematically integrate knowledge about the endothelium from different organ-specific disciplines, including neurology, pulmonary, cardiology, gastroenterology, rheumatology, infectious disease, hematology-oncology, nephrology, and dermatology. It's interdisciplinary approach, which draws on expertise from such diverse fields as evolutionary biology, comparative biology, molecular and cell biology, mathematical modeling and complexity theory, translational research, and clinical medicine.

Available in PDF, EPUB and Kindle. Book excerpt: The participation of endothelial cells in various physiologic and pathologic processes has been hypothesized since before the turn of the century. However, until recently, direct evidence for endothelial involvement in these processes has been extremely difficult to obtain due to the inability to study endothelial cell function in vitro. Though the possibility of using cultured endothelial cells to study endothelial cell function in vitro was recognized many years ago, the inability to culture unambiguously identifiable endothelial cells limited investigators in their studies of endothelial function. As a result, the field of endothelial cell biology lay relatively fallow for many years. The development in the early 1970's of routine and easily implemented methods for culturing human endothelial cells and the demonstration that cultured endothelial cells synthesized a physiologically relevant protein, Factor VIII/von Willebrand Factor, quickly changed this state of affairs. Over the following decade the scope of endothelial cell research rapidly widened, spreading in a number of directions. First, methods were developed to culture endothelial cells from a variety of species. Second, methods were developed to culture endothelial cells from different organs and types of blood vessels (arteries, veins, and capillaries) within a single species. Third, and most important, investigators began using cultured endothelial cells as tools to study the potential involvement of endothelial cells in a wide assortment of biologically interesting processes. The net result has been a tremendous increase in our understanding of endothelial cell function.

Book excerpt: Although blood capillaries were first observed through a flea-lens microscope by Malpighi in 1661,200 more years elapsed before the cellular nature of the vessel wall was conclusively demonstrated. Beginning with the middle of the 19th cen tury, our knowledge of the histological organization of blood vessels has steadily increased. However, the endothelium, which for a long time was considered to be just an inert barrier lining, had been barely explored until three decades ago. Since then, there has been an upsurge of interest in the fine structure and function of endothelial cells. Intense in vivo and in vitro investigations have revealed that the endothelial cell is a key element in a wide variety of normal activities and diseases. A large number of investigators and laboratories have been attracted to endothelial cell research, thus supporting the expansion of the continuously grow ing and diversifying field of endotheliology. The number of articles published annually on this subject has increased from a few score at the beginning of the 1970s to more than a thousand in recent years, and an increasing number of journals, books, societies, and symposia focused primarily on the vascular en dothelium have marked the last decade.

Book excerpt: Endothelial cell biology has developed into a vibrant discipline and has become a critical instrument to study several disease processes on the cellular and molecular level. It is now widely recognized that dysfunctions of normal endothelial cell homeostasis are involved in some of the most important human diseases, including ischemic heart diseases, hypertension, atherosclerosis, tumors, diabetes, arthritis, and inflammation. Further, the increasing importance and recognition of the field of vascular biology in general requires in vitro and in vivo techniques in order to address the complex questions. Methods in Endothelial Cell Biology is a comprehensive practical "how-to"-guide summarizing the most relevant established techniques as well as a number of new emerging techniques. Easy-to-follow reliable protocols provide a useful lab bench resource for the experienced researcher and newcomer to the field.

Book excerpt: Endothelial Signaling in Vascular Dysfunction and Disease: From Bench to Bedside provides a detailed understanding of the endothelium, its activation and their link to some common clinical disorders. In addition, the book covers earlier discoveries, including those from the last and 19th centuries. It is split into five sections that cover the vascular tree as an integrative structure, the endothelium in inflammation, endothelial signaling, activation and toxicity with chemotherapy, radiation induced endothelial dysfunction and vascular disease, and therapies in combating vascular diseases. Each section is discussed with a translational approach in order to make the content truly applicable. This book is a valuable source for basic researchers, clinicians in the fields of Oncology, Cardiovascular Medicine and Radiology, and members of the biomedical field who are conducting studies related to the endothelium and vascular disease. Discusses the most relevant discoveries in endothelial biology and their link to manifestations of clinical disease Presents history and diagrams in each section to highlight the original biological discovery and its link of clinical manifestations of vascular disease Includes recent findings on the relationship between endothelial signaling, chemotherapy and radiation induced endothelial dysfunction

Available in PDF, EPUB and Kindle. Book excerpt: This book focuses on the structural, biochemical, and diverse functional properties of the endothelial luminal membrane glycocalyx (ELMG), an organelle which constitutes the endothelial cell "membrane." It is intended to provide the newcomer with a broad, basic, and brief perspective of the luminal endothelial vascular membrane, and for the more established investigator, a basic overview and integrated perspective of the "universe" we explore. The endothelium is an assortment of heterogeneous regulatory cells whose cytoplasm and cell membranes are joined, forming functional units. There is a tremendous amount of literature on the endothelial cell, constituting seemingly isolated and distinct fields of encapsulated research. However, the multifunctional properties of some molecules give rise to an overlap of findings, frequently ignored between the different fields. The book is divided into three parts. The first part concentrates on the structure of the ELMG, with emphasis on morphological and biochemical composition. The importance of the chemical composition to the physiological functions of the ELMG, such as sieving properties, pharmacology, and flow sensing, is the focus of the second part of the book. Finally, some of the pathologies associated with ELMG dysfunction are explored in the last section. The aim is to provide basic and well-established knowledge in the various individual fields, identify the current concepts in each area, and discuss their respective strengths and weaknesses (including hidden problems). Finally, the overall goal is to integrate areas where overlap is clearly indicated, bringing them all together to provide the first ever basic, integrative, panoramic bird’s-eye view of the field.

Book excerpt: Endothelium and Cardiovascular Diseases: Vascular Biology and Clinical Syndromes provides an in-depth examination of the role of endothelium and endothelial dysfunction in normal vascular function, and in a broad spectrum of clinical syndromes, from atherosclerosis, to cognitive disturbances and eclampsia. The endothelium is a major participant in the pathophysiology of diseases, such as atherosclerosis, diabetes and hypertension, and these entities are responsible for the largest part of cardiovascular mortality and morbidly. Over the last decade major new discoveries and concepts involving the endothelium have come to light. This important reference collects this data in an easy to reference resource. Written by known experts, and covering all aspects of endothelial function in health and disease, this reference represents an assembly of recent knowledge that is essential to both basic investigators and clinicians.
